Abstract. Desrita, Muhtadi A, Leidonald R, Sibagaring RD, Nurfadillah 2020. Biodiversity of nekton in Batangtoru River and its tributaries in North Sumatra, Indonesia. Biodiversitas 21: 2344-2352. Batangtoru River which flows from the Tapanuli highlands (i.e., Sipirok, Tarutung, Siborong-borong, and Silangit) to the southwestern Indian Ocean is an important habitat for aquatic organisms in the western region of North Sumatra. This research was done to provide basic data on biodiversity of aquatic organisms in Indonesia, especially North Sumatra. The study was conducted in July-August 2019. Nekton was captured using backpack electrofishing units (12 volts and 9 Amperes), traps, nets, and bamboo trap. This study found 68 nekton species, consisting of fish (57 species), crab (3), shrimp (6), lobster (1), and 1 freshwater turtle (1). In general, nekton found in the Batangtoru River and its tributaries are native and economically valuable fish (Tor and eel), including the newly discovered species, which was, Hemibagrus caveatus. Invasive fish found consisted of 6 species, i.e., Cyprinus carpio, Oreochromis mossambicus, Oreochromis niloticus, Xiphophorus helleri, Hypostomus plecostomus, and Trichogaster pectoralis. The only invasive crustacean found was freshwater crayfish (Cherax quadricarinatus).

Abstract. Cahyanto T, Efendi M, Ramdan DM. 2020. Structure and composition of trees in Mount Tilu Nature Reserve, West Java, Indonesia. Biodiversitas 21: 2674-2680. An understanding of the structure and composition of stands in a conservation area is needed to support sustainable management strategy. However, this information in the Mount Tilu Nature Reserve, Bandung District, West Java area is still lacking. This research was aimed to analyze the structure and composition of tree species in the block of Malagembol forest, Mt. Tilu NR. Data collection was carried out through vegetation analysis using sampling plot method with size of 10x100 m2 at three-level altitudes of 1530 m, 1745 m, and 1950 m asl. Observation parameters included species names, number of individuals, and diameter at breast height (dbh). Data were analyzed to determine the floristic composition, species structure based on their diameter class, relative basal area, diversity indices, and analysis of the importance of the main components of trees species through Principal Component Analysis (PCA). A total of 32 tree species from 23 families was found in the observation plots which was dominated by Fagaceae, Lauraceae, and Myrtaceae families. Some pioneer plants covered the gap in vegetation due to minor disturbance and residual damage in the past. Nonetheless, the dominance of stands with small diameters indicated good regeneration status following such disturbance. Based on these findings, we recommend protecting the vegetation in Mt. Tilu NR by limiting community activities that can disturb the forest.

Abstract. Ambarwati A, Wahyuono S, Moeljopawiro S, Yuwono T. 2020. Antimicrobial activity of ethyl acetate extracts of Streptomyces sp. CRB46 and the prediction of their bioactive compounds chemical structure. Biodiversitas 21: 3380-3390. The goal of the research was to determine the antimicrobial activity of ethyl acetate extracts of Streptomyces sp. CRB46 and predict their chemical structure of the bioactive compounds based on the AntiSMASH analysis of the whole genome sequencing. Streptomyces sp. CRB46 was isolated from Cyperus rotundus L. rhizosphere in the Cemoro Sewu highland, Indonesia (1920 meters above sea level). The results showed that the supernatant ethyl acetate (EAS) extract could inhibit 11 of 12 tested microorganisms with a diameter of the inhibition zone ranging from 17-35 mm (paper disc method). The supernatant extract was found more effective in inhibiting microorganisms than the ethyl acetate extract of pellets (EAP). The EAP extract was only able to inhibit the growth of seven tested microorganisms (16-26 mm). The results of LCMS analysis showed that crude extract of EAS consisted of 20 mixtures of organic compounds. Based on FTIR analysis, 16 functional groups were found in the EAS extracts. In addition, based on AntiSMASH analysis, it was also found that Streptomyces sp. CRB46 produced eight classes of bioactive compounds, but only six compounds have been identified as their chemical structures. It can, therefore, be concluded that Streptomyces sp. CRB46 has the potential to produce bioactive compounds that can be used for microbial infection treatment.

Abstract. Bousba R, Gueraiche S, Kanouni MR, Bounar R, Djekoune A, Khammar H, Nadia Y. 2020. Genotypic diversity assessment of some durum wheat (Triticum durum) genotypes using RAPD analysis. Biodiversitas 21: 2696-2701. Knowledge of genetic variability in durum wheat (Triticum durum Desf.) is of major importance in the development of breeding programs and the preservation of local landrace resources. The objective of this study is to highlight the molecular polymorphism among six durum wheat genotypes from different origins and characterized by different sensitivity to drought tolerance (tolerant, semi-tolerant, and non-tolerant). 15 Random Amplified Polymorphic DNA (RAPD) markers were used to assess the molecular diversity of these genotypes. Our results show a total of one hundred and sixty-nine amplicons, where one hundred and twenty-four are polymorphic bands. The number of bands per primer ranged from two (OPJ-06) to fifteen bands (primers OPE-13 and OPB-01). The values of the Allelic frequency varied from 1 (OPJ-06) to 0.20 (OPA-17) and 0.19 (OPE-13, OPO-06, B-19 and OPA-20). Also, the values of the coefficient of genetic similarity range from 0.69 to 0.80, these results indicate a large variation between tested genotypes. According to the dendrogram generated by the RAPD approach, we obtained four distinct groups: the first one (G1) contains GTADUR and KORIFFLA x SHAM, the second group (G2) contains the local genotype BIDI-17. However, the genotype WAHA was in the third group (G3), the fourth and fifth groups contained the genotypes CIRTA and TELL, respectively. A complementary analysis was done to estimate genetic differentiation, using CPA Analysis that indicated four groups among the six genotypes, where, the local genotypes BIDI-17 and CIRTA were classified together. For allele’s richness, the local genotypes show in this investigation, the highest values in comparison to the introduced genotypes, which suggested the performance of the RAPD markers (high polymorphism and fast genetic analysis). The molecular markers RAPD-PCR type, despite their non-specific characteristics, has shown a strong aptitude for genetic characterization in durum wheat and a high level of polymorphism, which makes it possible in a preliminary study to make exploitable discrimination. These results may be helpful in the improvement and varietal selection, and useful in accelerating breeding programs of durum wheat.

Abstract. Fajri M, Pratiwi, Ruslim Y. 2020. The characteristics of Shorea macrophylla’s habitat in Tane’ Olen, Malinau District, North Kalimantan Province, Indonesia. Biodiversitas 21: 3454-3462.  Shorea macrophylla is a tree species in Tane' Olen forest area. This study analyzed the soil’s physical and chemical properties, topography, and microclimate of S. macrophylla’s habitat. A purposive method was used to select a sampling plot and to place the subplots. Soil was analyzed to determine the physical properties, i.e., texture, bulk density, porosity, and water content, and the chemical properties, i.e., pH, CEC, total N, organic C, C/N ratio, P, K , and Al saturation. Importance value index was determined for each tree species to know the species composition in the study site. Only the dominant species were presented. The soil at the study site had bulk density of 0.60-1.31 gram cm³-1, porosity 50.60%-77.35%, water content 34.88%-95.37%, and soil texture sandy clay. The chemical properties of the soil were as follows: pH was 3.6-4.8, N 0.05%-0.19%, organic C 1.40%-3.65%, P 0.41-1.22 mg 100 gr-1, K 58.68-232.55 mg 100 gr-1, and Cation Exchange Capacity (CEC) 5.35-10.81 meg 100gr -1. Slope ranged between 0 and 25%. The microclimate characteristics were as follows: temperature was 24-26.5°C, relative humidity 76-87%, and light intensity 145-750 Lm. Trees species with an IVI ? 10% were S. macrophylla, Madhuca spectabilis, Myristica villosa Warb, Scorodocarpus borneensis, Eugenia spp., Palaquium spp., Macaranga triloba, Syzygium inophyllum and Shorea sp. Positive associations were observed between S. macropylla and S. borneensis, Eugenia spp., Palaquium spp.. and M. triloba, and negative associations were observed between S. macropylla and M. spectabilis, M. villosa Warb, S. inophyllum, and Shorea sp. S. macrophylla grows on riversides with flat and gentle topography, acidic soil, and lower fertility but with suitable microclimate. This species can be recommended to be planted in degraded tropical forest areas but the microclimate and soil properties should be taken into account.

Abstract. Laojun S, Chaiphongpachara T. 2020. Comparative study of larvicidal activity of commercial essential oils from aromatic rosemary, vanilla, and spearmint against the mosquito Aedes aegypti. Biodiversitas 21: 2383-2389. Dengue fever is a dangerous mosquito-borne viral disease of humans. This disease is caused by dengue virus, a member of the genus Flavivirus of the family Flaviviridae. The Aedes aegypti mosquito is a principal vector for this virus. To control Ae. aegypti populations, there is extensive focus on larval eradication, because breeding sites are often close to human populations, especially man-made containers. The objective of this research was to evaluate the efficacy of commercial essential oils from rosemary (Rosmarinus officinalis), vanilla (Vanilla planifolia), and spearmint (Mentha spicata) with regard to larvicidal activity against Ae. aegypti larvae after 24 and 48-hours of exposure in the laboratory. The results showed highly efficacious larvicidal activity, with median lethal doses (LC50) of 0.23, 0.10, and 0.12 ppm after 24-hour exposure to commercial rosemary, vanilla, and spearmint oil, respectively. The results of this study will be useful for the control of a common dengue vector and may replace the use of pesticides that may have broad environmental impacts.

Biodiversitas 21: 2935-2945. The use of peatlands is a significant contributor to the world’s palm oil production. A serious problem of oil palm plantations in peatlands is the high incidence of basal stem rot (BSR) disease caused by Ganoderma, which has a higher attack rate than on mineral soils. There is no effective way to control Ganoderma in peatlands. At present, the effort for the same focuses on environment-friendly biological methods; however, this is constrained by the unavailability of appropriate biological agents for peatlands. The development of biological control agents for peatlands is hampered by limited data on biological control of Ganoderma in peatlands. This research was conducted to evaluate the in vitro antagonistic activity of fungi isolated from a peatland in acidic pH conditions. Twenty-seven Ganoderma-antagonistic fungi from peatland were evaluated for their activity and their ability to antagonism in vitro within a pH range of 2-7. The results show that most antagonistic fungi from peatland, based on biomass weight, the sporulation ability, and germination of conidium, were able to grow optimally at pH 3.0-4.0, indicating that most of the Ganoderma-antagonistic fungi from peatland can be used as biological control agents for BSR on oil palms in peatlands.

Abstract. Ku-Or Y, Leksungnoen N, Onwinom D, Doomnil P. 2020. Germination and salinity tolerance of seeds of sixteen Fabaceae species in Thailand for reclamation of salt-affected lands. Biodiversitas 21: 2188-2200. Over the years, areas affected by salinity have increased dramatically in Thailand, resulting in an urgent need for reclamation of salt-affected areas using salinity tolerant plant species. In this context, seed germination is an important process in plant reproduction and dispersion. This research aimed to study the ability of 16 fabaceous species to germinate and tolerate salt concentrations of at 6 different levels (concentration of sodium chloride solution, i.e., 0, 8, 16, 24, 32, and 40 dS m-1). The germination test was conducted daily for 30 days, and parameters such as germination percentage, germination speed, and germination synchrony were calculated. The electrical conductivity (EC50) was used to compare the salt-tolerant ability among the 16 species. Our results showed that the germination percentage, germination speed, and germination synchrony of all species decreased with an increase in salinity concentration. The mean germination time increased from 2-4 days under 0 dS m-1 to 4-10 days under 16 dS m-1, indicating that the seeds germinated at a slower rate as the salinity levels increased. Seed germination percentage was more than 80% across all the species at salinity levels between 0 to 16 dS m-1. Sesbania grandiflora, Senna siamea, and Dalbergia cochinchinensis had a high EC50 value of 33.56, 32.93, and 30.83 dS m-1 respectively, suggesting that these species were the three most salt-tolerant species in this study. As such, the establishment of these species in salt-affected areas should be studied further in order to reclaim such affected lands.

Abstract. Ginting E, Nugrahaeni N. 2020. Evaluation of physical and chemical characteristics of sixty peanut germplasm. Biodiversitas 21: 2840-2847. Nutritional values of peanut, particularly protein and fat is essential in terms of predominant use of peanut for foods. The seed size is also important for end use of the products. This study aimed to evaluate the physical and chemical characteristics of sixty ILETRI's peanut germplasm was performed. The results showed that the major genotypes had rose seed coat colors with two seeds per pod and average dried pod yield of 3.09 t/ha. Twenty-one genotypes belonged to large-seeded (? 55 g/100 seeds) and thirty-six were medium (40-55 g). The ash contents slightly varied from 2.53% to 2.98% (dw). One genotype (MLGA 0444) contained high protein (> 30% dw), while high-fat contents (> 43% dw) were noted in all genotypes. Selected genotypes had favored combinations of seed size, protein, and fat contents with high pod yield. Based on the similarity of 100-seed weights, ash, protein, and fat contents, sixty genotypes were grouped into three clusters with the main differences in seed sizes. Nineteen genotypes in cluster II with large seed size, medium protein, and high-fat content characteristics, particularly are suitable for oil extraction breeding purposes.

Autecology of Drosera burmanni in the Wolobobo Botanic Gardens, Ngada District, Flores Island, Indonesia. Biodiversitas 21: 2137-2145. Drosera burmanni Vahl is a native carnivorous plant from tropical and subtropical Asia to the West Pacific, including Flores Island. During the botanical survey, a small population of the species was found in a limited area in the Wolobobo Botanic Gardens, Flores Island. Conservation of the species is necessary, since the habitat might be developed for the botanic gardens infrastructure. This research aims to study the autecology of D. burmanni, including environmental preferences and its associated species. Thirty plots with 2 x 2 m each were made to recognize its associated species and the pattern of its population distribution and its soil requirements. In the Wolobobo Botanic Gardens, D. burmanni flourishes with an average annual rainfall of 1835 mm; a temperature of 12.50-26.2° C; 48-99% humidity; at an altitude of 1489-1491 m asl.; a soil pH of 6.66, which contains very high C-organic, a high C/N ratio, moderate N total, K2O, P2O5, low CEC, and very low K, Na, Ca, Mg, and BS. Drosera burmanni displays a density of 8.5 individuals per m2 and is associated with 20 species, which belong to 19 genera and 9 families. The species has a clustering pattern and a positive association with Erigeron sumatrensis, Spinifex littoreus, and Imperata cylindrica. To conserve D. burmanni in the Wolobobo Botanic Gardens, it is necessary to designate its natural habitat as an in situ conservation area.
