Abstract. Tapwal A, Kumar R, Borah D. 2015. Effect of mycorrhizal inoculations on the growth of Shorea robusta seedlings. Nusantara Bioscience 7: 1-5. Shorea robusta is one of important timber yielding tree species of northeast India and known to have both ectomycorrhizal (EcM) and endomycorrhizal (AM) associations. It is hypothesized that under favorable conditions different mycorrhizal fungi present in soil develop symbiotic association with fine roots of trees. In present investigations, mycorrhizal inoculum of EcM and AM fungi applied to S. robusta seedlings raised in polyethylene bags in nursery. Observations on growth characters and mycorrhizal colonization were recorded at the interval of three months. The results revealed that irrespective of type of mycorrhizal inoculation, growth of the seedlings increased significantly in comparison to control. Maximum growth was observed for the seedlings inoculated with EcM alone, followed by dual inoculations (EcM+AM), seedlings inoculated with AM fungi and minimum in control.

Transplantation of carica (Vasconcellea pubescens) at various altitudes of Mount Lawu, Central Java with treatment of shade and different types of fertilizers. Nusantara Bioscience 7: 6-14. Ex-situ conservation efforts to increase crop production can be done by means of transplantation. The aim of this study was to assess the effect of altitude, shade, and type of fertilizer and their interactions to the performance of carica (Vasconcellea pubescens) vegetative plants transplanted to Mount Lawu, Central Java, Indonesia. The research was conducted in divided plot design at three altitudes (± 1400, 1600 and 1800 m asl.) with shade treatment as main plots and fertilizer (manure, Zwavelzuur ammonium/ZA fertilizer, combination of manure with Ammonium fertilizer, and control) as subplots. Parameters of vegetative variability were observed. The results of the performance of carica vegetative plant were analyzed using ANOVA followed by Duncan's test at 5% standard test. It was observed that an increase in altitude caused slower growth of carica vegetative parts, but better vegetative growth performance in the presence of shade. For the fertilizers, the best growth parameters were obtained in the treatment of manure with ZA fertilizer. Interaction between altitude, shade, and type of fertilizer significantly influenced the performance of carica vegetative parameters. The best growth parameters are at an altitude of 1400 m above sea level with shade treatment and manure combined with ZA fertilizer.

Abstract. Pandit R. 2015. Green synthesis of silver nanoparticles from seed extract of Brassica nigra and its antibacterial activity. Nusantara Bioscience 7: 15-19. We report the green synthesis of silver nanoparticles using seed extract of Brassica nigra. UV-visible spectroscopic analysis showed the absorbance peak at 432 nm which indicated the synthesis of silver nanoparticles. Nanoparticles Tracking and Analysis (NTA) was used to determine the size of synthesized silver nanoparticles. Zeta potential analysis was carried out to study the stability of nanoparticles while FTIR analysis confirmed the presence of proteins as capping agents that provided stability to nanoparticles in colloid. Antibacterial activity of silver nanoparticles was evaluated against Propionibacterium acnes, Pseudomonas aeruginosa, and Klebsiella pneumoniae. The activity of Vancomycin was significantly increased in combination with silver nanoparticles showing synergistic activity against all bacteria while the maximum activity was noted against P. acnes.

Abstract. Xu M, Xie H, Zhong Y, Ma J, Liu S, Yang H. 2015. Treatment with MG-132 and TSA induced apoptosis in OS-RC-2 cells by increasing oxidative stress and decreasing the expression of NF-kappa B. Nusantara Bioscience 7: 20-25. Here we assessed the effect of the proteasome inhibitor MG-132 alone or in combination with the histone deacetylase inhibitor TSA on human renal cell on human renal cell carcinoma cells in vitro and we explored the underlying molecular mechanisms.OS-RC-2 cells were treated byMG-132 alone or in combination with TSA and/or NAc. The MTT assay and flow cytometry (FCM) were used to determine cell viability, ROS levels, and apoptosis. Expression of Bax and NF-kappa B p65 proteins was quantified by Western blotting. MG-132significantly increased the level of ROS, inhibited cell growth and induced apoptosis in a dose- and time-dependent manner. Both ROS and apoptosis were further increased following the combined addition of MG132 and TSA. In all cases, expression of p65 was down-regulated. MG-132 alone or in combination with TSA induced apoptosis in OS-RC-2 cells through the generation of ROS and the down-regulation of p65 expression.

Response of Syngonium podophyllum L. ‘White Butterfly’ shoot cultures to alternative media additives and gelling agents, and flow cytometric analysis of regenerants. Nusantara Bioscience 7: 26-32. Syngonium podophyllum L. (arrowhead vine) is a popular leafy indoor pot plant whose tissue culture has been established, primarily through in vitro shoot culture, but several interesting aspects have not yet been explored. In this study, cv. ‘White Butterfly’ was used to investigate the response of shoot formation to alternative gelling agents and media additives. Gellan gum (Gelrite®) at 2 g/L resulted in greater leaf production, plantlet fresh weight and higher chlorophyll content (SPAD value) than all other gelling agents tested, including agar, Bacto agar, phytagel, oatmeal agar, potato dextrose agar, barley starch and corn starch, when on a basal Hyponex® (NPK = 6.5: 6: 19; 3 g/L) medium. Several alternative liquid medium additives tested (low and full-fat milk, Coca-Cola®, coffee, Japanese green, Oolong, and Darjeeling teas) negatively impacted plant growth, stunted roots and decreased chlorophyll content (SPAD value) of leaves. Plant growth on medium with refined sucrose or table sugar responded similarly. Poor growth was observed when crude extract from a high rebaudioside-containing stevia (Stevia rebaudiana Bertoni) line - an artificial sweetener - was used. Leaf tissue from the control did not show any endopolyploidy but low levels of endopolyploidy (8C) were detected in some treatments.

Abstract. Grygorieva OO, Berezovsjka MA, Dacenko OI. 2015. Cell response of Chlamydomonas actinochloris culture to repeated microwave irradiation. Nusantara Bioscience 7: 38-42. Two cultures of Chlamydomonas actinochloris Deason et Bold in the lag-phase were exposed to the microwave irradiation. One of them (culture 1) was not treated beforehand, whereas the other (culture 2) was irradiated by microwaves 2 years earlier. The measurement of cell quantity, as well as measurement of change of intensities and spectra of cultures photoluminescence (PL) in the range of chlorophyll a emission, was regularly conducted during the cell cultures development. Cell concentration of culture 1 exposed to the microwave irradiation for the first time has quickly restored while cell concentration of culture 2 which was irradiated repeatedly has fallen significantly. The following increasing cell concentration of culture 2 is negligible. Cell concentration reaches the steady-state level that is about half of the cell concentration of control culture. Initially, the PL efficiency of cells of both cultures decreases noticeably as a result of irradiation. Then there is the monotonic increase to the values which are significantly higher than the corresponding values in the control cultures. The ratio of the intensities at the maxima of the main emission bands of chlorophyll for control samples of both cultures remained approximately at the same level. At the same time effect of irradiation on the cell, PL spectrum appears as a temporary reduction of this magnitude.

Abstract. Tarraf SA, Talaat IM, El-Sayed AEB, Balbaa LK. 2015. Influence of foliar application of algae extract and amino acids mixture on fenugreek plants in sandy and clay soils. Nusantara Bioscience 7: 33-37. Two pot experiments were conducted to study the effect of foliar application of algae extract and amino acids mixture on the growth and chemical constituents of fenugreek plants (Trigonella foenum-graecum L.). Plants were sprayed with different concentrations of algae extract (0.0, 2.5, 5.0 g/L) or equivalent of amino acids mixture (0.0, 0.625, 1.25 g/L). The results indicated that foliar application of amino acids mixture enhanced the vegetative growth of fenugreek plants, especially when plants were sprayed with 1.25 g/L amino acids mixture. Data also show that total nitrogen, essential oil percentage, and yield followed the same trend. These results hold true for plants cultivated in either clay soil or sandy soil. Data also indicated that foliar application of algae extract to fenugreek plants significantly increased plant height, number of leaves, number of branches and fresh and dry weights of plant at vegetative growth stage and flowering stage of fenugreek plant, especially in plants treated with 5 g/L algae extract in sandy and clay soils, respectively. Treatment of fenugreek plants with algae extract markedly increased nitrogen, phosphorous and potassium contents, especially at 5 g/L.

Abstract. Paralikar P. 2015. Fabrication of ketoconazole nanoparticles and their activity against Malassezia furfur. Nusantara Bioscience 7: 43-47. In the present study, ketoconazole nanoparticles were synthesized from commercially available ketoconazole powder. Sonication is the physical method used to fabricate ketoconazole nanoparticles. UV-Visible spectroscopy, FTIR spectroscopy, NTA analysis, and TEM analysis reveals the formation of polydispersed ketoconazole nanoparticles with 51nm particle size. The antifungal study demonstrates that synthesized ketoconazole nanoparticles exhibit significant activity against Malassezia furfur as compared with commercially available ketoconazole powder. Further, nanogel was prepared using ketoconazole nanoparticles which showed significant antimalassezial activity. After systematic trial, the ketoconazole nanoparticles containing gel can be used as antidandruff gel.

Abstract. Patil S, Maknikar P, Wankhade S, Ukesh C, Rai M. 2015. Antifungal effect of cumin essential oil alone and in combination with antifungal drugs. Nusantara Bioscience 7: 55-59. We report evaluation of antifungal activity of cumin seed oil and its pharmacological interactions when used in combination with some of the widely used conventional antifungal drugs using CLSI broth microdilution, agar disc diffusion and checkerboard microtitre assay against Candida. The essential oil was obtained from cumin seeds using hydrodistillation technique and was later evaluated for the presence of major chemical constituents present in it using gas chromatography and mass spectrometry (GC-MS) assay. The GC-MS assay revealed the abundance of ?-terpinene (35.42%) followed by p-cymene (30.72%). The agar disc diffusion assay demonstrated highly potent antifungal effect against Candida species. Moreover, the combination of cumin essential oil (CEO) with conventional antifungal drugs was found to reduce the individual MIC of antifungal drug suggesting the occurrence of synergistic interactions. Therefore, the therapy involving combinations of CEO and conventional antifungal drugs can be used for reducing the toxicity induced by antifungal drugs and at the same time enhancing their antifungal efficacy in controlling the infections caused due to Candida species.

Abstract. Fazlollahi Mohammadi M, Ahmadlo F, Keshtiban SM. 2015. Adaptive strip sampling in forest inventory of scattered species of Ulmus glabra in Hyrcanian Forests, northern Iran. Nusantara Bioscience 7: 48-54. Some populations of tree species may be sparse but they are highly geographically clustered. A simple random sample of plots could then yield mainly empty plots. Adaptive Cluster Sampling (ACS) is an efficient design for tallying of these populations. Presented study is designed to estimate the abundance of Wych elm (Ulmus glabra) as a scattered tree species in mountain forests of Iran. This study looks at design-based methods for a forest inventory survey, with the goal of improving estimation when the target species has a patchy spatial distribution. In particular, we examined the efficiency and feasibility of modified adaptive cluster sampling in combination with strip sampling (SACS) as a new sampling design in Hyrcanian forests, Iran for scattered species. To collect data, we recorded a number of 845 trees to represent locations of individuals in a spatially aggregated population by using GPS 60CX model. In overall, results showed that SACS was underestimated in estimating coefficient of variance (CVµ) and (Varµ) while it was overestimated in estimating mean of tree density at all quadrate size except for size 15 m square in compared to conventional strip sampling (CSS). Results evidenced that HH estimator was more efficient than HT ones in estimating mean, (CVµ) and variance (Varµ) in compared to (CSS) approach. Practically results of our study using SACS confirmed that a 15 m square sample size of quadrate with four adjacent units was optimized network size to sampling of such populations. Future studies using SACS may be required, especially, in combine to two-phase sampling strategies when cover of these species is of concern.
