Abstract

The research wasaimed to theidentifyspecies and density of sea cucumbers at southernbeach of Bali are Mertasari beach, Segara beach, and Mengening beach. The identification ofspecies was conducted in the Laboratorium of EcologyDepartment of Biology, Faculty ofNatural Science and Mathematics, University of Udayana from December 2013 toJanuary 2014.A size of 1 x 1m quadrat squaretransect sampling method was employed in this research.Identification of sea cucumbers was performed based on their morphology and shape ofspicules. GSIwas used to observed reproductionphase of sea cucumber.Two species of seacucumbers found ware Holothuria atra and Holothuria sp. Holothuria atra was found atMertasari beach, Sanur village, southern of Denpasar and Segara beach, Tuban, Kuta, Badungwhile Holothuria sp. was faound atMengening beach. Segara beach showed the highestdensityof4.333 individual/m2,while Mertasari beach showed the lowest density 0.875individual/m2. Abstract

This research was conducted to determine the morphological characteristics and thename of species tebel-tebel plant that grows on rontal trees (Borassus flabellifer L.) in CekikCampgrounds area of West Bali National Park. The method was used observation anddescription morphological character of plant parts along with species identification. Theresults showed tebel-tebel plant have the following characteristics: epiphytic plants, rootsticker, woody stems (lignosus), climbing with the twisted branches (scandens), oval leafshape (ovatus) until lancet (lanceolatus), bouquet of flowers bunches, petals 5, yellowishwhite, corona 5, base colored purple, pistil and stamens united to form ginostegium, fruittube, needle-shaped seed include spread instrument such as fine hairs, is a character trait ofthe species Hoya parasitica Wall.Keyword: morphology, identification, tebel-tebel

Abstract

Phosphate solubilizing bacteria are decomposer bacteria functioning as major role in soilenrichment. That is because it is able to perform phosphate dissolution mechanism by excreting lowparticles weight of organic acids. The bacteria utilize simple carbon compounds (rootexudatesofplantsandthe restof theplant).Phosphate solubilizing bacteria activity will be high at atemperature of 300C - 400C. This study aims to isolate and identify thephosphate solubilizingbacteria intheconventional soil and organic soil originated from Japan. Conventional and organic soil sample aresent from Collage Of Agricultural Ibaraki University in Japan. The Isolation of pathogenic bacteria isperformed by the dilution method (platting method). Results of isolation of bacteria lisolates obtained 2on ground phosphate conventional solubilizing (TKO1 andTKO2) and 4 isolates of bacteria onsoilorganicsolubilizingphosphate(TOR1, TOR2, TOR3andTOR4). Bacteria lisolates TKO2 highestphosphate solubilizing. Based on the identificati on results using the Microgen ™ Kit GNA + B - IDSystem and Determine Bergeys 's Manual of Biology (Garrity et al., 2006)TKO2 isolate is identified asYersinia sp.Keywords : Phosphate solubilizing bacteria, conventional soil, organic soil, Yersinia sp.

Abstract

his purpose of this research was to determine the benefits of the lotus plant inSumampan Village, District of Sukawati, Gianyar, Bali. The research was conductedfrom 4 to 16 February 2013. The method was used in this study is exploratory survey bydirect observation and interviews with one main informant and 15 KK from 3 banjar. Theresults showed there were 11 kinds of lotus : lotus sudamala ( Nymphoides indica ) (4.54%), yellow lotus (21.21 %), dark blue lotus ( Nymphaea stellata Wild ) (12.12 %), pinklotus (16.66 % ), violet lotus (9.09 %), purple lotus (9.09 %), white lotus (Nymphaeanouchali Burm f.) (18.18 %), light blue lotus (Nymphaea stellata Wild ) (3.03 %), lotustutur (1.51 %), lotus dedari (1.51 %) and lotus brumbun (3.03 %). The lotus plant is usedas a upakara / banten 77.41 % , 16.12 % as ornamental plants, while 6.45 % as amedicine for breast cancer drug, arthritis, headaches, stress, fear, and cleanser the liverand pancreas. As a medicine, lotus plant was used in it’s from as tempel, solutions, pupukand boreh.Key word: exploratory survey, benefits lotus, tempel, pupuk, boreh

Abstract

DNA extraction is the first step to study plant systematic and biodiversity analysis usingmolecular markers. This study aimed to conduct DNA extraction from herbarium materialsusing different extraction methods. A total of 0.05 grams of herbarium powders ofCalantheemarginata (Blume) Lindl. and Goodyera procera(Ker-Gawl) Hook. (terrestrialorchid) were used for samples by three different methods. The first method was from Doyleand Doyle with modification of incubation time for 1,5 hours at 65oC and increasing EDTAconcentration to 50 mM. Second method was Dellaporta et al. with modification of incubationtime for 1,5 hours (at 65oC) and increasing EDTA concentration to 100 mM. Third methodwas Rogers and Bendich with modification of incubation time for 1,5 hours (65oC) andadding ethanol twice. The results of electrophoresis revealed that method of Doyle and Doyleobtained DNA from C. emarginata herbarium, while method from Rogers and Bendich,unfortunately it was inconsistent. The method from Dellaporta et al.obtained DNA from G.procera herbarium, while method from Doyle and Doyle revealed inconsistent DNA forG.procera. PCR-RAPD revealed the quality of DNA isolated using Doyle and Doyle methodwas not optimal, showed by unclear patterns of DNA bands. PCR-RAPD using DNA isolatedwith method from Rogers and Bendich revealed clearer DNA bands but only for small sizefragment.Keywords : orchid, DNA extraction, herbarium, PCR

Abstract

The aim of this study is to determine the total amount of Staphylococcus aureus in atraditional sausage (urutan) sold in Denpasar traditional market in Bali. Sausage samples aretaken from four areas in Denpasar (North Denpasar, East Denpasar, West Denpasar, and SouthDenpasar). From each area, three traditional markets are selected and from each market 2merchants are chosen. The calculation of the number of Staphylococcus aureus is done byplatting method and by pour plate method. The results show that the highest number ofStaphylococcus aureus belongs to the North Denpasar region in the mount of 241,067 CFU/gwhile the lowest belongs to the region of West Denpasar with 71,233 CFU/g. These two resultsare significantly different with one another (P<0,05). The maximum limit of microbialcontamination in processed meats (sausages) for Staphylococcus aureus based on IndonesianNational Standard (SNI) is equal to 102 CFU / g. Based on the testing that has been done, allurutan contamination by the Staphylococcus aureus in Denpasar traditional market, hasexceeded the threshold amount of SNI.Keywords : Traditional Sausages (urutan), Staphylococcus aureus
