Abstract

Uncontrolled use of antibiotic caused the emergence of bacteria strain resistant toward of antibiotic. The sensitivity assay of E. coli toward p-lactam antibiotic has been done. The sensitivity assay was started with isolated bacteria from patients urine at Dr. Sardjito hospital followed by count total of colony/number of germ which growth and identified the bacteria. The Kirby Bauer method was performed. Antibiotic impregnated disk (ampicillin 30 ng, sulbactam/ampicillin 20 ng, cefotaxim 30 ng, ceftazidime 30 jig, ceftriaxon 30 ng, cefpirom 30 ng, cefepim 30 ng and imipenem 10 ng) were placed on agar plate previously streaked with suspension of E. coli (1.10 CFU/ml). The plates were incubated for 18-24 hours at 37° C. The diameters of the zone inhibition were measured, and compared to standar interpretive zone sizes. The result of the study showed that all of E. coli resistant toward antibiotic ampicillin; 65% resistant toward antibiotic sulbactam/ampicillin; 50% still sensitive toward antibiotic third cefalosporin generation that is cefotaxim, ceftazidime, ceftriaxon; more than 50% sensitive toward fourth cefalosporin generation like cefepim, cefpirom and all of pathogen bacteria E. coli sensitive toward antibiotic imipenem.

Abstract

ABSTRACTFurosemide is a diuretic drug, which is insoluble in water. Due to this condition, it is needed away to increase the dissolution rate with forming of inclusion complex in copresipitation systemwhich produce as solid dispersion product using β-cyclodextrin carrier. The copresipitation systemwas made up of 1 : 0,5; 1 : 1; 1 : 1,5 and 1 : 2 variation concentration of furosemide- β-cyclodextrin. The characteristic forming of inclusion complex in solid dispersion system wasevaluated by infrared analysis and then followed by HyperChem molecular model analysis. Thedissolution test was done in order to see the increasing of dissolution rate and this test is usedbuffer phosphate pH 5,8 as the medium with rotation speed 100 rpm at 37 ± 0,50C for 60 minutes.The amount of dissoluted furosemide was then analyzed by spectrophotometric test. Abstract

ABSTRACTAim of this research was to determine chemical structure of flavonoid from water fraction ofSappan wood. The flavonoid was isolated by Soxhlet xtraction using methanol and infundation.First, the detection of flavonoid was done using Thin Layer Chromatography (TLC) with statioramphase silica gel GF 254 and mobile phase ethyl acetate : acetyc acid 15 % (9:1 v/v) smoked byNH3, and detected under UV 366 nm, showed 5 spots with hRf 7, 50, 61, 68, and 80. PreparatifTLC was done to water fraction, disolveed in methanol and detected with UV-Visspectrophotometry using diagnostic reagents NaOH, NaOAc, NaOAc+H3BO3, AlCl3, ACl3+HCl andcompared with references. In fraction 1, 2, and 5 showed isoflavon with 7-OH, substitution oxygenin position six. In fraction 3 showed isoflavon with 7-OH, substitution oxygen in position six, 6,7diOH in A ring, and o-diOH in 6,7 or 7,8. In fraction 4 showed isoflavon with 7-OH, substitutionoxygen in 6, o-diOH in 6,7 or 7,8 and 5-OH.Key Words: Caesalpinia Sappan. L, Flavonoid, TLC, UV-Vis Spectrophotometry

Abstract

ABSTRACTToxoplasmosis is caused by infection with the protozoan Toxoplasma gondii. CongenitalToxoplasmosis is an infection resulting from the transplasental passage of the parasites frominfected mother to the fetus. An acute infection in pregnant women associated with a range ofoutcomes from subclinical infection to intra uterine death.Early diagnosis is important to makedecision, therapy or abortion. At present, prosedur diagnostic of congenital toxoplasmosis usesconventional methode especially serological test,but the conventional methode is time consuming,and has high risk. PCR amniotic fluid is one methode to make dignostic of conegenitaltoxoplasmosis. This is very sensitif and rapid (24 hours) and safer than serological tes of thefetalblood.PCR can be done from 15 week after gestation.Key word: congenital toxoplasmosis- PCR- amniotic fluid.

Abstract

ABSTRACTAcetaminophen is a kind of analgetic-antipyretic drug which the usage in excessive dose willcause the liver damage. Pegagan herb (Centella asiatica, L) is one of plant which is used to protectthe liver from various damage of drug. A research to study the hepatoprotective effect of pegaganherb infusion on acetaminophen induced wistar rat has been conducted. This research was aimedto find how excellent pegagan herb protects the liver from the damages caused by acetaminophenand find the data of hepatoprotective of pegagan herb’s dose effect span in infussion form throughthe SGPT enzym and hyspatology of liver cell analysis. This research used the completed randomof unidirectoral pattern method, using wistar strain white rat as the tested animals, weight 200 g 10%, age 5-8 weeks. The way of attemp: 36 rat were divided into 6 groups were each groups had 6rat. For the treatment, Group I was controlled by aquadest. Group II was given acetaminophensuspention dose 2,5 g/kg BB. Group III-IV was given pegagan herb infussion 0,027; 0,054; 0,0108;0,216 g/kg BB once in a day for a week and 8 hours after that, on the seventh day was givenacetaminophen suspention dose 2,5 g/kg BB. Blood taking was conducted by orbitalis sinus beforeth treatment, 8 hours after the seventh day treatment and 24 hours after acetaminophen was given.SGPT data was analysed by ANOVA statistical test, where if found the significant difference, itwould continue by Tuckey Test with 95% as significant standar. Then continued by hyspatology teston liver cell of rat. The result of this research showed that pegagan herb infussion dose 0,027;0,054; 0,108; 0,216 g/kg BB had a hepatoprotective effect on acetaminophen inducted white ratsuccessively 34,19%; 62,52%; 78,62%; 85,69%. And the result of qualitative analysis by histologyanalysis showed that pegagan herb had hepatoprotective effect.Keyword: Hepatoprotective, Pegagan Herb,Acetaminophen

Abstract

ABSTRACTThe latest discoveries in the field of microbiology have proved that bacteria communicate eachother. The process of cell to cell communication is called quorum sensing. Quorum sensing wasfirst discovered in two luminous bacteria called Vibrio harveyi and Vibrio fischeri. These bacteriaemit light in response to increase in cell population density. Density-dependent light production isaccomplished trough the release and detection of hormone-like molecules called autoinducers thataccumulate in the environment as the bacterial density increases. Quorum sensing is believed toregulate competence in development, sporulation, virulence factor induction, sporulation andnutrient flux along with other events in pathogenic bacterial infections. Recently, many scientisthave been doing research in the field of quorum sensing. If the signaling systems among thebacteria were able to be blocked, the dangerous effects from bacteria then possibly be prevented.In other word, if the quorum sensing mechanism were stopped, the disease can be prevented oreven cured instead of antibiotics use.Keyword : bacterial infection – quorum sensing - inhibiton

Abstract

ABSTRACTIgA-deficient in individuals have developed mucosal infections, atopy, autoimmune diseasesand rheumatic. Under the circumstance, the development of an immunomodulator that stimulatesimmune system, especially IgA, is truly desired. The research is objected on finding out the effect offermented horse milk treatment on IgA antibody in Hepatitis A immunized mice. IgA concentrationin serum and lymphocytes was evaluated using indirect ELISA method. Fermented horse milkgroup was compared with water group to calculate the percentage of IgA concentration increase.Univariate analysis was used to analyze Optical Density (OD) product from ELISA. The resultshowed that fermented horse milk increased 46.20 % IgA concentration in serum and 95.47 % inspleen lymphocytes. Univariate analysis showed that fermented horse milk increased significantlyIgA spleen lymphocytes but not IgA serum.Keywords : immunomodulator, IgA, hepatitis A vaccine, ELISA
