Abstract

Human erythropoietin (hEPO) is a glycoprotein that regulates the formation of erythrocytes and mainly used in anemia patients. Previously, we have reported the expression of modified human EPO with 2 additional N-linked in mammalian cell CHO-K1. The aim of this current research was to study the optimum condition for modified recombinant hEPO (rhEPO) production in CHO-K1. To do this, several parameters of culture conditions were applied including antibiotic concentrations, seeding densities, time of incubations, fetal bovine serum (FBS) concentrations and cell culture media. The result showed that the presence of antibiotic G418 improved the expression level with the highest was at 1% of concentration. Meanwhile, seeding density of 2–3x105 cells/6 cm dish and seven day of incubation time were the best condition for rhEPO protein expression. From five different combination media used, F12 medium with 10% FBS gave the highest expression of rhEPO protein. From this study was also found that at passage 16 the expression level was still increasing proving that the clone expressing the protein of our interest is promisingly stable.Keywords : EPO, erythropoietin, protein expression, CHO-K1, optimation

Abstract

There are so many potential effects that green tea, such as antibacterial activity against Propionibacterium acnes, that caused acne. In the previous study, green tea had been made in gel. In this study, formula of green tea water extract gel is developed with addition of antioxidant so that stability of the gel would increase. Gel of green tea water extract were made using Carbopol 940 with glycerin as the solvent. The chosen antioxidant was sodium sulfite. It is also combined with chelating agent, EDTA (Edetic Acid). The activity of green tea water extract gel was determined and tetracycline HCl gel was used as the reference. The addition of sodium sulfite 1% w/w into gel with Carbopol 940 1% w/w as the base prevent the change of color in more than eight weeks. Minimum Inhibitory Concentration (MIC) of green tea water extract without either sodium sulfite or EDTA was 1.20 ± 0.11 cm against Propionibacterium acne. MIC of green tea water extract with addition of sodium sulfite 1% w/w was 1.19 ± 0.28 cm. MIC of green tea water extract with addition of sodium sulfite 1% w/w and EDTA 0.1% w/w was 1.25 ± 0.22 cm. The addition of sodium sulfite 1% w/w caused viscosity of gel decrease 42.1%. The addition of sodium sulfite 1% w/w increased the color stability of green tea water extract. The addition of sodium sulfite 1% w/w didn’t decrease its antibacterial activity against Propionibacterium acnes.Keywords: green tea, anti acne, Propionibacterium acnes, gel, antioxidant

Abstract

The pumpkin belongs to the family of Cucurbitaceae, is a well-known edible plant that has been frequently used as functional food or herbal medicine. Pumpkins contain rich unsaturated fatty acids, phytoestrogens and vitamins E in their seeds that have potential pharmaceutical, nutraceutical, and cosmeceutical properties. Information regarding their nutritional components and therapeutic properties of pumpkin seeds has expanded dynamically in the recent years and this review focus on the three main components of pumpkin seeds that described before. Several types of unsaturated fatty acids are the dominant component in pumpkin seeds which can play a role in the disease prevention and promote health. Pumpkin seeds also contain the important phytoestrogen compounds, i.e., secoisolariciresinol and lariciresinol that have estrogenic-like effect such as preventing hyperlipidemia and osteoporosis for menopausal women. Phytoestrogens in pumpkin seeds also could be related to a reduced hormone-dependent tumor. Pumpkin seeds are rich in vitamin E contents as an emerging free radical scavenger, anti-aging and antioxidant such as a-tocopherol and g-tocopherol. Findings of these studies prove that patents field for the innovation product of pumpkin seeds holds promise for the future along with their immense nutraceutical properties.Keywords : pumpkin seeds, estrogenic, anticancer, antioxidant, nutraceutical

Abstract

Indonesian medicinal herbs have so many biological activity, including anticancer activity. This assay aims to evaluate three Indonesian medicinal plants (Trigonella foenum-graecum L. (TFG), Aglaia elliptica Blume. (AE) and Foeniculum vulgare Mill. (FV)) which have potency in anticancer activity. The experiment was done by MTT assay to observe the cytotoxicity activity on breast cancer cell lines MCF7.  Apoptotic potency was evaluated based on % population of MCF7 cell in subG0/G1 phase.  The results showed that IC50 from ethanolic extract of TFG was 241.24 ppm, ethanolic extract of AE was 19.442 ppm and  ethanolic extract of FV was 69.41 ppm.  Apoptotic potency for TFG was 6.46%,  AE was 11.94% and for FV was 8.73%. Thus, it is concluded that ethanolic extract of AE gives the lowest IC50 and the higher potency of apoptotic by significantly (p<0.05) increase % population of cells in subG0/G1 phase on 100ppm concentration.Keywords :  Trigonella foenum-graecum L., Aglaia elliptica Blume. and Foeniculum vulgare Mill., MCF-7, Apoptotic, Cytotoxic

Abstract

The development of cancer treatment were initiated by the existence of human’s effort to treat by applying certain materials which is mostly part(s) or extracts of plants, which are now adapted as traditional herbal medicine. The discovery of new drugs was based on intuition and empirical evidence. Thus, high luck factor was involved in a successful treatment with unguaranteed reproducibility. One example of drug being developed through conventional drug development is Taxol. Taxol is an extremely complex natural product and requires a bunch of hard work with high level of serendipity to be discovered as antitumor agent. Recently, rapid development in human biology and technology allow a change in drug discovery strategy by minimizing the luck factor. Targeted therapy has been a very promising strategy of drug development research, especially in cancer treatment. Although cancer has been known as a disease with very complex cellular and histo-pathophysiology, the abundance of studies on proteins, such as receptors and hormones, as the hallmarks of cancer allows us to explore carcinogenesis suppression further based on molecular targeted therapy. Kinases, one type of protein involved in signal transduction regulating cell growth and differentiation, could be the proteins that are proposed to be inhibited in suppressing tumor growth. An interesting example of the drug being discovered based on molecular modeling is the discovery of lapatinib as anti- cancer with specific target on HER-2 and EGFR to overcome the resistance of cancer to Herceptin caused by elevated level of EGFR expression.Keywords : targeted therapy, cancer, translational drug development

Abstract

Many natural products have been widely explored for their pharmacological activities, including anticancer activity. Hedyotis corymbosa L. is known for its anticancer properties toward several cancer cell lines. The study aims to investigate whether ethanolic extract of Hedyotis corymbosa L. (EEH) performs anticancer properties by inhibiting migration and metastasis on breast cancer cells. Cytotoxic evaluation by using MTT assay was carried out to determine EEH effect on 4T1 breast cancer cells, meanwhile to investigate the treatment of EEH in migration and metastasis inhibitory effect, scratch wound healing assay and gelatin zymography were conducted in this study. The data showed that EEH possessed cytotoxic activity with IC50 value of 400 μg/mL.  Interestingly, migration inhibitory effect was shown up to 42 hours and the activity of MMP-9 was also decreased after the treatment with EEH. According to these findings, we suggest that Hedyotis corymbosa L. promotes another anticancer properties by inhibiting migration and metastasis towards breast cancer cells.Keywords : Hedyotis corymbosa L., cytotoxicity, migration, metastatic, 4T1 breast cancer cells

Abstract

Osteoporosis in menopause woman is caused by estrogen deficiency which plays an important role in bone formation. Corn silk (Stigma Maydis) contains stigmasterol, a phytosterol compound predicted to act as phytoestrogen. The aim of this research is to observe the activity of Corn Silk as the source of phytoestrogen by in vivo study in ovariectomized rats. Bone density analysis was examined by using x-ray. Meanwhile, histological profile of bone matrix was determined by HE-staining microscopic observation. Affinity of stigmasterol to ER (α,β) were evaluated by molecular docking. The results showed that treatment of EECS after ovariectomy has not been able to increase bone density compared to the control group OVX. Moreover, histological observation of bone matrix showed that EECS performed improvement effect compared to was observed in the administration of estradiol. Docking between stigmasterol and ER (α,β) gave the docking score which are almost the same as that seen in docking with estradiol. In summary, EECS produced positive effects on bone density in estrogens-deficient OVX rats by reducing bone resorption. Therefore, EECS may also prove to be helpful in preventing osteoporosis in postmenopausal women whose estrogen is insufficient.Keywords: osteoporosis, phytoestrogen, corn silk, stigmasterol

Abstract

Ursolic acid has been widely known to possess biological activity against numerous tumor cell lines. Previous studies revealed its cytotoxicity on several cancer cells in vitro by either inducing apoptosis or cell cycle modulation. This study was conducted to investigate ursolic acid’s cytotoxicity solely and in combination with a chemotherapeutic agent, doxorubicin, on MCF-7 breast cancer cells, followed by observation on its mechanism. Cytotoxicity of single and combinational treatment of ursolic acid and doxorubicin on MCF-7 breast cancer cells were conducted by using MTT assay. Single treatment was then evaluated by determining IC50 value, while combinational treatment was evaluated by analyzing cell viability and evaluating combination index (CI). To explore the mechanism underlying cytotoxic effect on respected cells, further analysis on cell cycle profile of single and combinational treatment was conducted by flow cytometry. Twenty four hours treatment of ursolic acid inhibited MCF-7 cells’ growth with IC50 value of 37 µM, while combinational treatment showed that several concentration combinations of ursolic acid and doxorubicin exhibited synergism of cytotoxic activity on MCF-7 cells, giving optimum CI value of 0.54. Flow cytometric analysis showed that combinational treatment induced G2/M arrest in MCF-7 cells. These results show that ursolic acid is promising to be developed as either single chemopreventive agent, or as doxorubicin’s co-chemotherapeutic agent in breast cancer treatment. Observation on the selectivity as part of safety aspect together with in silico, in vitro, and in vivo study on its molecular mechanism should be conducted.Keywords: ursolic acid, doxorubicin,co-chemotherapeutic agent, breast cancer, cell cycle

Abstract

Degenerative disease caused by decreasing organ function in old people has become the biggest death cause in the world. This aging process marked by senescence activity. An anti-aging agent from the medicinal plant has high potency to be developed, such as pumpkin seed (Cucurbita moschata) which contain tocopherol as antioxidant. The aim of this study is to investigate the anti-aging effect of pumpkin seed extract (PSE) on NIH 3T3 fibroblast normal cell induced by doxorubicin. This anti-aging effect was observed using MTT assay continued by SA- βgal (Senescence-Associated Beta-Galactosidase) activity detection test. The stability of  molecular interaction between tocopherol and doxorubicin to CYP 3A4 as the oxidase enzyme was conducted using molecular docking. Based on in vitro test, PSE is not cytotoxic to NIH 3T3. PSE at the dose of 100,200,400, and 800 μg/mL decreased % cell senescence by 2,77; 4,5; 6; and 18 times respectively. Meanwhile, in silico test indicated that tocopherol (-107,409) has a higher interaction to CYP 3A4 compared to doxorubicin (-70,52). Both compounds have a similar binding site in Leu 364; Phe 435; Pro 434; Cys 442; Ile 369; Thr 309; dan Ala 305. The overall result of this study showed that PSE has anti-aging effect by decreasing SA- βgal activity. This anti-aging activity possibly due to the interaction between tocopherol to CYP 3A4 based on molecular docking.Keywords: anti-aging, Cucurbita moschata, tocopherol, SA-βgal, molecular docking

Abstract

Previous study reported that Nigella sativa has in vitro and in vivo cancer activity.This study was conducted to observe the effect of chloroform extract of Nigella sativa seed (NCE) on 7,12-Dimethylbenz[a]anthracene (DMBA)-induced female rats’ splenocyte. The experiment consisted of five groups, corn oil solvent control group, DMBA group, DMBA+250 mg/kgBW NCE, DMBA+500 mg/kgBW NCE and DMBA+750 mg/kgBW NCE. Extract was dissolved in corn oil and oral administered daily for 2 weeks before and during the DMBA induction. Observation of cell proliferation was performed using haematoxylin and eosin (H&E) and AgNOR stainings. H&E staining showed decreased necrocis activity extract groups compared to DMBA group. From AgNOR staining results, mean AgNOR (mAgNOR) of extract groups was less in number compared to DMBA group. The mAgNOR in corn oil solvent control group, DMBA group, DMBA+250 mg/kgBW NCE, DMBA+500 mg/kgBW NCE and DMBA+750 mg/kgBW NCE were 1.22, 1.91, 1.29, 1.36 and 1.33, respectively. Our current results showed that NCE reduces the proliferation of DMBA-induced rat spleenocytes  Thus, NCE has potency to be developed as a chemopreventive agent.Keywords: Nigella sativa, spleen, DMBA, anti-proliferative
