Abstract

The aim of this study is to determine the effect of coconut (Cocos nucifera) leaf enrichment to soil of mangrove forest of Bali Suwung coastal on the cellulase activity. The measurement of cellulase activity was conducted by CMC method (Carboxymethyl Cellulose Assay) on soil sample with and without enrichment of coconut leaf substrate in incubation time of 1, 2, 3, and 4 weeks. The results showed that the enrichment increased cellulase activity. In addition, cellulase activited the soil samples, increase with incubation times.

Abstract

The aims of this research are to determine the concentration of reducing sugar and starch in Glacilaria sp. and to investigate the influence of the concentration of ammonium sulfate as nitrogen source on the production of bioethanol using Glacilaria sp. as the carbon source. The concentrations of ammonium sulfate employed were 0,25%; 0,50%; 0,75%; and 1% (w/v). The methods employed in this research were the determination of water content, glucose and starch concentration using Nelson-Somogyi method, determinating the optimum duration of fermentation, pH measurement, and the determination of  ethanol concentration. The results showed that the concentrations of reducing sugar and starch in Glacilaria sp. are 17,14% and 15,42%. The concentration of (NH4)2SO4, which produced the optimum concentration of ethanol (1,04% (v/v) on seventh day), was 1% (w/v). The fermentation media containing (NH4)2SO4 of 1%; 0,75%; 0,5%; and 0,25% (w/v) produced optimum concentrations of ethanol, which were 0,96% (v/v) on fifth day; 0,78% (v/v) on seventh day; 0,74% (v/v) on seventh days; and 0,73% (v/v) on seventh day, respectively. The optimum concentration of (NH4)2SO4 was 1% (w/v), which produced maximum concentration of  bioethanol from Glacilaria sp., of 0,96% (v/v).

Abstract

Protium is the main genusl in the family of Burseraceae which has been used traditionally as medicine, cosmetic as well as insecticide. This research aimed to isolate and identify antifeedant active compounds from leaf of Protium javanicum Burm, F which is locally known as tenggulun. Epilachna sparsa (E.sparsa) larvae were used as bioindicator for antifeedant activity assay. The assay was performed both on crude extract, fractions, and isolate. Dried leaf powder of tenggulun (1000 g) was extracted with methanol to yield 20.89 g dark green extract. This extract showed 71.61% antifeedant activity at 0.1% (b/v). This active extract was then fractionated into n-hexane, chloroform and water fractions. The chloroform fraction was the most active which give 89.62% activity at 0.1% (b/v) compared to n-hexane (36.83%), and water fractions (1.96 %). Purification of chloroform fraction was done using silica gel column chromatography with n-hexane : chloroform (3:4) as eluent and three groups of fractions (isolates) were obtained. The most active isolate which showed 70.53% antifeedant activity at 10 ppm was found relatively pure, therefore it was then identified.  Based on phytochemical reaction, analysis of infrared spectra and ultraviolet-visible spectra, the antifeedant active isolate was identified as triterpenoid class of compounds which have functional group such as OH, CH3, CH2, C=O, C=C and ?max at 245 and  416  nm.
