Abstract

The aim of this study is to determine the adsorption capacity of NaoH activated bentonites in reducing metal ions Pb2+ and Cu2+ from their solutions. The characterization of the activated bentonites was carried out by determining the specific surface area using methylene blue adsorption method, the numbers of the active site by acid-base titration, and determining the basal spacing using X-ray diffraction (XRD). Adsorption parameters such as contact time, pH, and the isotherm adsorption curves were determined by atomic absorption spectrophotometer (AAS). The specific surface area and the number of active sites of the NaOH-activated Bentonite were 22.1017 m2/g and 32.8716 x 1020 atoms/g respectively. This vakue was higher than non-activated ones which were 12.6602 m2/g and 31.5847 x 1020 atoms/g respectively. The optimum condition of Pb2+ adsorption on activated bentonite was obtained at pH 3 with contact time under 5 minutes following the Langmuir isoterm pattern while the optimum condition of Cu2+ adsorption was obtained at pH 4 with contact time under 5 minutes following the Langmuir isoterm adsorption suggesting that the adsorption took place by chemical adsorption. The adsorption capacities of Pb2+ and Cu2+ were 185.50 mg/g and 30.00 mg/g respectively.

Abstract

Isolation, identification and antibacterial activity of flavonoid compounds from jackfruit leaf (Artocarpus heterophyllus Lmk) extract have been done in this research. Extraction of 500 g dry powder of jackfruit leaf using maceration method produced 31.20 g concentrated brownish green ethanol extract. Partition of 30.00 g the ethanol extract gained three concentrated extracts in n-hexane (4.46 g), chloroform (0.84 g) and n-buthanol (6.65 g) fractions. The phytochemical test showed that n-buthanol extract contained flavonoid compound which was active as antibacterial agent with 15.75 mm inhibition capacity. Column chromatography analysis on n-buthanol extract with mobile phase n-hexana: ethyl acetate: n-butanol (8:2:1) gave 8 fractions (FA, FB, FC, FD, FE, FF, FG, FH).  Phytochemical testing showed that fraction FA and fraction FH belong to flavonoide compounds. Analysis of FA using UV-Visible gained 2 peaks at ? 323.40 nm (band I) and ? 285.60 nm (band II) which indicated the flavonoide groups of flavanone or dihydroflavonol. Analysis of FH using UV-Visible gained 2 peaks at ? 323.40 nm (band I) and ? 285.60 nm (band II) suggested the flavonoide groups of flavone or flavonol (3-OH substitution).` By using shiffting reagent the isolate FA was suggested to contain dihydroflavonol group and FH was suggested to contain flavone with hydroxyl groups at C-2’, C-4’.C-5’, C-6’, C-6, C-7, and C-8. Infrared analysis showed that the isolate had –OH, C=O, C-O, C=C aromatic, CH aromatic, and CH aliphatic groups. Antibacterial testing of flavonoid compounds of fraction A and fraction H on Staphylococcus aureus with a concentration of 10.000 ppm inhibited 10.50 mm for fraction A and 7.25 mm for fraction H.

Abstract

The aim of this research was to find specific activity of bromelain enzyme extracted from crude extract of pineapple fruit (Queen variety) by salting out method using three different salts, which were (NH4)2SO4, NaCl, and Kusamba traditional salt. Crude extract of bromelain enzyme was obtained by precipitation method using salt with different levels of saturation. This study included determination of total protein concentration by using Biuret method and protease activity by using Anson method to get the specific activities of the crude extract. The results showed that the highest specific activities of bromelain enzymes precipitated with the three salts (NH4)2SO4, NaCl and Kusamba traditional salt with the ratio of 1: 0.94: 0.56 were 0.2887 U/mg (at 20% saturation) ; 0.2705 U/mg (at 40% saturation) and 0.1623 U/mg (at 80% saturation). Fractination of enzyme with the NaCl and Kusamba traditional salt gave smaller specific activity value than (NH4)2SO4. This results may caused of NaCl had smaller ionic strength, the presence of other ions in Kusamba traditional salt and the different solubility of bromelain enzyme in different salts. However, due to the ratio of NaCl approached (NH4)2SO4 and the availability of Kusamba traditional salt they could be used as alternative salts to replace (NH4)2SO4 in fractination bromelain enzyme from crude extract of pineapple fruit
