Abstract

Six compounds in one fraction of seaweed Gracilaria coronopifolia (Red algae) that have radicalscavenging activity have been identified. Extraction of those compounds was carried out by maceration method using 96% ethanol. Separation and purification of the fraction were performed by liquid-liquid partition and column chromatographic methods. The obtained fractions were treated with 2,2-diphenyl-1- picrylhydrazyl and found that their radical-scavenging activities of the first (A) and the second (B) fractions from the column with a 60 min retention time were 83.12% and 90.72%, respectively. The compounds contained in the A and B fractions identified by GC-MS were 1-nonadecene; hexadecanoic acid or palmitic acid; octadec-9-enoic acid; cholesta-4,6-dien-3β-ol; cholest-5-en-3β-ol or cholesterol; and cholest-4-en-3- one. These compounds could be expected to have radical scavenging activities.

Abstract

Telah diidentifikasi gabungan senyawa yang paling aktif sebagai antibakteri dalam tumbuhan kentut-kentut (Paedaria foetida Auct.) Ekstraksi metabolit dilakukan dengan cara maserasi menggunakan pelarut etanol 85%. Pemisahan dan pemurnian metabolit mula-mula dilakukan dengan metode partisi cair-cair menggunakan pelarut n-heksan, kloroform, dan etilasetat, dilanjutkan dengan metode kromatografi kolom (silikagel) dengan eluen campuran kloroform : n-heksana  (4:1). Fraksi yang paling aktif dianalisis menggunakan panduan kromatografi gas-spektroskopi massa. Bakteri yamg digunakan sebagai bioindikator adalah Micrococcus luteus dan Eschericia coli. Fraksi yang menggunakan sifat paling aktif antibakteri andalan fraksi etilasetat. Hasil identifikasi fraksi yang paling aktif tersebut adalah sebagai berikut:  yang aktif menghambat pertumbuhan bakteri Micrococcus luteus dan Eschericia coliadalah gabungan enam senyawa, yaitu:  asam benzen asetat, 4-vinil-2-metoksi-fenol, 2-hidroksi asam benzoat, asam miristat, stigmasterol, dan kolesterol. Sedangkan yang hanya aktif menghambat pertumbuhan bakteri Eschericia coli adalh gabungan tiga senyawa, yaitu:  fenpropatrin, stigmasterol, dan kolesterol.

Abstract

ABSTRACTAnticancer activity test of the ethanol extract of sponge Hyrtios erecta from Pari Island beach (Jakarta) has been conducted. Extraction of the sponge was carried out by 70% ethanol at room temperature. Toxicity screening test was carried out based on Bhrine Shrimp Lethality Test (BSLT). Invitro anticancer activity test of the extract was carried out using HeLa cell line. Based on the results, it was found that ethanol extract of Hyrtios erecta sponges has anticancer activity with LC50 of 26.35 ppm.ABSTRAKTelah dilakukan uji antikanker ekstrak etanol spons Hyrtios erecta yang berasal dari perairan Pulau Pari Kepulauan Seribu, Jakarta. Ekstraksi dilakukan dengan cara maserasi menggunakan etanol 70% pada temperatur kamar. Skrining toksisitas dilakukan dengan metode Bhrine Shrimp Lethality Test (BSLT). Uji antikanker secara invitro ekstrak tersebut menggunakan sel HeLa. Berdasarkan hasil penelitian ini diperoleh bahwa ekstrak etanol spons Hyrtios erecta bersifat antikanker dengan harga LC50 sebesar 26,35 ppm.

Abstract

Coleus scutellariodes L, Benth, locally known as Iler, has been traditionally used as medicines such as for treatment of fever, ears infection, diabetes, and dyspepsia. The polar constituents in Iler extracts are believed can inhibit the growth of leukemia cell. This research aims to isolate and identify any toxic compounds from toxic fraction of Iler which could be potential as anticancer agents. Extraction of metabolites from Iler leaves was carried out at room temperature using maceration methods with 85 % ethanol eluent. The crude extract was then fractionated into n-hexane, chloroform, and ethyl acetate respectively. Both extract and fractions were tested towards Artemia salina. The most active fraction was purified on silica gel column chromatography using n-hexane:chloroform (2:1) as eluent. Four compounds were identified in the most toxic fractions (LC50 90.72) using Gas Chromatography-Mass Spectrometry as palmitic acid ( 24.41), stearic acid (19.53%), 9-Octadecenamide (22.95%), and hexadioic acid, diocttyl ester (33.11%). Abstract

Telah dilakukan uji antikanker isolat toksik 3 (tiga) jenis spons yang berasal dari perairan Indonesia. Ketiga spons tersebut adalah Callyspongia aerizusa, Haliclona fascigera, dan Lanthella basta. Isolasi metabolit dalam spons dilakukan dengan cara maserasi, kemudian dilanjutkan dengan tahap pemisahan dan pemurnian menggunakan cara partisi serta kromatografi kolom. Skrining toksisitas dilakukan dengan metode Bhrine Shrimp Lethality Test (BSLT). Uji antikanker secara in vitro isolat yang paling toksik menggunakan sel HeLa. Berdasarkan hasil penelitian ini diperoleh bahwa spons Callyspongia aerizusa bersifat sebagai antikanker dengan LC50 sebesar 5,50 ppm. Spons Haliclona fascigera tidak bersifat antikanker terhadap sl HeLa, karena harga LC50 sebesar 44,67 ppm. Sedangkan spons Lanthella basta bersifat antikanker dengan harga LC50 sebesar 18,62 ppm.Kata Kunci: aktivitas antikanker; Callyspongia aerizusa; Haliclona fascigera; Lanthella basta

Abstract

Based on previous research, seaweed extract of Eucheuma sp was reported having antitumour activities. Eucheuma cottonii collected from Pari Island (Seribu archipelago) showed toxicity towards Artemia salina. Toxicity test using Artemia salina as bioindicator was a prescreening testing for anticancer agent. The aims of this research were to isolate and identify any potentially anticancer agent from the most toxic fraction of E. cottonii extract. Sample was extracted with 85 % ethanol using maceration method. The crude ethanol exract was then partitioned into n-hexane, chloroform, and ethyl acetate. The most active fraction towards artemia salina toxicity test was then purified using silica gel column chromatography with n-hexane:chloroform (2:3) as eluent. The pure active fraction having LC50 14.80 ppm was contitued of five compounds which identified using Gas Chromatography-Mass Spectrometry as Cyclododecana (3.23%), Tetradecanoic acid (7.10%), Hexadecanoic acid (54.74%), Cholesta-4,6-dien-3?-ol (6.67%), and Cholest-5-en-dien-3?-ol (Cholesterol) (28.26%).

Abstract

The isolation and identification of antibacterial and toxic compound on Coleiis scutellarioides [L.] Benth. has been conducted. The extraction applied was maceration method using ethanol 85 %. The ethanol extract then partitioned using three kinds of solvents namely n-hexane, chloroform, ethyl acetate respectively. The most toxic and active antibacterial extraction was separated by chromatography column with n-hexane: chloroform as the mobile phase. Identification of the active isolate was conducted by gas chromatography-mass spectroscopy with the aim of'Wiley 229.L database. Toxic test and antibacterial activity test showed that FB2 was proved to be the most toxic and active antibacterial one. The most toxic and active antibacterial fraction of FB2 showed a retardation area of 66,98 mm2 to Micrococcus luteus and 54,43 mm2 to Escherichia coli, and LC50 at 90,72 ppm. Gas chromatography-mass spectroscopy data show that the FB2 fraction contains 4 compounds namely palmitate acid (24,41%); stearate acid (19,53%); 9-oktadekenamida (22,95%); and Esther dioctyl hexadioat (33,11 %»)•

Abstract

Infectious diseases caused by Staphylococcus aureus and Escherichia coli can be treated by milk banana peels (Musa × paradisiaca L.). This study aims to reveal the antibacterial activity of milk banana peel ethyl acetate extract against Staphylococcus aureus and Escherichia coli, and to identify their active compounds. Extraction was conducted by maceration at room temperature, followed by partition. Separation was carried out by column chromatography with mobile phase of chloroform: ethyl acetate (7.5:2.5) and stationary phase of silica gel 60. Antibacterial assay was performed by well diffusion method and identification of active compounds was analyzed by UV-Vis spectrophotometry and LC-MS/MS. Extraction of the peel powder produced methanol extract, partition of the methanol extract resulted n-hexane, ethyl acetate, n-butanol, and water fractions. Antibacterial activity assay shows that ethyl acetate extract was the most active to inhibit the growth of both bacteria. Separation with column chromatography resulted 5 fractions. Identification of the most active fraction with UV-Vis showed that the isolate gave maximum absorption at λ 339.00 nm (band I) and λ 262.00 nm (band II) which were thought to be flavonol (3-OH substituted). Analysis with LC-MS/MS shows that the most active fraction contained 3-methacrylate flavonols and 3-(furan- 2yl) acrylate flavonols.
