Abstract

This study concentrated on the effect of Saccharomyces cerevisiae Hansen extract on gosipol content of Gossypium hirsutum L callus cultures. There were many ways commonly used to obtain high secondary metabolites product by using tissue culture,i.e. elicitation.The objective of this study was to increase the production ofgossypol on G. hirsutum callus cultures using elicitor derived from S. cerevisiae.The callus grew optimally and produced gossypol on Linsmaierand Skoog (LS) medium with the addition of 10 M NAA and 10 M 2,4-D. Callus derived from cotyledon was subcultured five times.Callus was then elicited with elicitor derived from autoclaved S. cerevisiae and concentration of elicitor tested were 0.5, 1.0 and 2.5% (b/v). The harvesting times were 0, 2, 4, dan 6 days after elicitation. The gossypol was analyzed qualitatively and quantitatively by using High Performance Liquid Chromatography (HPLC).Gossypol content was influenced significantly by concentration of elicitor and harvesting time.The maximum ofgossypol content was obtained on the 4 day after elicitation i.e. 469.233 ± 2.332 pg/g dry weight in the elicited callus of G. hirsutum.

Abstract

Field dodder fCuscuta campestris Yunck.)is an obligate parasite plant (holoparasit) with twining stems that coil and fasten to host plants.The qualitative and quantitative studies were tested to 20 twining stems pieces, that infected on host plants (Durantha sp.).The results showed that the twining stems are very sensitive and responsive to the contact of host canopy, but it is not for grasses (Poaceae).A single piece has been found to grow at rate of more than 1 centimetres per day. The ability and speed of reaction of the twining stems by the contact stimulating to their host (haptotropism) were discussed.

Abstract

Study on the productivity of two species of local crickets; Gryllus testaceus Walk and Gryllus mitratus Burn which are generally found in the local market have been conducted with the main aim at obtaining information on the capability of those species in producing eggs including the length of production cycles.Three levels of box square density (50 female/box,100 female/box and 150 female/box) and two levels of additional feed (green master and papay leaves) were used as treatments.The results showed that the number of eggs produced by G.mitratus were larger with longer production cycles compared to those of G.testaceus. Species and density has highly significant effects on egg production and feed consumption, whereas,additional feed significantly influencing egg production and feed consumption of the two species.The use of green master as additional feed resulted in a higher egg production than those given papay leaves.

Abstract

The development of method to explore natural chemical product initially from plants were started from the beginning and generously from time to time from the conventional ones until sophisticated equipments.The value of plants as food,cosmetic and medicines resources needs specific instruments and methods for determining the quantity as well as quality of the materials in plants.This paper describe in detail one of the sophisticated equipments commonly used for analyzing and isolating bioactive compounds the so called High Performance Liquid Chromatography (HPLC).

Abstract

Various biological aspects o/Telenomus sp.(Hymenoptera: Scelionidae) an eggs parasite of the rice stem borer Scirpophaga incertulas (Lepidoptera: Pyralidae) was conducted in the Pest Laboratory for Crop Institute, Bogor, Indonesia.The results showed that the parasite is solitary in habit and mainly live in eggs of the rice stem borer.The arfu Telenomus could maintain longer life span (3.4 days) in the cage and produced a large number of progenies (29.4 individuals) when they were given food of 10% honey solution compared to those which were given only water (2.1 days of life span, and 22.4 individual progenies).The parasite life span and the number of progenies were the least (1.8 days of life span, and 18.8 individual progenies) when they were treated without any food.The number of progeny was also the greatest when they were provided with one day old of stem borer eggs (45.87 individuals), while the number of progeny declined when they were provided with 2 days (36.93 individual progenies) and 3 days (24.87 individual progenies) old stem borer eggs respectively. Pairs of day old parasites are able to parasite more host eggs compared to that of successive days old parasites.The sex ratimf 2$ : 2$ produced more progenies compared to that with the sex ratio IS : 2$ and 2$ : lQ.Parthenogenetic reproduction in arfu Telenomus parasite being tested resulted in the male progenies only.

Abstract

Impatiens arriensii (Zoll.) T.Shimizu is endemic to Madura island. This species is confined only to certain locations on this island.It is remarkable in having four locular ovary with the fusing together of all four lateral petals.The populations of I.arriensii in Madura island was found to be extinct in 1999, (presumably) due to the habitat disturbance.Detailed description and illustration are provided.

Abstract

Antipyretic effect of alcohol extract ofilalang flmperata cylindrica L) root on white rat males had been studied using fever tolerance method.Experimental animals were induced to fever by injecting 0.9 ml/kg BW vaccine kotipa orally.Alcohol extract 5, 10, 30, 50,100 and 150 mg/kg BW was given one hour after injection.The body temperature was measured before injection and every half an hour until the fifth hour after injection.Paracetamol 200 mg/kg BW was used as control.Data were analyzed using ANOVA with the body temperature (T), the time interval (P) and extract doses being given (C) as parameters.The results showed that body temperature increased TC from the basal temperature at the third hour after injection of vaccine.The body temperature did not change significantly at the lower extract doses(5-30 mg/kg BW)(P>0.05),while higher doses (30-150mg/kg BW) linearly lowered the body temperature significantly (P<0.05).The extract dose at 50 mg/kg BW lowered the body temperature similar to paracetamol 200 mg/kg BW.

Abstract

Based on the distribution data of 13 species of Curcuma found in Java,spatial analyses using land status and land used thematic maps has been earned out.The purpose of this analysis is to detenvine thehabittat that usually Curcuma grows both cultivated or wild.Cluster analysis based on distance Eucledian using Statistical Program for Social Sciences(SPSS) for Window Release 6 software was administered to classify the relation between Curcuma species and their habitat.The result showed that Curcuma domestica; C. aurantiaca, C. aeruginosa, C. zedoaria, C. xanthorhiza and C.viridifolia are wide distributed in Java, but C. mangga, C. heyneana, C. purpuracens, C. soloensis, C. euchroma, C.colorata and C. borg are strickly found in specific areas only. Eventhough C. phaeocaulis, C. sylvatica and C. ochrorhiza has been reported found in Java, that species should be monitor due to limited information of this species in Java.Spatial analyses using ArcView software showed that Curcuma found in the 13 kinds of land status and land used.Curcuma mainly occupies in the habitat that classify as a "daratan" status withlanduse status as resettlement.Beside that Curcuma are also grown in the teak forest areas, but some of them have been changed to secondary forest after the timber harvested proven by landuse thematic map.Those areas are classified as a high-risk habitat due to probability status can be changed easily.Cluster diagram analyses using land status thematic maps, showed that there are some habitat similarities among C. xanthorrhiza and C. zedoaria, and then among C. aeruginosa, C. aurantiaca, C. viridiflora and also among C. borg, C. soloensis, C. mangga, C. heyneana, C. colorata, C. euchroma and C. purpuracens. Cluster diagram analysis using land use thematic showed that maps some similarities habitat also occurred betweenC. aurantiaca and C.aeruginosa, and then between C. zedoaria, C. xanthorhiza and C. viridifolia, and also between C. mangga, C. heyneana,C. purpuracens, C. soloensis, C. euchroma, C. colorata and C. borg.

Abstract

Efficiency of biodegradation of organic carbon and ammonium of sawit palm oil industry wastewater was investigated by using batch reactor with fluidized porous support particles.Two bioreactors, each had an effective volume of 3.0 L, were operated in parallel. Bioreactor-l is a batch reactor with fluidized porous support particles and bioreactor-ll is a suspended growth reactor.The wastewater consisting of 1500-2000 mg/L COD and 50-60 mg/L nitrogen was fed by fill and draw mode with one cycle per day,where 1.5 L treated water was drawn before adding the new wastewater.Efficiency of COD degradation and nitrification were calculated by measuring concentration of COD, ammonium-N, nitrite-N and nitrate-N.The experimental results showed that efficiency of COD degradation and nitrification tend to be higher in the bioreactor-l than in the bioreactor II.The highest efficiency of COD degradation in the reactor-l was 83.7% and in the bioreactor-ll was 63.44 %.The highest efficiency of nitrification in the bioreactor-l and bioreactor-ll was 76.72% and 56%, respectively.Ammonium removal occurred in the bioreactor might not only by biological nitrification, but also by other phisical or chemical processes.
