Abstract

Nine resveratrol oligomers have been isolated from the stem bark of Hopea mengarawan i.e. diptoindonesin D (1), balanocarpol (2), ampelopsin A (3), hopeaphuran (4), heimiol A (5), parviflorol (6), a-viniferin (7), isohopeaphenol (8), and vaticanol B (9).The structures of these compounds were determined based on spectroscopic evidence such as UV, IR, 1-D, 2-D NMR and comparison with the reported data. Those compounds implied that Hopea tends to produce resveratrol dimers. Moreover, in subgenus level, besides ampelopsin A (3) and isohopeaphenol (9), resveratrol oligomers in H. mengarawan the same as in H.dryobalanoides. It indicated that H. mengarawan and H. dryobalanoid.es came from the same subgenus.

Abstract

The aim of this study was to evaluate the effects of 2,4-dichlorophenoxyacetic acid (2,4-D) and N -benzyladenin (BA) to embryogenic callus induction of meristem culture in ginger {Zingiber officinale Rose). Completely Randomixed Design was applied using factorial pattern, replicated 3 times. Two factors were assigned i.e: 2,4-D (1.0, 2.0 and 3.0 mg/L) and BA (0.0, 1.0, 3.0 and 5.0 mg/L). Result showed that 2,4-D and BA significantly affected the embryogenic callus induction. The highest embryogenic callus (93.33%) was achieved on MS (Murashige & Skoog) containing combination of 1.0 mg/L 2,4-D and 3.0 mg/L BA, after 8weeks incubation.Histological evaluation informed that the incubation of embryogenic calli started from cortex cells adjacent the meristem vascular tissues.

Abstract

Purwodadi Botanic Gardens-LIPI (The Indonesian Institute of Sciences), East Java, is an ex-situ conservation area accommodating various living plant collections.Identification of the parasitic plants conducted in August 200S found five species parasiting 10S trees belonging to SI species of 24 genera. The highest parasite frequency was found on Ficus (Moraceae). The mistletoes found in the study area were Dendrophthoe pentandra (L.) Miq. (Loranthaceae), Macrosolen tetragonus (Bl.) Miq. (Loranthaceae),Scurrula atropurpurea (Bl.) Dans. (Loranthaceae), Viscum articulation Burm. f. (Viscaceae), and Viscum ovalifolium DC. (Viscaceae).The damaging effect of the parasitic mistletoes generally occurred on the distal part of branches or twigs of the host tree species.

Abstract

DNA isolation is an important issue in molecular fields especially for specific plant group such as Pinanga (Palmae). Typically,leaflets structure of Pinanga consists of shiny leaflets as common species of palms and mottled leaflets. Pinanga javana and P.coronata form commonly the two types of leaflets. The high quality of DNA of Pinanga with the shiny leaflet is readily purified and that with the mottled leaflet needed DNA purification due to the presence of high polyphenolic compound. Application of the silica binding method was recommended to purify the template DNA. Modified DNA-isolation protocol is relatively quick, simple,least expensive, minimum equipments and chemicals required, and suitable for PCR and endonucleases digestion reaction.

Abstract

Terpenoid is a fundamental cells constituent in living organisms. In living organisms, terpenoid biosynthesize via classical pathway of mevalonate and via deoxyxylulose diphosphate (DXP). The distribution patterns of both pathways are unique and specific in living organisms. In human and animal, terpenoid biosynthesize via mevalonate and IPP isomerase type I enzyme system. In plant, terpenoid biosinthesize via combination of the mevalonate and DXP pathways with IPP isomerase type I enzyme system. In parasitic protozoa like Plasmodium falciparum and in major human bacterial pathogen such as Mycobaclerium tuberculosis, Salmonella typhii and Helycobacter pylori, terpenoid biosynthesize via DXP and repeatitive IPP isomerase type II enzymesystem. The specific diversity of the terpenoid biosynthetic pathways in living organisms can use as targets for development ofnovel antiinfective drugs.

Abstract

Previous studies shown that Indonesian Thin-Tailed (ITT) sheep are more resistant to infection than Merino sheep infected with Fasciola gigantica. Eosinophils isolated from ITT sheep between 2-6 weeks postinfection could effectively kill immature parasites in vitro by superoxide radicals produced by these cells. The present study was undertaken to identify immunological mechanisms associated with such resistance by comparison of immue responses to fluke antigen in ITT and Merino sheep, a breed susceptible to infection with F. gigantica. This knowledge will enable the development of targeted control strategies using immunomanipulation such as by vaccination.Two critical observations from this in vivo studies were made including (1) eosinophils were significantly elevated at the time of F. gigantica parasite killing in the ITT host relative to the F. giganfica-susceptible Merino host, (2) IgG, responses correlated with resistance to infection in the ITT host.
