Abstract

The study of stomata characteristics of plantain and horn plants has been carried out. This study aimed to compare the characteristics of plant stomata by plantain and horn banana AAB genome. Therefore, it is possible to find out the similarities and differences in characteristics between plantain and horn banana that have the same genome, including by stomata location, cover cell shape, stomatal type, number of epidermal cells, number of stomata cells, stomata length, stomata width, stomata area, stomata index, and stomatal density. Stomata observations were made by making an incision using the whole mount method. The results obtained were then analyzed descriptively qualitatively by describing the stomata characteristics of the plantain and horn banana plants

Abstract

The most important problems in using polymerase chain reaction (PCR) are the efficiency of energy, cost and time due to gene amplification. Oligonucleotide primer design of sucrose synthase gene was conducted as a model of preliminary experiment to amplify gene using PCR. In plant cells, this gene plays an important role in carbohydrate metabolism, a sucrose molecule break down into glucose. This design involved some computer software as bioinformatics tools. Five data sequences of legumes were downloaded from gene bank using accession number of AF030231, AJ311496, X92378, X69773, and D10266 belongs to soybean, pea, alnus bean, fava bean, and mung bean, respectively. After sequences alignment, some conservative regions were determined as the basis to construct forward and reverse primer candidates. Furthermore, the candidates were tested for compatibility. The results showed that the oligonucleotide primers can amplify sucrose synthase gene with ± 1462 bp fragment size using 5’-AACTTTgTgCTTgA-3’ and 5’-TCCTTTgACTCCTTC-3’ for forward and reverse primer, respectively. Even the PCR process weren’t applied, those primers might be universal primers to amplify sucrose synthase gene of legume plants.

Abstract

The Durian Tengkurak (Durio tanjungpurensis Navia) is one of the endangered exotic species in the Malvaceae family. The species is important for conservation of the germplasm and is considered a potential genetic resource for the development of durian in the future. The objective of this research project was to assess the molecular diversity of D. tanjungpurensis in West Kalimantan, based on Inter Simple Sequence Repeat (ISSR) markers. We applied ten ISSR primers to reveal the genetic diversity of 60 individuals from six natural endemic D. tanjungpurensis populations. The genetic diversity parameters were estimated based on binary data about PCR products (present or absent bands). The results showed that the mean number of observed alleles, the mean number of effective alleles, the genetic diversity, the Shannon’s Information Index score, the number of polymorphic loci, and the percentage of polymorphic loci were 1.53, 1.29, 0.17, 0.26, 77.83, and 52.59, respectively. An analysis of molecular variance (AMOVA) showed that the genetic diversity within a population (65%) was higher than that found between the populations (35%). UPGMA clustering and principal coordinate analysis, based on the DICE similarity matrix, were used to classify the populations into three groups: 1) Hutan Rejunak and Tembaga, 2) Bukit Merindang, and 3) Hutan Rawak, Bukit Sagu 1, and Bukit Sagu 2. Further analysis of the population structure using STRUCTURE software was used to classify all the individuals into two major categories, thus uniting Groups 2 and 3 as one major category. In conclusion, a high level of genetic diversity in the Durian Tengkurak was revealed utilizing the ISSR markers employed in the study.

Abstract

Digital literacy has been supported by the development of information technology in Industry 4.0 which has significantly changed and enabled many opportunities for advancement in the community learning dimension. The system connected to the internet network and the availability of information (i.e. science, technology, and art) is a golden opportunity to create the achievements of the marine-isle community, which is always under the stigma of being fallen behind, so that they can get out of the poverty trap into a space of empowerment and mental readiness to enter the era of a new revolution. The approach were used socio-scientific interpretive and narrative method. This article aim is to reflect and capture the existence of the marine-isle community to inspire and motivate young learners to take advantage of easy access to information, and pinned a number of mentality attitudes (soft skills) to achieve success in life. A number of high awareness attitudes in changing the way of thinking and behavior patterns to motivate persons and our society. Thus, at least at the social level, inherent values, hidden internally to develop into young learners with digital characters who are adaptive, proactive, and dynamic. Otherwise, with anxiety, the community will be crushed and shackled by the hustle and bustle of digital advances

Abstract

Apis cerana Fabricius is a honeybee commonly cultivated in Indonesia to produce honey. Additional feed can acts as a controller of honeybee colonies to protect them from migrate, in uncertain local weather conditions. Providing additional feed based on ‘tongka langit’ banana, Musa troglodytarum L. which have a high β-carotene nutritional content influences the adaptation, eating, and resting behaviors of Apis cerana. This study describes and analyses those three behaviors of Apis cerana to the provision of the feed. The study was conducted for 10 days with the provision of three treatment feeds, namely K+ (sugar water), P1 (fresh banana pulp), and P2 (banana paste) on the first day, and data collection was carried out the next day with a total of 5 times data collection on the number of individuals. Data represents for three types of behavior parameters (adaptation, eating, and resting) will be subjected to a two-way analysis of variance and continued with Duncan's test. Of the three treatments, result was found that P2 has the largest number of individuals in adaptation behavior, while in eating behavior it showed that K+ (as control) has the largest. Treatment K+ and P2 were also not significantly different, after Duncan’ comparison test. The number of individuals of resting behavior from P2 treatment observed on ninth day slightly increased to that of K+ treatment. This is reflected that P2 has potential as additional natural feed for Apis cerana, with high β-carotene content, then might inclusively turn to the honey, even in low levels.

Abstract

Suplementary feed based on the flesh of tongka langit bananas has never been done, even though tongka langit bananas are very qualified as supplementary feed because there is 55% glucose as a substitute for nectar and other similar ingredients and the presence of β-carotene in these bananas as a precursor of vitamin A which is expected to be present in honey when given additional feed based on tongka langit banana. This study aims to determine the effect of giving tongka langit banana-based supplementary feed on the β-carotene content of Apis cerana (F.) bee honey. The research method used was experimental with a Completely Randomized Design at the A. cerana bee farm to obtain data related to feed consumption in four treatments located in Eri Village, Nusaniwe District, Ambon City, Maluku. Then the composition of honey in the form of β-Carotene and vitamin C was further analyzed using the KCKT method of normal phase chromatography (normal phase chromatography) related to the concentration of the four treatments at the Laboratory of the Bogor Agricultural Postharvest Research and Development Center. The results of the analysis of β-carotene content using the HPLC method showed that the highest β-carotene content was in the tongka langit banana feed of 2.275 mg/100g followed by fresh ripe tongka langit bananas of 2.198 mg/100g while in the water + sugar diet it was 2.075 mg/100g and natural feed without treatment as the lowest β-carotene content, namely 1.975 mg/100g. Abstract

Infrared (IR) spectroscopic analysis systems provide advantages in characterizing organic compounds and polymeric material formulations. In this study, an FTIR analysis of makila wood-activated charcoal (Litsea Angulata B.L) was conducted to determine the relationship between wavenumber and the type of makila activated charcoal and the correlation of wavenumber with the quality of makila wood activated charcoal. Makila wood-activated charcoal is made in 2 stages: carbonization at a temperature of 500 ºC for 8 minutes and then starting at a temperature of 700 ºC for 60 minutes. The results of the independence test (14.722) showed that at the α=0.05 level (24.992), the wavenumber was not related to activated charcoal. The correlation between wavenumber and iodine adsorption capacity is negative and tends to be below; in the area of ​​wave number 400-1500 cm-1, it is -0.5476, and in the area of ​​wave number 1500-2000 cm-1 is -0.3931. While in the wavenumber region, 2500-4000 cm-1 is positive and tends to be low at 0.3660, and in the wavenumber region 2000-2500 cm-1, the value is zero (0) or there is no correlation. The correlation between wavenumber and benzene absorption is positive and tends to below, in the area of ​​wave number 400-1500 cm-1, it is 0.1902, and in the region of wave number 1500-2000 cm-1 is 0.3271. While in the wavenumber region 2500-4000 cm-1, the correlation is negative and tends to be high at -0.8310, and in the wavenumber region 2000-2500 cm-1, the value is zero (0) or there is no correlation.

Abstract

Plant cell differentiation can only be understood visually by students through observation with a microscope. The ability of junior high school students to use microscopes is still limited so that practicum activities for making preparations are needed. This activity aims to improve student understanding and skills about monocular and binocular microscopes and making plant preparations to observe stomatal and epidermal cells. The implementation method includes lectures, interactive discussions, question and answer sessions, and making fresh plant preparations and proper microscope usage techniques. The results of the activity showed that participants were able to operate a microscope, make fresh plant preparations, and observe cells. The level of enthusiasm of the participants was quite high, especially when they managed to observe stomatal and epidermal cells directly through the microscope.
