Abstract

Teh hijau diketahui mengandung senyawa polifenol, yang sangat mendukung aktivitas antioksidannya. Aktivitas tersebut membuat ekstrak teh hijau bisa dimanfaatkan secara oral ataupun topikal. Pada penggunaan topikal, perlu dilakukan upaya untuk meningkatkan kemampuan penetrasi perkutan, salah satunya dengan pengembangan sediaan nanoemulsi. Penelitian ini bertujuan untuk mengembangkan sediaan nanoemulsi antioksidan mengandung ekstrak teh hijau dengan minyak calendula sebagai fase minyak dengan karakteristik yang baik. Teh hijau diekstraksi dengan metode refluks menggunakan etanol 96% sebagai  pelarut. Pengujian aktivitas antioksidan ekstrak dilakukan dengan metode peredaman DPPH. Ekstrak selanjutnya dikembangkan menjadi sediaan nanoemulsi dengan menggunakan surfaktan tween 80 dan kosurfaktan PEG400. Hasil penelitian menunjukkan, ekstrak etanol teh hijau memiliki aktivitas antioksidan yang sangat kuat dengan nilai IC50 2,14 ± 0,01 ppm. Ekstrak etanol berhasil dikembangkan menjadi sediaan nanoemulsi dengan menggunakan minyak calendula sebagai fase minyak, pada perbandingan minyak dan Smix (campuran surfaktan dan kosurfaktan) 1:12  dan perbandingan surfaktan dan kosurfaktan 2:1. Sediaan nanoemulsi teh hijau memiliki karakteristik fisik yang baik, ditandai dengan  penampilan yang transparan,  ukuran globul  15,97  ± 0,49 nm dan nilai indeks polidipersitas  0,29 ± 0,01. Sediaan nanoemulsi juga stabil merujuk pada pengujian heating cooling, sentrifugasi, dan  freeze thaw. Telah berhasil dikembangkan sediaan nanoemulsi mengandung minyak calendula dan ekstrak teh hijau dengan karakteristik dan stabilitas fisik yang baik.

Abstract

White tea and pomegranate seed oil are known to contain many bioactive compounds that make them potentially have antioxidant and photoprotective activities. This study aimed to examine the antioxidant and photoprotective activities of white tea leaf extract and develop it in an emulgel dosage form. White tea is extracted with 70% ethanol at 600C. The extracts were tested for antioxidant activity by the DPPH method and for photoprotective activity by the Mansur method. White tea extract was developed into an emulgel with pomegranate seed oil as the oil phase, stearic acid and TEA as emulsifiers, and viscolam mac 10 as a gelling agent. The results showed that the ethanol extract of white tea had a very strong antioxidant activity with an IC50 value of 10.80 ± 0.75 ppm. White tea ethanol extract also has photoprotective activity with an SPF value of 12.35 ± 0.63, %Te value of 2.13 ± 0.01 %, and %Tp value of 45.13 ± 0.08 % (ultra protection category). White tea extract has been successfully developed into an emulgel preparation with good characteristics and stability in terms of organoleptic, pH, viscosity, spreadability, and rheological behavior. White tea emulgel are stable based on centrifugation and freeze-thaw tests.

Abstract

Diabetic foot infection (DFI) is a common complication of diabetes, most frequently caused by Staphylococcus aureus. Green tea contains antibacterial compounds and may serve as a potential supportive therapy for DFI. This study aimed to develop a spray gel formulation containing green tea ethanol extract and evaluate its antibacterial activity against S. aureus. The extract was obtained using 96% ethanol through the reflux method and subsequently tested for antibacterial activity using the agar well diffusion method. The extract was incorporated into a spray gel formulation with Viscolam MAC 10 as the gelling agent. The resulting spray gels were evaluated for their physical properties and antibacterial activity. Green tea extract demonstrated antibacterial activity against S. aureus with a minimum inhibitory concentration (MIC) of 0.1%. Furthermore, the ethanol extract was successfully formulated into a spray gel using 5% Viscolam MAC 10, meeting quality requirements in terms of organoleptic properties, pH, viscosity, spray pattern, spray weight, drying time, and adhesion time. Formulations containing 0.75% and 1% GTE exhibited potent antibacterial activity against S. aureus. In conclusion, a green tea–based spray gel formulation with good physical characteristics and potent antibacterial activity has been successfully developed.
