ABSTRAK Daun Kirinyuh (Chromolaena odorata L.) dapat dimanfaatkan untuk pengobatan secara tradisional sebagai penyembuhan luka, penyakit infeksi dan antibakteri. Tujuan dari penelitian ini adalah untuk mengetahui kandungan senyawa kimia dari daun kirinyuh serta mengetahui aktivitas antibakteri ekstrak metanol daun kirinyuh (Chromolaena odorata L.) terhadap bakteri Staphylococcus epidermidis. Uji skrining fitokimia meliputi flavonoid, alkaloid, fenol, saponin, steroid dan tanin. Pengujian aktivitas antibakteri dilakukan dengan metode difusi sumuran dengan menggunakan 5 seri konsentrasi yaitu 15%, 30%, 45%, 60% dan 100% dan kontrol positif klindamisin 2µg/disk, dan kontrol negatif menggunakan Na-CMC 0,5%. Hasil skrining fitokimia menunjukkan bahwa ekstrak metanol daun kirinyuh (Chromolaena odorata L.) positif mengandung flavonoid, fenol, alkaloid, steroid, saponin, dan tanin. Hasil uji menunjukan bahwa konsentrasi 15%, 30%, 45%, 60% dan 100% ekstrak metanol daun kirinyuh (Chromolaena odorata L.) termasuk dalam kategori zona hambat sedang dengan rata-rata zona hambat yaitu 6,41 mm; 6,86 mm; 7,16 mm; 7,97 mm; dan 9,35mm. Analisis data uji Mann Witney memiliki adanya perbedaan aktivitas antibakteri ekstrak metanol daun kirinyuh (Chromolaena odorata L.) dengan konsentrasi 15%, 60% dan 100% terhadap bakteri Staphylococcus epidermidis dengan kategori sedang. ABSTRACT Kirinyuh leaves (Chromolaena odorata L.) can be used for traditional medicine as wound healing, infectious diseases and antibacterial. The purpose of this study was to determine the chemical compound content of kirinyuh leaves and to determine the antibacterial activity of methanol extract of kirinyuh leaves (Chromolaena odorata L.) against Staphylococcus epidermidis bacteria. Phytochemical screening tests include flavonoids, alkaloids, phenols, saponins, steroids and tannins. Antibacterial activity testing was carried out using the well-diffusion method using 5 series of concentrations namely 15%, 30%, 45%, 60% and 100% and the positive control was clindamycin 2µg/disk, and the negative control was using 0.5% Na-CMC. The results of the phytochemical screening showed that the methanol extract of kirinyuh leaves (Chromolaena odorata L.) positively contained flavonoids, phenols, alkaloids, steroids, saponins and tannins. The test results showed that the concentrations of 15%, 30%, 45%, 60% and 100% of the methanol extract of kirinyuh leaves (Chromolaena odorata L.) were included in the medium inhibition zone category with an average inhibition zone of 6.41 mm; 6.86mm; 7.16mm; 7.97mm; and 9.35mm. Analysis of Mann Witney test data showed differences in antibacterial activity of methanol extract of kirinyuh leaves (Chromolaena odorata L.) with concentrations of 15%, 60% and 100% against Staphylococcus epidermidis bacteria in the medium category.
