Abstract

Background: Increased activity of Adenosine Monophosphate-Activated Protein Kinase (AMPK) can inhibit cell growth and biosynthetic pathwaysin some cancer cells. Recently, several endogenous ligands have been identified to activate AMPK including Adenosine Monophosphate (AMP).Virtual screening is one of the recent technique, which is used for drug discoveries.Therefore the aim of this study was to identify active compounds of Indonesian herbal plants that activate AMPK by using molecular docking. Methods: Samples used in this study wereall of active compounds which were derived from Indonesian herbal plantsand had three-dimensional structures. Molecular docking was performed by using AutodockVina version 1.1.2 to evaluate interaction between AMPK and active compounds. The AMP structure obtained from protein data bank (www.rcsb.org/pdb)was used as a molecule standard of AMPK interaction. Docking score indicated affinity of molecule interaction and was determined in Cal/mol. To visualize docking results that fit to the AMPK molecule, chimera software version 1.9 was used. Results: Curcumin has docking scores as same as docking score of AMP (-7.5 Cal/mol) and has hydrogen bound in γ subunit of AMPK (Thr86, Thr88, and Arg117). Conclusions: Curcumin might be an activator of AMPK. Another molecular docking which has a flexible substrate-enzyme algorithm is required for confirming the docking result. Abstract

AbstractIndonesia has the sixth largest number of people with diabetes in the world. This study aimed to investigate association of age of menarche, duration of diabetes, vitamin c (VC) intake and psychological stress with fasting blood glucose (FBG) levels in adult women with type 2 diabetes mellitus (T2DM). Psychological stress was measured using a STAI questionnaire, while anthropometry, duration of diabetes and age of first menarche were assessed using open questionnaires. Vitamin C intake and FBG level were measured using SQ-FFQ and the hexokinase method respectively. Data were analyzed using Spearman Rank correlation and multiple linear regression tests. A total of 188 adult women with T2DM had mean age 53±5.31 years old, Body Mass Index (BMI) 25±3.76 kg/m2, age at menarche 13.8±1.71 years old, duration of diabetes 4.4±3.38 years and VC 56.9±46.79 mg/day. Body Mass Index (r=-0.190 p=0.011) and VC intake (r=-0.153 p=0.049) were associated with FBG levels. While, age of menarche, duration of diabetes and psychological stress were not associated with FBG levels.AbstrakIndonesia menempati peringkat keenam terbesar jumlah penderita diabetes melitus (DM) di dunia. Tujuan penelitian ini adalah menganalisis hubungan usia menarche, lama menderita DM tipe 2 (DMT2), asupan vitamin c (VC) dan stres psikologis dengan kadar glukosa darah puasa (GDP) pada perempuan dengan DMT2 Stres psikologis diukur menggunakan kuesioner STAI sedangkan data antropometri, lama menderita T2DM dan usia menstruasi dikaji dengan kuisioner terbuka. Asupan vitamin C ditentukan dengan Semi Quantitative Food Frequency Questionnaire (SQ-FFQ) dan kadar GDP serum diukur dengan metode hexokinase. Data kemudian dianalisis dengan menggunakan Korelasi Spearman dan regresi linier ganda. Sebanyak 188 subjek memiliki rata-rata usia 53+5,31 tahun, IMT 25+3,76 kg/m2, lama menderita DMT2 4,4+3,38 tahun, usia mesntruasi 13,8+1,71 tahun dan asupan VC 56,9±46,79 mg/day. Indeks Massa Tubuh (r=-0.190; p=0,011) dan asupan VC (r=-0,153; p=0,049) berhubungan dengan kadar GDP. Sementara itu usia menstruasi, lama menderita DM dan stress psikologi tidak berhubungan dengan kadar GDP.

Abstract

ABSTRACTDrinking plain water can increase daily energy expenditure and decrease total energy intake. This study aimed to analyze the effects of drinking plain water before eating steamed rice with high glycemic index (GI) at breakfast, on body weight (BW) and blood glucose level (BGL) in obese male adolescents. Twenty male adolescents aged 15-18 years old and had BMI for age > 85 percentile participated in this randomized control trial study. Ten male students in G1 group drank 250 ml plain water before eating 180 g steamed rice with high GI and 10 male in the other group (G2) drank the same amount of water after eating the same rice for breakfast for 7 days. Paired and independent t tests were used to analyze BW and BGL in both groups with p value < 0.05. Mean BW change in G1 was significantly different from mean BW changes in G2 (p=0.006), whereas mean fasting and 1h after meal BGLs changes of both groups were similar. Drinking water before having breakfast reduces BW in obese male adolescents.ABSTRAKMinum air putih dapat meningkatkan pengeluaran energi harian dan mengurangi total asupan energi. Penelitian ini bertujuan untuk menganalisis pengaruh minum air putih sebelum sarapan nasi kukus dengan indeks glikemik (IG) tinggi terhadap berat badan (BB) dan kadar glukosa darah (KGD) pada remaja putra dengan obesitas. Dua puluh remaja putra berusia 15-18 tahun dan IMT/U > 85 persentil berpartisipasi dalam penelitian randomized control trial study ini. Sepuluh remaja putra kelompok K1 minum 250 ml air putih sebelum sarapan 180 g nasi kukus dengan IG tinggi dan 10 remaja putra kelompok K2 minum air putih dalam jumlah yang sama setelah sarapan nasi yang sama selama 7 hari. Uji t berpasangan dan independen digunakan untuk menganalisis BB dan KGD pada kedua kelompok dengan nilai p <0,05. Rata-rata selisih perubahan BB pada kelompok K1 berbeda secara signifikan daripada rata-rata selisih perubahan BB pada kelompok K2 (p = 0,006) sedangkan rata-rata KGD puasa dan 1 jam posprandial sama pada kedua kelompok. Kesimpulan: Minum air putih sebelum sarapan nasi menurunkan BB pada remaja putra dengan obesitas.

Abstract

ABSTRACTBackground: The prevalence of Protein Energy Deficiency (PED) has increased over the past decade. Inadequate food intake and infectious diseases are the primary causes of PED. Clinical management of toddlers with PED often involves costly commercial foods. Ganyong, catfish, and red beans as local food ingredients with high nutritional content have the potential to be additional food for children with PED.Objectives: This study aims to examine the effect of GANIME formula form and it’s efficacy on body mass index (BMI) and albumin levels in the rats model with PED.Methods: The study consisted of two phases: analyzing the proximate content of GANIME and conducting in vivo research using ten male Wistar rats aged 3 weeks (50-100 grams). The rats were randomly divided into two groups, G1 (GANIME pellets) and G2 (GANIME porridge), and were observed for 14 days. Body weight (BW), BMI, and albumin were measured before and after treatment. Paired t-tests were conducted to compare pre-and post-treatment results, while inter-group differences were assessed using One-Way Analysis of Variance (ANOVA) tests.Results: The results showed that the optimal GANIME formula was identified at P3, with the highest protein and energy  content. The G1 group of rats experienced a greater increase in body weight and BMI than the G2 group of rats but the increase was not significant (p > 0.05). Each group did not differ statistically significantly (0.644). Albumin in each group experienced a non-significant decrease (p > 0.05) and the two were not statistically different (0.690).Conclusions: Administering the GANIME formula in pellet form exhibited superior efficacy because it’s consumed more so it can increase body weight and BMI, and produce a lower decrease in albumin levels in rats model with PED. KEYWORD: protein-energy deficiency (PED), albumin, body mass index (BMI), local food, GANIME formula
