Abstract

The potential of aquaculture in this area, especially the Obi Islands, has not been fully utilized. One of the commodities that can be developed is Kappaphycus alvarezii seaweed. This commodity is the main commodity for aquaculture which has important economic value. Analysis of the suitability of waters for seaweed cultivation is an early stage of activity that will determine the success of Kappaphycus alvarezii cultivation. Therefore, this study aims to assess the suitability of the waters for the development of Kappaphycus alvarezii seaweed cultivation on the island of Obi. This research was conducted in the waters of Akegula Village, Kampung Baru, and Kawasi, South Halmahera Regency, North Maluku Province, Indonesia. There are 3 stations in Akegula waters, 3 stations in waters in Kampung Baru waters, 4 stations in Kawasi waters, and 3 stations in Soligi waters. Water quality parameters observed were temperature, brightness, depth, current velocity, salinity, pH, dissolved oxygen, nitrate, and phosphate. The results showed that the waters of Akegula, Kampung Baru, Kawasi, and Soligi were very suitable.Keywords: Kappaphycus alvarezii; Water quality; Obi IslandAbstrakPotensi perikanan budidaya di kawasan ini khususnya kepulauan Obi, belum dimanfaatkan secara maksimal. Salah satu komoditas yang dapat dikembangkan adalah rumput laut Kappaphycus alvarezii. Komoditas ini merupakan komoditas utama budidaya perikanan yang bernilai ekonomis penting. Analisa kesesuaian perairan untuk budidaya rumput laut merupakan tahapan awal keigatan yang sangat menentukan keberhasilan budidaya Kappaphycus alvarezii. Oleh karena itu, penelitian ini bertujuan untuk mengkaji kesesuaian perairan untuk pengembangan budidaya rumput laut Kappaphycus alvarezii di pulau Obi. Penelitian ini dilakukan di perairan Desa Akegula, Kampung Baru, dan Kawasi, Kabupaten Halmahera Selatan, Provinsi Maluku Utara, Indonesia. Terdapat 3 stasiun di perairan Akegula, 3 stasiun di perairan di perairan Kampung Baru, 4 stasiun di perairan Kawasi,  dan 3 stasiun di perairan Soligi. Parameter kualitas air yang diamati adalah suhu, kecerahan, kedalaman, kecepatan arus, salinitas, pH, oksigen terlarut, nitrat dan fosfat. Hasil penelitian menunjukan bahwa perairan Akegula, Kampung Baru, Kawasi dan Soligi sangat sesuai.Kata Kunci: Kappaphycus alvarezii; Kualitas perairan; Analisa Kesesuaian; Pulau Obi

Abstract

Abstract The supply of coconut crabs (Birgus latro Linnaeus, 1767) has been sourced from capture in the wild. Development efforts in aquaculture have succeeded in producing eggs. However, further development with larval maintenance failed because it was constrained by a low survival rate. This study aimed to identify the potential probiotic bacteria in coconut crabs. Coconut crab samples were collected from Moor Island, South Patani District, Central Halmahera Regency, North Maluku Province, Indonesia. The intestines of coconut crabs were taken as much as 1 g, then ground on a porcelain dish and added to Tri-Salt. The scouring liquid was taken in 0.1 ml and spread on a petri dish containing Nutrient Agar. The isolated used were marked with BL1, BL2, BL3, BL4, and BL5. Molecular identification was performed with sequencing and then analyzed using the Basic Local Alignment Search Tool (BLAST). The selection of isolates from the gastrointestinal tract of coconut crabs showed that the bacteria obtained were Enterobacter tabaci (BL1), Enterobacter hormaechei (BL2), Bacillus horneckiae (BL3), Pseudomonas stutzeri (BL4), and Acinetobacter variabilis (BL5). Further testing revealed that the bacteria A. variabilis (BL5) is a probiotic isolate in coconut crabs. Highlight Research The intestinal bacteria from coconut crabs identified were E. tabaci, E. hormaechei, B. horneckiae, P. stutzeri, and A. variabilis. The E. tabaci strain BL1, E. hormaechei strain BL2, B. Horneckiae strain BL3, and P. stutzeri strain BL4 are pathogenic bacteria. The A. variabilis strain BL5 is potential probiotics bacteria. The A. variabilis strain BL5 showed a better survival rate than other isolates.

Abstract

Genetic is key substantial approach conservation, managament and sustainability. This study aims to genetic structure populations Skipjack tuna in North Maluku Sea. Samples collection in Morotai Island (n=10), Central Halmahera District, Weda (n=10) dan South Halmahera District, Bacan, (n=10) and secondary data (n=4) in March-May 2018. Molecular analysis through stages extraction, PCR, electrophoresis and sequencing DNA. DNA sequences analysis used MEGA 5 (Genetic distance and phylogenetic) and arlequin (Fixation index). The result found that fragment length 546 (base pairs) in control mitocondrial DNA. Genetic distance analysis Skipjact tuna population based on primary (North Maluku) and secondary data (Sulu-Celebes and South China Sea, Bali, Indian coast, Kyushu Island Japan) show close genetic 0.037-0.056. Fixation indices (Fst) analysis value 0.801-0.936 the show that weak genetic differentiation between populations. High genetic flow between populations based on genetic distance and Fst. The result show that genetic distance and Fst show that genetic structure populations Skipjack tuna in North Maluku Sea undistrubed. The Skipjack tuna data obtained can uses data base to preserve and sustainability fish resource.
