Abstract

Pengembangan model uji kompetensi pedagogik yang bersifat spesifik sangat diperlukan guna menghasilkan model uji kompetensi yang tepat  karena karakteristik pedagogik antar mata pelajaran berbeda. Penelitian ini bertujuan untuk mengembangkan  model Uji Kompetensi Pedagogik Tata Boga pada Pendidikan Profesi Guru (PPG) Program keahlian Kuliner. Pengembangan model Uji Pedaboga P2M  ini menggunakan prosedur pengembangan dari Plomp (1997)  yaitu terdiri (1) fase investigasi awal; (2) fase desain; (3) fase realisasi; (4) fase uji coba, dan revisi. Uji validitas dilakukan oleh 2 orang ahli yang menilai perangkat dan instrumen pembelajaran/pelatihan dan uji. Tingkat kepakaran antar penilai terhadap hasil penilaiannya dianalisis dengan statistik Coefficient Cohen’s kappa dan percentage of agreement. Subjek penelitiannya adalah mahasiswa jurusan Pendidikan Kesejahteraan Keluarga (Tata Boga).  Hasil penelitiannya adalah: (1) model Uji Pedaboga P2M  memiliki kriteria keefektifan rata-rata 92%, kepraktisan 87%, dan keterlaksanaan 86%. (2) perangkat uji kompetensi seperti modul pembelajaran/pelatihan dan uji dengan kriteria sangat baik (rata-rata 3,73), lembaran cek kemampuan dan rubrik penilaian (justifikasi) peserta diklat/uji dengan kriteria sangat baik (rata-rata 3,62). Model Uji Pedaboga P2M dapat  dijadikan acuan pada  kegiatan uji kompetensi pedagogik Pendidikan Profesi Guru (PPG) yang akan datang sehingga penilaian uji Kompetensi lebih spesifik. Specific development of pedagogic competence test model is highly necessary to generate appropriate competence models due to pedagogic difference between subjects. This research aimed to develop Culinary Art (food processing and presentation) Pedagogic Competence Test model on Teacher Profession Education (PPG) of Culinary Expertise Program. This P2M Pedaboga Test utilized Plomp (1997) development procedures that comprised of (1) initial investigation phase; (2) design phase; (3) realization phase; (4) testing phase, and revision. Validity test carried out by two experts who assessed the devices and instruments of learning/training and test.  Expertise level between the assessors against their assessment were analyzed analyzed using Cohen’s Kappa Coefficient and percentage of agreement statistic. Subjects were Family Welfare (Culinary Art) Education department students. Research result revealed that: (1) Pedaboga P2M test had 92% averageof  effectiveness, 87% of practicality, and 86% of implementation criteria; (2) competency test devices such as learning/training and test module had excellent criteria (3.73 in average), skill check sheets and assessment rubric (justification) of training/test participants had excellent criteria (3.62 in average). Pedaboga P2M Test model could be utilized as reference for future pedagogic competence test activity of Teacher Profession Education for more specific Competence test assessment.

Abstract

Pamelo (Citrus  maxima  (Burm.)  Merr.) besar merupakan salah satu jenis tanaman hortikultura yang mendapat perhatian dari pemerintah untuk dikembangkan dan ditingkatkan produksinya karena memiliki prospek pemasaran yang baik. Permasalahan utama pengembangan tanaman pamelo Aceh adalah ketersediaan biji yang sangat sedikit atau bahkan sering dijumpai tanpa biji dan juga bijinya sulit sekali dikecambahkan dikecambahkan secara konvensional. Oleh karena itu untuk mengatasi permasalahan tersebut dapat dilakukan secara in vitro. Tujuan penelitian ini adalah untuk mengetahui pengaruh BAP terhadap keberhasilan perkecambahan biji pamelo lokal Aceh secara in vitro. Penelitian  dilaksanakan di Laboratorium Kultur Jaringan Tanaman Fakultas Pertanian Universitas Malikussaleh pada bulan Desember 2018 sampai dengan Februari 2019. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) faktor tunggal. Perlakukuannya adalah konsentrasi BAP yaitu 0 dan 2 mg/L dengan 30 ulangan, sehingga didapat 60 satuan percobaan. Pada percobaan ini setiap eksplan ditanam pada botol kultur dengan jumlah 2 eksplan per botol. Data hasil pengamatan dianalisis secara statistik dengan menggunakan analisis ragam. Jika hasil uji F menunjukkan adanya pengaruh yang nyata antar perlakuan, maka analisis dilanjutkan dengan Uji DMRT (Duncan’s Multiple Range Test) pada taraf 5%. Hasil penelitian menunjukkan bahwa pemberian BAP dapat mempengaruhi pertumbuhan eksplan biji pamelo secara in vitro.  Pemberian BAP 2 mg/L pada awal menyebabkan biji lebih lambat membentuk tunas dan akar, namun pada akhir pengamatan didapatkan biji menghasilkan tunas dan daun yang lebih banyak.

Abstract

Acehnese pomelo is considered as a potential fruit to be developed for its economic and nutritional values. However, there is a main problem in developing this fruit. Pomelo sometimes is grown from seeds, but nowadays the fruits are often found to be having lesser seeds or even seedless. Besides, it is quite challenging to grow this local pomelo not only due to the scarce of the seeds, but it also cannot be conventionally germinated. Therefore, we need to develop this fruit through tissue culture. The purpose of this study was to investigate the effect of growth regulators on germination of pomelo seeds in vitro and its organ formation from in vitro root explants seed-originated of pomelo. The study was conducted at Plant Tissue Culture Laboratory, Faculty of Agriculture, Malikussaleh University. In October 2018 to April 2019. The research was divided into two separate experiments: 1) in vitro germination of pomelo seeds and 2) in vitro organ formation from root explants from of pomelo seeds. The first experiment using Complete Randomized Design (CRD) with 10 replications. The only factor observed was the use of BAP (0 and 2 mg/l). The second experiment was the examination of plant organ formation from root explants of pomelo seeds. This experiment had 2 factors observed. The first factor was different concentrations of BAP (0, 1, and 2 mg/l). The second factor was the different concentration of IBA (0, 1.25, and 2.5 mg/l). The results indicated that the application of BAP influenced the germination of pomelo seeds. The application of 2 mg/l BAP attributed to slower growth of shoots and roots, but it demonstrated better number of shoots, roots and leaves compared to application of 0 mg/l BAP. Pamelo root explants can form adventitious shoots on media eventhough without growth regulator (BAP 0 mg/l + IBA 0 mg/l), but optimization still needs to be done in order to obtain the best concentration to initiate shoot formation.

Abstract

Sweet kaffir lime (Citrus hystrix Dc), also called "Boh Kruet Mameh" is one of Aceh's local plants, which is currently very difficult to obtain. Sweet kaffir lime is not like the ordinary kaffir lime, which tastes sour. The fruit of sweet kaffir lime has a larger size than the regular kaffir lime, sweet and sour taste, aromatic and fresh, which can be consumed as a fruit table. The problem of developing the propagation technique of sweet kaffir lime plants is the seeds' condition, which are conventionally difficult to germinate and limited in number, making it difficult to get a high-quality seed. Thus, to overcome these problems, suitable solutions to plant propagation techniques should be sought. As for the right plant propagation technique is tissue culture technology or in vitro propagation technique. This research was conducted at the Plant Tissue Culture Laboratory, Faculty of Agriculture, Malikussaleh University. The research was conducted from February to April 2018. This research used a single randomized complete design (CRD) with ten replications. The treatment is a combination of the basic media, which consists of MS0 (MS media without BAP) and MS1 (MS + BAP 4 ppm). The result showed that the adduction of 4 ppm BAP in MS media influenced the in vitro sweet kaffir lime seeds' growth. Sweet kaffir lime seeds that were given 4 ppm BAP grew buds faster with more percentage of shoot growth, the number of shoots, and a number of leaves than the seeds without BAP adduction.

Abstract

Penelitian ini memiliki tujuan untuk mengetahui tingkat penerimaan panelis dan formula terbaik bir pletok saffronaga latte.  Metode pada penelitian ini yaitu metode eksperimen dilanjutkan uji sensori.  Pada penelitian ini, kulit buah naga merah menggantikan kayu secang pada pembuatan bir pletok, kemudian terdapat penambahan saffron sebanyak 0,5 gr untuk perlakuan 1 (F1);  1 gr untuk perlakuan 2 (F2); 1,5 gr untuk perlakuan 3 (F3). Uji Sensori dilakukan terhadap 35 panelis tidak terlatih. Data dari hasil uji sensori dianalisis menggunakan uji nonparametrik Kruskall - Wallis serta uji lanjut Mann-Whitney pada taraf kepercayaan 95% (α=0,05). Formulasi bir pletok saffronaga latte yang paling diterima oleh masyarakat yaitu bir pletok dengan perlakuan 2 (F2).  Penggunaan kulit buah naga  dan penambahan bunga saffron memberikan pengaruh terhadap penerimaan pada aspek roma, warna dan rasa bir plethok saffronaga latte, tetapi tidak memberikan pengaruh terhadap penerimaan tekstur.

Abstract

The objective of this study was to assess the sensory acceptability of Italian Meringue Butter Cream in terms of texture, aroma, color, and taste, using varying proportions of sweeteners. The research was carried out in the Culinary Arts Laboratory of the Ibu Kartini Academy of Social Welfare. An experimental approach was employed, utilizing a Completely Randomized Design (CRD). The study involved three treatments with different ratios of granulated sugar to stevia: F1 (220 g : 20 g), F2 (150 g : 25 g), and F3 (80 g : 30 g). A total of 35 untrained panelists participated in the hedonic sensory evaluation. The resulting data were analyzed using the non-parametric Kruskal-Wallis test, followed by the Mann-Whitney U test at a 95% confidence level (α = 0.05). The findings revealed that the variation in sweetener proportions significantly influenced the panelists’ preferences regarding aroma, texture, color, and taste. Among all formulations, the F1 treatment received the highest level of overall acceptance.

Abstract

Transferrin receptor (TfR) levels are elevated in various types of cancer cells and correlate with the aggressive or proliferative ability of tumor cells. Therefore, TfR levels are considered useful as a prognostic tumor marker, and TfR is a potential target for drug delivery in therapy of malignant cells. In such kind of targeted delivery system, antibody fragments are frequently used as targeting moiety. Here, we report the generation of an anti-TfR single-chain antibody variable (scFv). The cDNA encoding the variable heavy and light chain domains of the scFv antibody fragment was derived from the anti-TfR monoclonal antibody LUCA31. The gene encoding the anti-TfR scFv fragment was codon optimized for expression in Escherichia coli, subsequently synthesized, and cloned into the pJExpress-804 Rhamex vector. The expression vector utilizes the E. coli rhaB promoter and corresponding regulatory genes, and is tightly regulated by the presence of L-L-rhamnose, it is also tightly regulated in the absence of L-L-rhamnose by the addition of D-glucose The His6-tagged anti-TfR scFv fragment was expressed in E. coli NiCo21 and purified by means of immobilized metal chelate affinity chromatography on TALON™ matrix. In SDS-PAGE, a single band corresponding to a molecular mass of approximately 30 kDa was observed which corresponded to the predicted molecular mass based on the amino acid sequence.

Abstract

An ideal therapeutic for cancer would be one that selectively targets to tumor cells, is nontoxic to normal cells, and that could be systemically delivered, thereby reaching metastases as well as primary tumor. Immunoliposomes directed by monoclonal antibody or its fragments are promising vehicles for tumor targeted drug delivery. Transferrin receptors (TfR) levels are elevated in various types of cancer cells and considered to correlate with the aggressive or proliferative ability of tumor cells. Therefore, TfR levels can be elaborated as a prognostic tumor marker, and TfR is a potential target for drug delivery in the therapy of malignant cells. Here, we report the preparation of an anti-TfR single-chain antibody variable (scFv) immunoliposome for tumor targeted delivery vehicle. The cDNA encoding the variable heavy and light chain domains of the anti-TfRscFv antibody fragment was derived from the murine monoclonal antibody Clone E6, which is specific towards transferrin receptor. The gene encoding the anti-TfR scFv fragment was codon optimized for expression in Escherichia coli, subsequently synthesized, and cloned into the expression vector pJexpress404. The His6-tagged anti-TfR scFv fragment was expressed in E. coli and purified by means of immobilized metal-ion affinity chromatography on TALON™ matrix. SDS-PAGE revealed that the scFv fragment had the size of approximately 27 kDa, which corresponded with the predicted size of the protein based on its amino acid sequence. Liposome containing 5% MPB-DOPE were prepared by ethanol injection method. Afterwards, the anti-TfR scFv fragments were covalently conjugated to the liposome to produce the anti-TfR scFv immunoliposome with the size of around 200 to 300 nm.

Abstract

A facile generation of a recombinant antibody fragment with intrinsic fluorescent properties of the monomeric mCherry fluorescent protein is described. The so-called mCherryBody was designed based on the structure model of the variable fragment of anti-transferrin receptor antibody LUCA31 and the X-ray crystallographic structure of the mCherry protein. mCherryBody was constructed to retain optimal spatial geometry between the C- and N-termini of the antibody light-chain (VL) and heavy-chain (VH) by mimicking the domains interface pairing in antibody Fab fragments and incorporation of the mCherry fluorescent protein as a bridging scaffold. The gene encoding the chimeric protein was cloned into the pJExpress414 expression vector, expressed and secreted into the periplasm of Escherichia coli NiCo21(DE3) for assembly and disulphide bond formation. Based on its amino acid sequence, mCherryBody was predicted to have a molecular weight of 51.46 kDa. The modular assembly used in the generation of mCherryBody may permit the interchange of binding sites and of fluorescent proteins to create robust panels of coloured antibody fragments. Thus, the mCherryBody platform facilitates rapid generation of colored single-chain variable fragment (scFv) chimeras that could be used for screening of antibodies against cell surface markers or receptors.
