Abstract

Severe exposure to ultraviolet (UV) rays leads to skin damage, including hyperpigmentation, freckles, melanoma, age spots, and melasma, all of which are related to the skin pigment enzyme, tyrosinase. Prevention can be achieved by avoiding harsh UV rays and inhibiting tyrosinase catalytic activity. Many compounds have been developed for the treatment of such conditions; however, most come with unwanted side effects. The purpose of this study was to determine the sun protection factor (SPF) value and tyrosinase enzyme inhibitory activity of plant secondary metabolites with high antioxidant activity, namely rutin, catechin, niazirin, piperine, quercetin, and quercitrin, as potential alternatives. Both tests were carried out using UV-Vis spectrophotometry. SPF determination was performed by observing the absorbances across a wavelength range of 290 to 320 nm. The determination of anti-tyrosinase activity was conducted by measuring dopachrome at 490 nm after enzymatic reactions and calculating the IC50 value. In the SPF assay, benzophenone-3 was used as the standard, and piperine, rutin, quercetin, and quercitrin exhibited high protective abilities with SPF values above 30 at 500 µg/mL. In the tyrosinase inhibition assay, kojic acid as the standard showed a strong potential for inhibition with an IC50 of 33.65 µg/mL, while quercetin, rutin, and piperine exhibited weaker inhibitory potential with IC50 values of 178.44, 271.73, and 347.62 µg/mL, respectively. On the other hand, quercitrin and niazirin showed little to no tyrosinase inhibition activity. However, catechin demonstrated more catalytic activity towards the enzyme. In conclusion, quercetin, rutin, and piperine have the potential to be developed as active ingredients to protect the skin from UV-induced damage due to their satisfactory SPF values and tyrosinase inhibition activity.

Abstract

Abstract

Platelet aggregation has the potential to form thrombi and result in cardiovascular system diseases such as myocardial infarction and ischemic stroke - one of the leading causes of death globally. Traditionally, people use binahong leaves as blood thinners. Therefore, this study aims to obtain scientific data on the efficacy of binahong leaf as antithrombotic. The efficacy test was carried out on male Swiss-Webster mice. The results showed that the ethanol extract of binahong leaves at doses of 50 (BLEE50) and 100 mg/kg bw (BLEE100) could increase bleeding time on H7 (7.61±1.79% and 3.72±1.76% vs 1.08±0.90%) and H14 (13.81±4.42% and 5.06±2.30% vs 1.66±1.09%) and coagulation time at H7 (5.01±1.36% and 4.18±1.67% vs 1.38±1.08%) and at H14 (7.92±1.97% and 7.19±1.96% vs 1.70±1.10%) significantly (p<0.05). The two doses of BLEE were formulated in the form of nanoemulsions with the Self Nano Emulsifying Drug Delivery System (NBLEE50 and NBLEE100) were also able to prolong bleeding time and coagulation time significantly (p<0.01) but only NBLEE50 prolonged bleeding time (p<0.05) significantly against BLEE50. In the test of the anti-platelet aggregation effect with ADP as an inducer, both doses of BLEE and the nanoemulsion preparation (NBLEE) could significantly (p<0.01) inhibit platelet aggregation with a percentage of inhibition >70% which was not different from the standard (acetylsalicylic acid). In the antioxidant effect test using the DPPH method, BLEE has an IC50 = 66.08?g/mL which is classified as a strong antioxidant. Both doses of BLEE and its NBLEE could significantly (p<0.01) inhibit lipid peroxidation in plasma and liver and NO radicals formation. BLEE50 can significantly (p<0.05) reduce mean platelet volume (6.05±0.24fL vs 6.55±0.34fL) and platelet distribution width (8.52±0.36% vs 9.25±0.42%). Based on those results, BLEE has the potential to be used as an antiplatelet aggregation and antioxidant.

Abstract

Poor oral hygiene can promote the transition of commensal microorganisms into opportunistic pathogens and contribute to the development of oral diseases. Antimicrobial mouthwashes may serve as preventive or adjunctive therapies in the early stages of oral infections. On the other hand, propolis and roselle (Hibiscus sabdariffa), both rich in phenolic compounds, possess well-documented antimicrobial activities and are promising candidates for natural mouthwash formulations. This study evaluated the antimicrobial activity of the combination of ethanolic extract of propolis (EEP) and aqueous extract of roselle (AER) against Staphylococcus aureus, Streptococcus mutans, and Candida albicans, and developed a combined-extract mouthwash formulation. The MIC values of EEP against S. aureus, S. mutans, and C. albicans were 1280, 640, and >20,480 μg/mL, respectively, whereas AER exhibited MIC values of 320, 1280, and 10,240 μg/mL. The MBC values of EEP were 1280 μg/mL for S. aureus and 2560 μg/mL for S. mutans, while AER showed MBC/MFC values of 1280, 10,240, and 10,240 μg/mL against S. aureus, S. mutans, and C. albicans, respectively. Checkerboard assays indicated additive interactions against S. aureus (FICI = 2) and S. mutans (FICI = 3). A mouthwash containing 2% EEP and 1% AER was successfully formulated, exhibiting a transparent light-brown appearance, menthol aroma, homogeneous characteristics, pH of 5.44 ± 0.03, and viscosity of 1.36 mPa·s. The undiluted formulation achieved complete microbial reduction within 2–10 min of contact. These findings suggest that the EEP–AER combination has potential as a natural antimicrobial mouthwash for oral health management.
