Abstract

Seekor ikan pari cambuk Jenkins jantan tangkapan liar ditemukan mati dalam tangki karantina dengan gejala klinis sebelum kematian adalah nafsu makan berkurang selama seminggu. Riwayat pengobatan yang diberikan adalah pemberian antibiotika enrofloxacin secara peroral. Periode terapi berlangsung selama sepuluh hari. Pengobatan terakhir adalah pemberian Hepavit® (ekstrak hati) dan injeksi intramuskular (IM) antibiotika enrofloxacin. Satu hari sebelum kematian, sampel darah dikoleksi dan kemudian diperiksa untuk mengetahui gambaran hematokrit dan beberapa parameter kimia darah. Hasil pemeriksaan darah ditemukan penurunan kadar blood urea nitrogen (BUN), alkaline phosphatase (ALP), dan alanine aminotransferase (ALT), peningkatan kadar glukosa, penurunan total protein dan kadar albumin, serta peningkatan kadar globulin. Pemeriksaan patologi anatomi ditemukan lesi pada ekor, di sekitar mata, dan clasper. Lesi hemoragik ditemukan di lapisan mukosa esofagus, lambung, dan kolon spiral. Gumpalan darah ditemukan di bawah lapisan tunika organ testis. Kerusakan organ hati secara makroskopis ditunjukkan dengan perubahan warna yang tidak homogen, pembengkakan organ, kongesti hati, dan konsistensi yang rapuh. Berdasarkan diagnosis morfologik, kausa kematian ikan diduga karena mengalami kondisi infeksi septikemia selama beberapa minggu sebelumnya.

Abstract

Bakteri Vibrio spp. adalah salah satu patogen yang umum menginfeksi pembenihan dan pembesaran udang, yang dapat menyebabkan kematian masal benur di panti benih udang. Penelitian ini bertujuan untuk mengetahui dinamika bakteri Vibrio spp. pada air laut di seputar tujuh area sentra pembenihan udang vanamei di Indonesia. Pengamatan dilakukan setiap bulan sejak Januari hingga Desember 2020, dengan menginokulasikan 100 µL sampel air laut ke media agar thiosulfate-citrate-bile-salt sucrose (TCBS), diinkubasikan pada suhu 30°C selama 18–24 jam, setelah itu dilakukan penghitungan koloni Green Vibrio Colony (GVC), Total Vibrio Count (TVC), dan persentase kejadian bakteri berpendar pada sampel air laut. Hasil pengamatan menunjukkan bahwa angka GVC dan TVC tertinggi terjadi di perairan Takalar pada bulan Mei 2020, sedangkan persentase sampel air laut dengan bakteri yang berpendar paling tinggi terjadi pada sampel dari perairan Pantai Cermin-Serdang Bedagai. Nilai GVC berkisar antara 0,01–130,00 × 102 CFU/mL dengan angka tertinggi terjadi pada bulan Januari, Oktober, November, dan Desember 2020, sedangkan nilai TVC berkisar antara 0.20–520.00 × 102 CFU/mL sepanjang tahun 2020. Tidak ditemukan adanya bakteri berpendar pada sampel air laut dari daerah Anyer dan Pangandaran. Para operator panti benih udang vanamei perlu senantiasa waspada terhadap dinamika populasi bakteri Vibrio spp. pada air laut yang akan dipakai dalam kegiatan pembenihan udang vanamei.

Abstract

This study was aimed to observe the survival rate (SR) and clinical signs, and the acute immune response of Pacific white shrimp (Litopenaeus vannamei), including total hemocyte count (THC), bacterial clearance, and prophenoloxidase (ProPO) activity due to infection with the Vibrio parahaemolyticus bacteria and its metabolites. Shrimp were infected by bacterial isolates with culture media (A), bacterial isolates without culture media (B), and metabolites suspensions from bacterial culture media (C) for 8 hours with observation every 2 hours. The results showed that experimental shrimps in groups A and B showed clinical signs and reduced SR, 24.44% and 62.22%, respectively. Experimental group C still showed the clinical signs but did not cause the mortality (SR 100.00%). The THC did not show significant changes, either based on experimental groups or duration of infection, but all groups tended to show decreasing THC at 8 hours post-infection. Bacterial clearance and ProPO activity also did not show significant differences between experimental groups or duration of infection at 8 hours post-infection. However, there was an increase in bacterial clearance activity at 6 hours post-infection in the experimental groups A and B. Based on the study results, the acute immune response in shrimp from all of the experimental groups did not show significant changes at 8 hours post-infection. However, group A significantly decreased SR compared to groups B and C.

Abstract

This study was conducted to explore the potential of adjuvant for the production of immunoglobulin Y (IgY) as antistreptococcosis in layer chicken with mass production orientation. Enterococcus faecalis which causes streptococcosis in the red tilapia was selected as a candidateantigen. The production of immunoglobulin Y (IgY) was carried out on Isa Brown layer chickens and aged around 20 weeks. Furthermore, thechickens were grouped into four groups (A, B, C, and D groups), each consisting of three chickens based on the type of adjuvant, while twochickens were used as a control group. Each group was treated by giving MONTANIDE ISA 71R VG adjuvant (A), Freund's adjuvant (B), aluminum potassium sulphate adjuvant (KAl(SO4)212H2O) concentration of 50 ppm in pH 7 (C), and only antigens without adjuvant (D). Chickens were kept for 35 days and each week was checked for presence the IgY antigen in the serum and egg yolk. Booster was conducted on 14th and 28th days of maintenance. The results showed that IgY in treatment group A was detected on day 28 in the serum and day 35 in the yolk. Whereas the treatment group B could be detected on day 35 in the serum. However, the IgY was not detected in the serum and yolk in C, D, and control groups until the end of the maintenance. Based on the results, it can be concluded that the appearance of IgY in serum and yolk in a relatively fast time is obtained in the combination of Enterococcus faecalis antigen with the emulsion of water-in-oil adjuvant (SEPPICMONTANIDE ISA 71R VG) compared to the other types of adjuvant that use in this study.
