Abstract

Protein 16 (p16) merupakan salah satu protein supresor tumor yang memiliki peran dalam patogenesis Karsinoma Sel Skuamosa (KSS) kepala dan leher. Peran protein ini berbeda sesuai dengan faktor risiko paparan karsinogen dan memiliki implikasi terhadap faktor prognostik patologik. Tujuan: Menentukan hubungan protein 16 dengan derajat histopatologik dan Invasi Perineural (IPN) pada KSS kepala dan leher. Metode: Penelitian ini merupakan cross-sectional study dengan sampel sebanyak 60 kasus KSS kepala dan leher yang telah didiagnosis di Laboratorium Patologi Anatomik RSUP Dr. M. Djamil Padang periode Januari 2016 sampai Desember 2018. Sampel diperoleh secara simple random sampling dari blok parafin yang berasal dari jaringan tumor. Dilakukan penilaian ulang pada slaid histopatologik untuk menilai derajat histopatologik dan invasi perineural. Ekspresi p16 pada sel tumor dilihat dengan pulasan imunohistokimia. Korelasi dilakukan analisis statistik bivariat menggunakan uji Chi-square dengan tingkat kemaknaan p<0,05. Hasil: Ekspresi p16 positif didapatkan pada 24 kasus (40%) dengan kasus terbanyak pada KSS kepala dan leher derajat tinggi, sedangkan pada 36 kasus (60%) tidak terdapat ekspresi p16. Ekspresi p16 positif memiliki hubungan yang bermakna dengan derajat histopatologik (p = 0,002) namun tidak terdapat hubungan antara ekspresi p16 dengan IPN (p = 1,000). Simpulan: Ekspresi p16 positif berhubungan dengan derajat histopatologik tumor yang lebih tinggi namun p16 tidak tidak berhubungan dengan kejadian invasi perineural pada KSS kepala dan leher.Kata kunci: derajat histopatologik, ekspresi p16, invasi perineural, KSS kepala dan leher

Abstract

Background: Head and neck squamous cell carcinoma (HNSCC) is a malignancy that occur in head and neck region and originates from squamous epithelial cell in the upper respiratory tract mucosa. This malignancy has a high heteregenicity. Carcinogenesis due to HPV infection has a different moleculer pathway from HNSCC without HPV infection. Methods: We searched Pubmed collection for open access and English language articles publish access from years 2016-2022 with keyword HPV, HNSCC and carcinogenesis. Results: We had 16 articles that matched from the keywords. The mechanism of HPV leading carcinogenesis in HNSCC because the HPV has a specific protein that can cause malignant transformation in squamous epithelial cells. The E6 protein inactivates the p53 tumor suppressor gene by activating the ubiquitin ligase E6AP causing degradation of p53. The E7 protein inactivates the pRb (protein retinoblastoma) tumpr suppressor protein by blocking the interaction between pRb and E2F, causing E2F depression. Conclusion: Carcinogenesis and malignant transformation in HNSCC correlated with HPV infection (high risk type) via the E6 and E7 proteins. This combination can causes abnormal cell cycle and leads cells to escape cell cycle control and get a malignant transformation.

Abstract

Background : Regenerative medicine stands at the forefront of transformative healthcare, offering innovative approaches to address tissue damage and degenerative conditions. Central to this paradigm is the profound impact of stem cell therapy, a dynamic and promising avenue within regenerative medicine. As we navigate the complexities of this evolving field, our systematic literature review seeks to unravel the diverse applications, breakthroughs, and challenges inherent in utilizing stem cells for regenerative purposes Methods : We searched in Science direct, Pubmed, NIH and another e-library with keyword stem cell, application and regenerative medicine. Our review aims to contribute to the ongoing narrative of stem cell therapy in regenerative medicine. Through this exploration, we aspire not only to enhance our understanding but also to guide future research endeavors and clinical applications in this dynamic and promising field. Result : We reviewed the article  with the theme article is stem cell and regenerative medicine. Stem cell therapy consistently demonstrated remarkable efficacy in promoting tissue regeneration. Across applications, stem cells showcased a remarkable versatility in enhancing tissue-specific cell behavior and activating regenerative pathways. his versatility positions stem cells as a promising tool with broad applications in regenerative medicine. Conclusion : This review emphasize the versatility and efficacy of stem cells in diverse regenerative scenarios. Evaluating methodological quality and comparing studies reveal nuanced insights, guiding future research directions. This review provides a comprehensive understanding of the current landscape, paving the way for innovative approaches in regenerative medicine.
