Abstract

The objective of this study was to assess the association of KiSS1 gene polymorphism with reproductive traits in Kaligesing goat. Genotypes of 48 ewes aged three years old were determined using PCR-RFLP and DNA sequencing. Sixteen out of 48 samples were monitored for the estrus cycle and determined their 17β-estradiol and progesterone levels using ELISA method. The results showed that KISS1 gene in the studied population was polymorphic with one single nucleotide polymorphism (SNP T125A). Then SNP was used to analyze genotype all individuals by PCR-RFLP method using MboII enzyme. Three genotypes (TT, TA and AA) were identified. Genotype frequencies of TA were 59.57%, AA were 40.43% while TT was not inculed in calculation for genetic diversity and assosication analysis since the number of genotype only one goat. The allele frequencies of T and A were 29.79% and 70.21%, respectively. The genotype distribution for the SNP was deviated from Hardy-Weinberg equilibrium (χ2=8.10; P<0.025). Hormonal analysis showed that the levels of 17β-estradiol in the follicular and luteal phase were 50.12±61.26 and 42.35±42.43 pg/ml, respectively, while the levels of progesterone hormone in the follicular and luteal phase were 7.87±10.59 and 12.39±15.93 ng/ml. No significant associations of the polymorphism were observed for any hormonal levels. However, it was first report about polymorfism in KiSS1 gene of local Indonesian goat especially Kaligesing goat.

Abstract

Livestock genetic improvement is the main purpose of animal breeding program. Biotechnology brings novel method to improve the animal breeding program qualitatively and quantitatively. Identification of gene sequence of polymorphism are used widely to analyze gene sequence to understand genetic variation. Calpain 1 (CAPN1) gene is the gene that involve in the process of improving meat quality such as marbling and tenderness of the livestock including the cattle, buffalo, and goat species. CAPN1 is responsible in encoding the µ-calpain that regulate postmortem proteolysis.  This study collect the gene database including calpain 1 [Bos taurus (cattle)] NC_037356.1, calpain 1 [Bos indicus (zebu cattle)] NC_032678.1, calpain 1 [Bos indicus x Bos taurus (hybrid cattle)] NC_040104.1, calpain 1 [Bubalus bubalis (water buffalo)] NC_059161.1 calpain 1 [Capra hircus (goat)] NC_030836.1 through NCBI. Those collected gene were analyze using the Clustal W alignment, MEGA 11, and BLAST program. The results indicate that the cattle, buffalo, and goat species CAPN1 gene of the 5’ UTR, Intron, Exon, dan 3’ UTR region show 9 SNPs, 1691 SNPs, 153 SNPs, and 197 SNPs, respectively. Total of the identified SNPs within the the cattle, buffalo, and goat species CAPN1 gene comparison are 2050. This study can be used for the initial report in order to develop further research related to CAPN1 gene for the livestock ruminant.

Abstract

The aim of this research is to study the phenotypic character and growth of layered chickens. In this study, crosses between G4 Layer chickens were carried out which resulted in G5 Layer chickens. Morphological characters, growth weights and measuring the feed-conversion ratio (FCR) were observed. The results obtained were 10 individuals of G5 Layer chickens with the proportion of male gametes (70%) and female gametes (30%). Phenotypic characters are brown fur color and a combination of brown and white. The value of Feed Conversation Ratio (FCR) is 1.67 and the average weight growth of G5 Layer chickens reaches 491.2 grams at the age of 7 weeks.

Abstract

Itik Magelang merupakan salah satu jenis unggas dwiguna yang mampu menghasilkan daging dan telur. Optimalisasi produktivitas itik dapat dilakukan dengan memberikan imbuhan pakan (feed aditif) berupa prebiotik. Penelitian ini bertujuan untuk mengkaji pengaruh penambahan inulin dalam pakan terhadap bobot karkas dan persentase organ dalam Itik Magelang jantan. Rancangan percobaan pada penelitian ini adalah Rancangan Acak Lengkap (RAL). Penelitian ini menggunakan itik Magelang jantan sebanyak 100 ekor. Penelitian ini terdiri dari 4 perlakuan dengan 5 ulangan. Perlakuannya adalah: P0 (pakan control) P1(pakan + Inulin 0,5%) P2 (pakan + Inulin 1%) P3 (Pakan + Inulin 1,5%) dengan jumlah itik ulangan adalah 5 ekor. Variabel yang diamati meliputi: bobot karkas, dan persentase organ dalam (hati, jantung, ampela, dan Limpa). Analisis data menggunakan analisis ragam (ANOVA) dan data yang berbeda nyata diuji lanjut Duncan's Multiple Range (DMRT). Hasil penelitian ini menunjukkan bahwa penambahan inulin pada pakan tidak berpengaruh nyata terhadap bobot karkas dan persentase organ dalam. Bobot karkas (899,35 ± 92,39) persentase jantung (0,87 ± 0,52%) persentase hati (2,70 ± 0,35%) persentase ampela (4,36 ± 0,47%) persentase limpa (0,11 ± 0,05%) yang masih ke dalam kondisi normal. Berdasarkan hasil penelitian dapat disimpulkan bahwa penambahan prebiotik inulin sebanyak 0, 0,5, 1, dan 1,5% dalam pakan itik Magelang jantan tidak berpengaruh nyata terhadap bobot karkas dan persentase organ dalam (jantung, hati, ampela, dan limpa).
