Abstract

Broad range applications of human mesenchymal stem cells (MSCs) neccesitate the best way to preserve MSC. Cryopreservation is the only one applicable methods for MSCs storage, which consists of vitrification and slow freezing cryopreservation. However, there is a lack of standard procedure of cryopreservation. Different freezing steps and cryo-mediums are often used, which may impact on MSCs characteristics. This article aims to evaluate the impact of cryopreservation on consensus criteria of MSCs derived from various tissues, which consists of surface morphology, differentiation capacity, and surface marker expression. Twenty-nine in vitro studies were analyzed, and their results organized according to the cell attributes they asses. This review shows that there are various procedures of slow-freezing cryopreservation. Our findings show that a temperature reduction rate of -1oC / min is the most suitable for MSC cryopreservation, and combination of DMSO and FBS in addition of growth medium is still the most used cryomedium in slow freezing cryopreservation. Whereas, in vitrification, combination of ethylene glycol, Ficoll, and sucrose is used as cryoprotective agents. In conclusion, our results show that although there are variations in slow-freezing cryopreservation, there is no significant effect on MSC either morphology, surface marker expression (positive markers consists of CD105, CD90, CD73; and negative markers consists of CD45, CD34, CD14 or CD11b, CD19, and HLA-DR), or differentiation capability (adipogenic, chondrogenic, and osteogenic). Whereas, vitrification effects could not be clearly defined yet, because of a limited number of studies.Key Words: Cryopreservation, Slow-Freezing, Vitrification, MSC characteristiccs, MSC storage

Abstract

Objective: This study evaluates the critical factors affecting COVID-19 vaccine acceptance in Indonesia using a proposed extended theory of planned behaviour (extended-TPB). Design/Methods/Approach: We conducted an online survey (N = 222 respondents) using a valid and reliable questionnaire to evaluate people's intention to take the COVID-19 vaccination. A structural equation modeling was performed to demonstrate the impact of religious acceptance (RA), vaccine safety (SF), trust (TS), perceived behavioural control (PBC), attitude towards vaccination (ATT), and subjective norms (SN), on intention (IN) to accept COVID-19 vaccine. Findings: The results show positive relationships of all hypotheses except the correlation between RA and TS. TS becomes a positive determinant of ATT, SN, PBC, and INT, and TS is influenced by SF but not by RA. TS is the key determinant factor of intention to uptake the COVID-19 vaccine. Originality: This study used a proposed extended TPB by adding trust, religious acceptance, and safety as additional components of TPB to predict the intention to participate in COVID-19 vaccination. This study is essential because the vaccine is believed to be the best strategy to deal with virus-related diseases, such as COVID-19. However, there is a lack of information on the multifactor effects of Indonesian people's acceptance of vaccination, especially among Surakarta residents. Practical/Policy implication: This research can be a recommendation for the Indonesian government to elevate public intention to take the COVID-19 vaccine, especially by implementing policies that accentuate people's trust by confirming and publishing information about the vaccine safety.

Abstract

One of the food-product halalness indicator is the absence of alcohol. In this case, traders as business actors must know about the contamination of khamr and alcohol in food ingredients that are traded so as not to endanger themselves and consumersas a condition when applying for halal certification. Because the content of khamr and alcohol besides endangering health, the Qur'an also clearly forbids them. Therefore this community service aims to increase the knowledge of shop owners around IAIN Surakarta regarding the contamination of khamr and alcohol in food as an effort to prepare for halal certification. This activity is carried out using the socialization method, namely by giving lectures and questions and answers to 10 food stall owners. Activity evaluation is carried out by providing a pre-test and post-test on the topic presented. From this community service activity, there was an increase in knowledge which could be seen from the pre-test and post-test scores, which were respectively 36 and 86. Specifically, the increase in knowledge about the definition of alcohol and khamr was 50%, the rules of the beverage trade. alcohol in Indonesia is 50%, types of alcoholic drinks are 60%, and the law of alcohol and khamr in Islam is 30%.

Abstract

Batik wastewater contains various chemical compounds, such as dyes, synthetic compounds, heavy metals, and organic compounds that can harm the environment. Therefore, proper waste management is required. The use of alum as coagulant in textile wastewater treatment is commonly used, but it is not effective enough to remove wastewater colour and reduce COD. This study aims to modify alum-based batik wastewater treatment using (i)double coagulant by combining alum - FeSO4; and (ii)coagulant-oxidizing agent with combination of alum – chlorine. This experimental study consisted of two stages, the first experiment aimed to determine the optimum composition dose of alum – FeSO4 and alum – chlorine in depositing waste pollutants, reducing the colour and pH of the effluent; the second experiment aims to determine the effect of alum - FeSO4 and alum - chlorine at the optimum dose in reducing the COD. The results showed that alum–chlorine in various compositions reduced colour ten times and precipitated pollutants two times better than alum-FeSO4. The optimum concentration ratio of alum:chlorine is 1:1 (1.2 g/L alum : 1.2 g/L chlorine). This dose reduced the pH to 8; depositing 0.385 g pollutants; removed 99.57% wastewater colour; and decreased COD from 686.9 mg/L to 235.53 mg/L

Abstract

Breast cancer is the most common cancer among women and a major cause of cancer-related death worldwide, including Indonesia. Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer, characterized by high mortality and resistance to standard treatments. Cancer stem cells (CSCs) is known to be a key factor of drug resistance in TNBC, mediated by the signal tranducer and activator of transcription factor 3 (STAT3). This study aims to explore the bioactive compounds from Indonesian’s natural product which potentially target CSCs by inhibiting STAT3. We used in silico methods, consist of literature review, followed by selection of promising compounds, ADMET analysis and selection using Lipinski’s rules, and molecular docking simulation. Our results found curcumin, quercetin, α-mangostin, gingerol, and rocalglamide as the bioactive candidates which target CSCs. ADMET testing showed that only curcumin, quercetin, and gingerol meet the Lipinski’s criteria. The molecular docking results showed that quercetin exhibited the strongest binding affinity (-8.3 kcal/mol) compared to curcumin (-6.9 kcal/mol) and gingerol (-5.9 kcal/mol). The results indicate that quercetin binds effectively at the active site of STAT3. The findings suggest that quercetin has potential as a STAT3 inhibitor. Further research is needed to determine the significant efficacy and potential as an anticancer.
