Abstract

Background: In the last few decades there has been an increase in cases of chronic diseases and premature aging which are some of the key factors in influencing public health. This condition is caused by various causes, but the main factor is the number of free radicals that exceed the body's capacity to neutralize them. Antioxidants have an important role as health protective factors because they can neutralize and protect cells from the presence of free radicals. Red castor leaves have been reported to have good activity as an antioxidant. The chemical content of this plant is anthraquinone, flavonoid, saponin, tannin and terpenoid. Objective: This study aimed to evaluate the antioxidant activity of red jatropha leaf extract using the DPPH method. Methods Samples were extracted using 70% ethanol by maceration method. The antioxidant activity test was carried out using the DPPH method (1,1-diphenyl-2-picrylhydrazyl) using a comparison of Vitamin C. Each test used a variation of the concentration of the solution. For red jatropha leaf extract using concentration variations of 20, 40, 60, 80 and 100 g/ml, while for vitamin C using concentration variations of 10, 15, 20, 25, and 30 g/ml. The absorbance of each test solution was measured using a spectrophotometer at a wavelength of 515 nm. The results are reported as IC50 values. Results: The results showed that the IC50 values of red jatropha leaf extract and vitamin C were 47.81 and 14.99 g/ml, respectively. This value indicates that the Chinese jatropha leaf extract has a very strong antioxidant activity (IC50 <50 g/ml). Conclusion: Red jatropha leaf extract showed very strong antioxidant activity with an IC50 value of 47.81 g/ml so it has great potential as an antioxidant that can be used for the development of traditional medicinal raw materials.

Abstract

Indonesia is an archipelagic country with many oceans, rivers and lakes. The number of drowning incidents is still unknown because many cases are not reported and many victims do not receive medical services. Based on data from the National Search and Rescue Agency (BASARNAS) of Southeast Sulawesi in 2015 from various locations, 12 people died from drowning, this number is relatively small compared to 2016, the number of drowning victims increased by 17 people and in the last year of 2017 until the end of February, 2 drowning victims were reported. The latest BASARNAS data in 2023 showed 43 cases of ship accidents and 24 cases of dangerous conditions for humans. Providing first aid education requires real action for the general public so that they can provide assistance to drowning victims. The implementation of this community service began with a pre-test followed by counseling on drowning and practicing how to perform CPR in drowning cases, ending with a post-test by the Sawa Village community of 40 people. The pre-test results showed that the level of public knowledge before being given counseling was mostly sufficient with a total of 22 people (55%), less knowledge 17 people (42%) and good knowledge 1 person (3%). After being given counseling, there was an increase in sufficient knowledge of 23 people (57%), good knowledge of 15 people (38%) and less knowledge decreased to 2 people (5%). Health Counseling on First Aid for Cardiopulmonary Resuscitation in Drowning Cases has succeeded in increasing public knowledge in Sawa Village, North Konawe Regency.

Abstract

Background: The high prevalence of cancer has a direct impact on the quality of life and nutritional status of sufferers, mainly due to decreased appetite, metabolic disorders, and side effects of conventional therapies such as surgery, mastectomy, chemotherapy, hormonal therapy, or radiation therapy. These therapies, although aimed at inhibiting the growth and metastasis of cancer cells, often cause severe side effects that worsen the patient's condition. Therefore, it is important to explore alternative treatments that are effective and have minimal side effects. In this regard, biological sources, such as endophytic fungi kelakai, contain various active secondary metabolites that can inhibit cancer cell proliferation so that they can be part of a more public health-friendly supportive therapy approach. Objective: This study aims to prove the cytotoxic activity of the extract of endophytic fungi in kelakai against MCF-7 and Vero cells. Methods: Isolation, purification, and extraction process of secondary metabolites from the endophytic fungi isolates were carried out in the same manner as our previous research. The ethyl acetate extract of kelakai was obtained by extractive fermentation from pure isolates of the endophytic fungi in kelakai. Extraction was carried out by the fractionation method using the ethyl acetate solvent at a ratio of 1:1. Identification of chemical contents, including tannins, alkaloids, flavonoids, saponins, and steroids/terpenoids, was conducted qualitatively. Cytotoxic activity was tested by the MTT method. Results: From the isolate extraction, 2.4 g of ethyl acetate extract was produced. The identification of chemical contents showed positive results, meaning that the extract did contain tannins, flavonoids, alkaloids, steroids, and saponins. The extract cytotoxicity test against MCF-7 and Vero cells yielded IC50 values of 517.3015 μg/ml and 1,074.9152 μg/ml, respectively. Conclusion: Based on these results, the extract of kelakai was not toxic to Vero cells, but it was toxic to MCF-7 cells. These findings suggest that the kelakai can be the basis for exploring local plants as a source of natural medicine while supporting the conservation of endemic plants to provide safer alternative therapies and improve public health.
