Abstract

The use of medicinal plants is increasing due to the search for alternative resources to treat diseases such as hypertensions and infection. Along with the development in science, preventive action should take place to prevent our body from suffering from these diseases. This can be done by increasing the human immune status with immunomodulatory agents. Binahong empirically have benefits for wound healing. The purpose of this research was to investigate the immunomodulatory effect of ethanolic extract of binahong leaves. The non-specific modulatory effects of the ethanolic extracts of binahong leaves on the immune systems were measured based on phagocytosis index and phagocytosis capacity. Tests were conducted on strain Balb/C male mice at the age of 6-8 weeks. Mice were administered orally with the extract of binahong leaves (doses of 25, 50, 75 mg/kgBW) for 14 days. The test results with the index parameters and macrophages phagocytosis capacity at doses of 50 and 75 mg/kgBW did not significantly increased when compared with the controls. From these results, we concluded that the ethanolic extract of binahong leaves with a dose of 25, 50 and 75 mg/kgBW cannot significantly the activity of macrophages by phagocytosis index parameters and phagocytosis capacity significantly.

Abstract

Cinachyrella sp. marine sponge produce many kinds of secondary metabolites. The purpose of this study was to examine the cytotoxic and anti-proliferative activity of marine sponge Cinachyrella sp. The sponge was extracted with 96 % ethanol. Ethanol extract cytotoxicity assay were performed with MTT method (Microculture Tetrazolium) against to cell lines of HeLa, T47D, WiDr and Vero. The results show that of the ethanol extract were only toxic to cell lines of HeLa IC50 897.809 μg/mL but did not toxic to cell lines of T47D, WiDr and normal cell lines of Vero. Fractionation of the ethanol extract was conducted by a Vacuum Column Chromatography (VCC) and have 4 fraction that were F1, F2, F3 and F4. Cytotoxicity and cell proliferation inhibitory were tested of fraction F1, F2, F3 and F4 against cell lines of T47D. The values IC50 of F1; F2; F3 and F4 against cell lines of T47D were 82.744; 163.679; 66.522 and 333.026 μg/mL and fraction F3 concentration 31.5 µg/mL inhibits cell proliferation cell lines of T47D at 48 hours of incubation

Abstract

Plants can be sources of immunomodulatory agents. Piper crocatum Ruiz & Pav. (red betel) leaves have been used to heal many diseases, especially to increase endurance. The aim of this research was to investigate the imunomodulatory effect of methanolic leaf extracts of Piper crocatum Ruiz & Pav. and its fractions by in vitro phagocytic macrophage activity test. The result showed that methanolic extract of Piper crocatum Ruiz & Pav. 75μg/mL could stimulate phagocytic macrophage activity equal to 100μg/mL product-X® that contained Echinacea extract. Separation of the extract by vacuum liquid chromatography using gradient solvent n-hexane - ethyl acetate yielded 5 fractions. The second fraction showed the highest activity. It contained alkaloid and terpenoid, while the extract also contained flavonoid and essential oil. Alkaloid and/or terpenoid could be the compound in the leaves of Piper crocatum Ruiz & Pav. that responsible increasing phagocytic macrophage activity.

Abstract

Munculnya penyakit-penyakit infeksi serta adanya sifat resistensi mikrob terhadap antibiotik yang telah ada, telah mendorong penelitian untuk menemukan antibiotik baru dari bakteri, fungi dan Actinomycetes. Saat ini, kelas Actinomycetes menjadi fokus para peneliti karena salah satu anggotanya, yaitu Streptomyces dipromosikan sebagai bakteri yang mampu menghasilkan zat antimikrob terbesar. Sebanyak 45,6% dari 8.000 antimikrob dihasilkan oleh Streptomyces. Tujuan penelitian ini untuk mengisolasi dan mengkarakterisasi Streptomyces yang berasosiasi dengan rizosfer rumput belulang (Eleusine indica) yang berpotensi sebagai penghasil zat antifungi. Jenis penelitian ini adalah eksplorasi dengan pemeriksaan laboratorium. Berdasarkan hasil penelitian ditemukan sebanyak 33 isolat dari rizosfer rumput belulang dan 17 isolat di antaranya berpotensi sebagai penghasil zat antifungi. Delapan isolat hanya mampu menghambat pertumbuhan salah satu fungi uji, 7 isolat mampu menghambat 2 macam fungi uji dan satu isolat, yaitu RRR67 mampu menghambat pertumbuhan ketiga fungi uji, yaitu Trichophyton mentagrophytes, Aspergilus fumigatus dan Candida albicans. Berdasarkan hasil pemeriksaan dengan mikroskop elektron (SEM) diketahui morfologi spora dan ornamen permukaan rantai spora isolat RRR67 termasuk anggota genus Streptomyces. Penelitian ini menyimpulkan bahwa 17 isolat dari rizosfer rumput belulang (Eleusine indica) berpotensi menghasilkan zat antifungi yang diperlukan untuk pengobatan penyakit-penyakit infeksi.

Abstract

Cubebin is a lignan isolated from Indonesian plant Piper cubeba L.f fruits. Cubebin has been found to strongly inhibit contraction of isolated-trachea of guinea pig induced by histamine or metacholine. ln the present study, the effcct of cubebin was investigated on histamine release from rat peritoneal mast cells (RPMCS), a connective tissue mast cell. Compound 48/80 (G Protein activator), thapsigargin (SERCA inhibitor), ionoinycin (calcium ionophore), and PMA (PKC activator) were used as inducers for histamine release from connective tissue mast cell. Histamine released in the medium was measured by HPLC-Huorometry. The results showed that cubebin at the concentration of 30 and 100 µM inhibited the histamine release from RPMCS induced by thapsigargin by 12,86 ± 1,84 % and 40.38 ± 1.93 %, respectively. In addition, there was a partial inhibition seen in respond to ionornycin, with no effects towards compound 48/80 or PMA, These data indicate that cubebin inhibited thc histamine release from connective mast cells, and might involve in the activation of sarco/ endoplasmic reticulum Ca2+ ATPase.

Abstract

Coleus amboinicus (Lour.) merupakan tanaman obat tradisional yang banyak digunakan oleh masyarakat dan telah diteliti secara ekstensif serta dilaporkan di beberapa bidang ilmu pengetahuan. Penulisan review artikel ini didasarkan pada studi literatur tahun 1993 hingga tahun 2021, yang diperoleh dari database google scholar, Scopus, Science Direct, PubMed, dan lainnya. Tanaman ini merupakan tanaman semak aromatis, berbunga, namun jarang berbiji dan dapat diperbanyak secara vegetatif. Profil senyawa fitokimianya dapat digolongkan menjadi beberapa kelas yang berbeda seperti terpenoid, fenolik, flavonoid, alkaloid, saponin, steroid dan tanin. Tanaman C. amboinicus (Lour.) kaya akan komponen fenolik seperti asam rosmarinat dan asam kafeat, dengan mayoritas komponen volatil utamanya yaitu karvakrol. Profil fitokimia C. amboinicus (Lour.) dapat bervariasi tergantung pada berbagai faktor, seperti lingkungan, keanekaragaman genetik, bagian tanaman yang digunakan, maupun metode dan proses ekstraksi. Potensi farmakologinya seperti antibakteri, antifungi, antioksidan, antikanker, antiinflamasi, antidiabetes, antihiperlipidemia, antimalaria serta laktagoga telah banyak dilaporkan. Selain itu, tanaman ini juga dimanfaatkan dalam kuliner serta sintesis biogenik senyawa nanopartikel. Eksplorasi dari tanaman C. amboinicus (Lour.) juga merambah kepada mikroorganisme endofitnya, dan dikaitkan dengan kemampuannya untuk menghasilkan senyawa bioaktif yang potensial sebagai antibakteri, agen biokontrol serta produksi enzim selulase. Review artikel ini memberikan informasi terkait botani, fitokimia, farmakologi, sintesis biogenik, pemanfaatan lainnya serta eksplorasi mikroorganisme endofitnya. Artikel ini diharapkan dapat menjadi sumber informasi untuk pengembangan potensi C. amboinicus (Lour.) yang semakin meluas.
