Abstract

Penelitian ini dilaksanakan untuk melakukan evaluasi terhadap nilai pemuliaan (NP) pada calon pejantan sapi Aceh sebagai pejantan unggul berdasarkan performans keturunan selama tahun 2010 sampai 2013. Sapi Aceh yang diikutsertakan dalam penelitian ini, masing-masing dengan nomor A001, A004, P0751 dan P0752. Analisis data dilakukan berdasarkan performans berat sapih terkoreksi 205 hari (BB205), berat setahunan terkoreksi 365 hari (BB365) dan berat akhir atau dewasa terkoreksi 550 hari (BB550) hasil keturunan keempat calon pejantan tersebut. Analisis nilai heritabilitas (h²) BB205, BB365 dan BB550 secara berurutan adalah 0,48; 0,40 dan 0,49. Hasil penelitian menunjukkan bahwa calon pejantan nomorP0752 memiliki NP BB205, BB365 dan BB550 tertinggi masing-masing sebesar 54,24 kg; 83,99 kg dan119,08 kg. Kecermatan relatif terbaik dalam uji zuriat yaitu sebesar 1,55 pada sifat BB550 sehingga seleksi pada calon pejantan sapi Aceh dapat dilakukan berdasarkan rata-rata performans BB550 keturunannya.Kata kunci: Sapi Aceh, Uji zuriat, Heritabilitas, NP, Kecermatan relatif

Abstract

This research aimed to find out the genetic progress for Aceh cattle by several growth traits selection such as birth weight (BW), weaning weight (WW), yearling weight (YW), mature weight (MW) of does to identify the best selection response from bulls and cows breeding patterns. The recorded data of production and reproduction of Aceh cattle from 2010 to 2014 at breeding centre were analyzed and used as a technical coefficient on estimate heritability (h2), genetic correlation (rG), direct selection response (RY) and correlated selection response (CRY) on several breeding patterns. Most of  h2 and rG value among growth traits were positive and high. The highest value of Ry found on BW (0.04), WW (1.21), YW (2.05), MW (3.28) and this was obtained on breeding pattern for 3 years and cow for 6 years. The indirect selection or CRY value based on BW were lower than WW. It is concluded that WW might be used as selection criterion in order to increase WW, YW and MW.

Abstract

Penelitian ini bertujuan untuk membuat estimasi nilai pemuliaan (NP) dan most probable producing ability (MPPA) sifat produksi sapi Aceh di Kecamatan Indrapuri, Provinsi Aceh. Penelitian dilaksanakan mulai tanggal 7 Maret 2013 sampai dengan 7 April 2013 di Balai Pembibitan Ternak Unggul (BPTU)-Hijauan Pakan Ternak (HPT) Sapi Aceh Indrapuri. Materi penelitian ini terdiri dari catatan produksi dan populasi sapi Aceh tahun 2010 sampai 2012. Jumlah sapi yang digunakan untuk mengestimasi NP dan MPPA pada sifat produksi adalah 38 ekor. Estimasi nilai estimasi heritabilitas dengan metode korelasi saudara tiri sebapak (paternal half-shib correlation), sedangkan estimasi repitabilitas berdasarkan dua catatan individu menggunakan metode korelasi antar kelas (interclass correlation). Hasil penelitian menunjukkan bahwa nilai heritabilitas berat lahir, berat sapih, berat satu tahun, dan berat akhir masing-masing adalah 0,15+0,12, 0,48+0,58, 0,49+0,58, dan 0,56+0,69. Nilai repitabilitas yang dapat diestimasi berdasarkan data recording adalah berat lahir sebesar 0,33+0,22. Sapi Aceh pejantan nomor P.075 memiliki NPabsolut berat akhir tertinggi yaitu sebesar 14,99 kg. Nilai MPPAabsolut berat lahir tertinggi pada induk sapi Aceh nomor A.0668 yaitu sebesar 15,38 kg.(Kata kunci: Sapi Aceh, NP, MPPA, Heritabilitas, Repitabilitas, Sifat produksi)

Palembang: Penerbit & Percetakan Universitas Sriwijaya (UNSRI).This study aimed to determine the diversity of phenotypes and genotype distribution of crossbred chickens based on qualitative traits. The crossbred chickens used are crossbred Merawang chickens with other chickens. Merawang chickens are dual-purpose chickens that have great potential to be developed. The main problem with Merawang chickens is that the characteristics of Merawang chickens have not been registered. Efforts made to overcome this problem were by analyzing the genotype distribution of the qualitative traits of Merawang Cross chickens and PCA analysis of the chicken morphostructure. One way to analyze the genotype distribution can be done by calculating the genotype frequency, heterozygosity, and genetic ingression values. This study was conducted using a sample of 26 male and 44 female Merawang Cross chickens. From the entire population of Merawang Cross chickens, the genotype frequency values showed that the dominant phenotype in Merawang Cross chickens was recessive in feather color, feather features, feather shimmer, and comb shape. But it is dominant in shank color which has an allele frequency value of 100% white/yellow. The heterozygosity value shows a medium-value feather color pattern and other phenotypes are low (<30). So, the genotype distribution pattern in Merawang Cross chickens is more dominantly carried by European chickens, the high genetic integration value is found in Rhode Island Red (RIR) chickens reaching 100% in males and 87% in females. This shows that crossbred chickens or Merawang Cross chickens have dominant genes from Rhode Island Red (RIR) chickens.

Abstract

Objective: This study was carried out to characterize Pasundan (Rancah) cattle from West Java of Indonesia based on a partial Zinc Finger Y-linked (ZFY) gene sequence.Methods: Total of fourteen (14) DNA samples of unrelated Pasundan bulls at the breeding station (BPPIBT-SP Ciamis, West Java) were used for sequencing analysis. The blood samples were collected from jugular vein of each bull for the DNA extraction analysis. Along 979 bp of ZFY gene was amplified using primer pairs of forward: 5’- GGT GAG GGC ACA TGA GTT C -3’and reverse:  5’- CTC TGC AGG TGG TTG TGT AA -3’. Therefore, a forward sequencing method was performed in the present study to obtain the ZFY gene sequences. Along 714 bp of bovine ZFY gene sequences were used for determining the Neighbor-Joining (NJ) tree for Pasundan bulls with 1000 × bootstrap replications using a MEGA computer program. The genetic diversity parameters of haplotype diversity (Hd), nucleotide diversity (pi), Tajima’s D test and Fu’s Fs statistics were calculated using a DNAsp computer program.Results: Total of fourteen (14) haplotypes of ZFY gene and seventy (70) mutation sites were detected in the Pasundan bulls. The haplotype diversity (Hd) and nucleotide diversity (pi) in Pasundan’s ZFY gene diversity were 1.00 (high) and 0.01 (low), respectively. The neutrality test of Tajima’s D and Fu’s Fs statistic values were -1.42 and -6.47, respectively. Bovine ZFY gene can be classified into two clades of ZFY-1 and ZFY-2. Therefore, the Neighbor-joining (NJ) tree (1000×bootstrap) revealed that the Pasundan cattle are classified into clade ZFY-1 together with Bos taurus (AF241273, AF465179). Meanwhile, a clade ZFY-2 consisted of Bos taurus (AF465181, DQ336536), Bos indicus (DQ336537), and Bos javanicus (DQ336578).Conclusion: The ZFY gene of Pasundan bulls were polymorphic with seventy mutation sites. Therefore, Pasundan bulls were characterized into ZFY-1 clade or Taurine lineage based on ZFY gene sequence variaton. The crossbreeding program with Bos taurus straw to increase meat production of cattle may affecting the genetic structure of Pasundan bulls since this program is widely applied in West Java through many years ago.
