Abstract

Relation of successful ICSI (intracytoplasmic sperm injection) with DNA stability and thiol-disulfide status of rat spermatozoa nuclei was investigated. Testicular, caput and cauda epididymal spermatozoa have been used for this research. The nuclei of these spermatozoa were stained with aeridine orange for DNA stability test and labeled with mBBr for thiol-disulfide status before injesting into oocytes. Cauda epididymal spermatozoa were very stable and contained high disulfide than caput epididymal and testicular spermatozoa. Results from spermatozoa injection into oocytes show that they were capable to fertilize oocytes. However, testicular spermatozoa can not develop up to blastocyst stage. These results indicated that successful ICSI at least in the rat depends on DNA stability and disulfide status of sperm nuclei.   Key words: ICSI, DNA stability, disulfide status, acridine orange, mBBr

Abstract

Rat cauda epididymides were kept in 1.5-ml centrifuge tubes containing 1 ml milli-Q water or physiological saline (0.9% NaCl) and stored and freezed at -196ºC without cryoprotectant for up to 7 days. After thawing of the cauda epididymis, all spermatozoa were non-motile immediately after collection. All oocytes injected with sperm heads (nuclei) of spermatozoa collected from frozen-thawed cauda epididymis in saline were activated (100%) and gradually decreased (P<0.05) in cauda epididymis frozen in milli-Q water at -196°C (86%), and in control (69%). In activated oocytes, a large proportion of sperm heads had transformed into male pronuclear formation (66-78%). When 1-cell embryos were cultured for 120 h, 7% developed to blastocyst stages. These results indicate that genetic materials of species (at least in the rat) that had unexpectedly die can be saved by a simple method. Key words: ICSI, Sperm Heads, Piezo-Injection, Frozen, Rat

Abstract

The objective of this study was to determine the strategic program for buffalo development in South Sulawesi. The study was a thought of idea and concepts to create programs and strategic activities for buffalo development by using SWOT analysis that preceded the determination of the internal and external factors. Based on the results of SWOT analysis, the strength map for buffalo development in South Sulawesi was in quadrant I (aggressive), this indicated that by utilizing the opportunities and minimizing the threats, buffalo development in South Sulawesi was able to improve faster and making center of development buffalo in Indonesia. Operational policy strategy in determining the program of activities, were: (1) take advantage of the ability of buffalo convert low-quality feed to fulfill the demand for meat;(2) to intensify AI in order to improve the birth that the demand for meat can be fulfilled, (3) utilizing buffalo ability to convert low-quality feed to reduce importation of meat, and (4) to intensify AI in order to improve the birth so that meat imports could be reduced.

Abstract

Beberapa teknik fertilisasi mikro telah diterapkan untuk mengatasi masalah infertilitas pria, namun hanya intracytoplasmic sperm injection (ICSI) yang mampu meningkatkan secara nyata keberhasilan fertilisasi, implantasi dan kelahiran. Pada perkembangannya teknik ICSI juga telah diterapkan pada beberapa jenis hewan sebagai suatu model untuk mempelajari kemampuan fertilisasi berbagai jenis spermatozoa yang secara alami atau melalui fertilisasi in vitro tidal( bisa membuahi sel telur. Walaupun keberhasilan teknik ICSI telah dapat mengatasi masalah infertilitas pada pria, teknik ICSI masih mempunyai potensi negatif yang mungkin muncul pada anak hasil ICSI. Namun demikian, hal ini perlu penelitian lebih lanjut untuk membuktikannya.

Abstract

ABSTRACTDiprosopus Monauchenos (DM) is one of congenital defect that can be caused by a genetical factor. This study was conducted to reveal the genetic mutation of DM cattle. Research showed that along 354 bp of partial BTA13 sequence (69,985,393th - 69,985,745th nucleotide) in DM and normal samples were obtained by sequencing analysis. Research showed that one insertion/deletion (indel) mutation at 28th and two transition mutations at 241th and 269th positions were detected in both samples. The obtained sequence in the present study was located at the intronic region of LPIN3 gene (69,972,281th - 69,991,534th nucleotide). In the future, a depth research to investigate the genetic mutation in DM animal with whole genome analysis is important to get the results accurately.Keywords: cattle, DM, mutations, sequencing, BTA13

Abstract

This study aims to determine the effect of cattle origin and ownership status on the reproductive performance of beef cows in four locations of the Sekolah Peternakan Rakyat (SPR). The study locations include SPR Sungai Lilin (Musi Banyuasin Regency), Mesuji Raya (Ogan Komering Ilir Regency), Pelepat Ilir (Bungo Regency) and Wanaraya (Barito Kuala Regency). The study was conducted through a survey method with direct observation and interviews with SPR members. The variables observed included age at first calving, calving interval, empty period, and service per conception (S/C). The factors analyzed were cattle origin (within or outside the SPR) and ownership status (personal or profit-sharing). The results showed that cattle origin had no significant effect on AFC, CI, DO and S/C. Ownership status had a significant effect on AFC, CI and DO. Recommendation of reproductive optimization strategies based on livestock origin include strengthening breeding stock, standardization of the quality of parents, both from within and outside the SPR area and certification of cattle origin, In contrast, reproductive optimization strategies based on ownership status include reformulation of the profit-sharing model, incentive programs for profit-sharing farmers, development of a gradual ownership scheme.
