Abstract

The use of insecticides in the intensive control of Helicoverpa armigera caterpillars increases the environmental deterioration, the natural enemies, and threatening the user's health. The study aims to isolate, characterize, and determine the biological potential of local isolates of entomopathogen bacteria as the biological controlling agent on H. armigera larvae. Bacterial isolation was performed using a serial dilution followed by bacterial macroscopic and microscopic characterizations. A total of eight entomopathogen bacterial isolates were successfully obtained from this study, namely IBE 01, IBE 02, IBE 03, IBE 04, IBE 05, IBE 06, IBE 07, and IBE 08 with diverse morphological characters. The dead larvae generally showed soft, blackish/brownish, smelly, and watery characteristics. Characteristics due to infection with entomopathogen IBE 04 bacteria show the body of larvae that are soft, reddish, and smelly. The treatment of IBE 07 isolates results in the body of the dead larvae being easily ruptured, reddish, and smelly. The percentage of IBE on the mortality of H. armigera larvae is > 50%. The isolates with the highest percentage of killing the larvae were IBE 04 and IBE 07, at 94% and 88%, respectively. Keywords: biocontrol, larvicide, entomopathogenic bacteria, Helicoverpa armigera

Abstract

Protease (enzim proteolitik) merupakan enzim yang mampu menguraikan atau memecah molekul protein. Salah satu kelompok mikroorganisme yang mampu menghasilkan protease, yaitu aktinobakteri. Penelitian ini bertujuan untuk mengisolasi dan menguji aktitivas enzim proteolitik (protease) dari aktinobakteri isolat lokal (AKJ-09) Aceh. Penelitian ini dilaksanakan dengan melakukan peremajaan isolat, penapisan isolat yang berpotensi sebagai penghasil protease, penentuan kurva produksi protease, dan pengukuran aktivitas protease dan kadar protein. Isolat aktinobakteri AKJ-09 diseleksi untuk diisolasi dan diuji aktivitas proteolitiknya karena menunjukkan aktivitas proteolitik tertinggi dengan Indeks proteolitik (IP = 19,93) di antara kedelapan isolat aktinobakteri lainnya yang secara kualitatif menunjukkan aktivitas proteolitik pada media NAS (Nutrient Agar + Susu Skim). Berdasarkan uji aktivitas proteolitiknya, isolat AKJ-09 menunjukkan aktivitas protease tertinggi pada hari kedelapan sebesar 0,088 U/ml dengan kadar aktivitas spesifik mencapai 7,56 U/mg.

Abstract

Human herpesvirus 6 (HHV-6) is a widespread causative agent of exanthum subitum or roseola infantum. The species of HHV-6 is divided into two variants, HHV-6A and HHV 6-B, which might be detected and identified by PCR and RE methods. The three samples of eye swabs from children with suspected HHV-6 neurological illness were extracted by guanidium isothiocyanate method before running in PCR followed by agarose electrophoresis as a detection stage. The positive results were then identified by RE analysis to determine the variant of the viruses. One of the three samples (sample 2) shows positive result in direct electrophoresis and it was identified as HHV-6B which produced two fragments of 153bp and 70bp in the RE analysis. PCR and RE analysis are useful methods in detection as well as identification of HHV-6

Abstract

Searching for novel bioactive compounds of microorganisms is increasing. Actinomycetes are a group of bacteria that have potential as a major producer of bioactive compounds. This study aimed to isolate and evaluate the isolates of actinomycetes collected from soil. This research was carried out in three stages, namely the field sampling, isolation of actinomycetes, screening the potential as a producer of bioactive compounds (antimicrobials). A total of 11 isolates of actinomycetes have been isolated in this study. The isolates varied in morphological characteristics. The antibacterial evaluation showed that nine isolates (81.8%) that were able to inhibit either the test organism. Three isolates of ACT-04, ACT-05, ACT-06 and ACT-11 were capable of inhibiting both bacterial targets.

Abstract

Abstract: Entomopathogenic fungi are essential to consider as one of the biological agents to control termite populations. This research aimed to molecularly identify entomopathogenic fungi isolates in termites based on the ITS rDNA region and to determine the relationship of fungi isolates. Identification was performed by DNA extraction, PCR amplification, electrophoresis, purification, and sequencing. Phylogenetic trees were generated using MEGA X.  Molecular identification showed that the ISO1 sample was Penicillium oxalicum, the ISO2 sample was Trichoderma ghanense the ISO3 sample was Aspergillus niger, the ISO4 sample was Aspergillus fumigatus and the ISO5 sample was Aspergillus pseudonomius. The phylogenetic tree showed that the ISO1, ISO2, ISO3, ISO4, and ISO5 samples had the closest relationship with Penicillium oxalicum strain FR6-CGR12, Trichoderma ghanense isolate TM2, Aspergillus niger isolate 77, Aspergillus fumigatus, and Aspergillus pseudonomius strain DTO 267D6, respectively.Abstrak: Kelimpahan jenis fungi entomopatogen adalah hal yang terpenting untuk dipertimbangkan sebagai agen hayati bagi populasi rayap.Penelitian ini bertujuan untuk mengidentifikasi secara molekular isolat fungi entomopatogen pada rayap, berdasarkan daerah ITS rDNA dan mengetahui hubungan kekerabatan dari isolat fungi tersebut. Identifikasi dilakukan dengan cara ekstraksi DNA, amplifikasi menggunakan PCR, elektroforesis, purifikasi dan sekuensing. Selanjutnya kontruksi pohon filogenetik menggunakan aplikasi MEGA X. Berdasarkan uji molekular menunjukkan bahwa sampel ISO1 merupakan Penicillium oxalicum.ISO2 merupakan Trichoderma ghanense.ISO3 merupakan Aspergillus niger. ISO4 merupakan Aspergillus fumigatus.ISO5 merupakan Aspergillus pseudonomius.Konstruksi pohon filogenetik menunjukkan bahwa, sampel ISO1 berkerabat dekat dengan Penicillium oxalicum strain FR6-CGR12. Sampel ISO2 berkerabat dekat dengan Trichoderma ghanense isolat TM2. Sampel ISO3 berkerabat dekat dengan Aspergillus nigerisolat 77. Sampel ISO4 berkerabat dekat dengan Aspergillus fumigatus.Sampel ISO5 berkerabat dekat dengan Aspergillus pseudonomius strain DTO 267D6.
