Abstract

The aim of this study was to determine the effect and the best dose of Sambiloto (Andrographis paniculata) extract supplementation on total erythrocytes and total leukocytes in broilers. The study was conducted for 6 months, May – October 2021. Maintenance and treatment of broilers for 30 days was carried out at the Karang Anyar Farm broiler cage unit in Karang Anyar Village, Jati Agung District, South Lampung Regency, Lampung Province. The experimental study used a completely randomized design with five treatments. Each treatment consisted of 25 broilers of the Cobb CP 707 strain which were divided into 5 cages, so a total of 125 broilers in 25 cages. Sambiloto (Andrographis paniculata) as an immunomodulator was added to drinking water with different doses according to broiler body weight, namely drinking water without a liquid extract of Sambiloto (Andrographis paniculata) (P0); drinking water with Sambiloto liquid extract 0.05 cc/kg BW (3 mg/kg BW) (P1); 0.1 cc/kg BW (6 mg/kg BW) (P2); 0.2 cc/kg BW (12 mg/kg BW) (P3); 0.4 cc/kg BW (24 mg/kg BW) (P4). Total of 25 blood samples from 30-day-old broiler were taken through the brachial vein using a 3 mL syringe and stored in a 5 mL EDTA tube. Calculation of total erythrocytes and total leukocytes at the Palembang Health Laboratory and then the data were analyzed descriptively. The highest total erythrocyte in broilers was P0, the lowest was P4, the mean total erythrocyte was within normal limits for all treatments. The highest broiler leukocyte total was P2, the lowest was P3, the average total leukocyte for all treatments was above the normal range. The active compound in Sambiloto (Andrographis paniculata) is andrographolide which can act as an immunostimulant for specific and non-specific immune functions through NK cells, macrophages, and cytokine induction. The flavonoid content in sambiloto will also send intracellular signals to cell receptors to increase its activity when immune system activity is reduced which can increase the production of IL-2 thereby increasing lymphocyte proliferation, increasing the proliferation and differentiation of B and NK cells. The conclusion was that supplementation of Sambiloto (Andrographis paniculata) extract through drinking water can maintain the number of erythrocytes within normal limits and increase the number of leukocytes as an immune response after vaccination with the best supplementation dose in drinking water of 6 mg/kg BW which is equivalent to a volume of 0.1 cc/liquid extract. kg BB broiler.

Abstract

The research was conducted at Jati Agung District Lampung Selatan Regency on December 2019, aimed to know infestation levels of gastrointestinal helminths on Mud Bufallo. The research used cencus method. Data collection has been done by taking all faecal samples from 80 Mud Bufallo at 6 village in Jati Agung District. Faecal samples examination checked with Mc. Master and Sendimentation test at Lampung Veterinary Hall. Data were analyzed descriptively. The results showed that of the 80 buffalo faeces samples examined, 23 samples (28.75%) were positively infested with gastrointestinal helminths in both single and multi-species infestations. The highest infestation found in Sinar Rejeki village about 100% and the lowest infestation found in Jatimulyo village about 18,51%. Helmiths species that found in Mud Bufallo are from Trematode class (Paramphistomum sp.) Nematode class (Toxocara sp., Bunostomum sp., Strongyloides sp., Haemonchus sp., Oesophagustomum sp., Trichostrongylus sp., and Mecistocirrus sp.), and Cestode class (Moniezia sp.).

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi Vitamin E, Selenium dan Zinc dalam air minum terhadap berbagai parameter histomorfometri usus halus broiler. Penelitian dilaksanakan selama 30 hari (Juni-Juli 2021) di unit peternakan broiler di Desa Adimulyo, Gading Rejo, Pesawaran, Lampung. Penelitian ini bersifat eksperimental menggunakan rancangan acak lengkap, 5 kelompok perlakuan dengan 5 ulangan setiap kelompok. Setiap ulangan terdiri dari 5 ekor broiler, sehingga total 125 ekor broiler. Suplementasi Vitamin E, Selenium dan Zinc pada air minum dengan 5 perlakuan dosis berbeda yaitu air minum tanpa Vitamin E, Selenium dan Zinc (P0); air minum yang mengandung Vitamin E, Selenium dan Zinc dengan dosis 0,5 g/30 kg BB/hari (P1); 1 gr/30 kg BB/hari (P2); 1,5 g/30 kg BB/hari (P3); dan 2 g/30 kg BB/hari (P4), tiap gramnya mengandung Vitamin E 40 IU, Selenium 0,4 mg dan Zinc 160 mg. Lima ekor ayam broiler per kelompok diambil secara acak dan dilakukan nekropsi pada umur 31 hari, diambil sampel usus halusnya, difiksasi dengan formalin 10% dan dibuat preparat histologi di Balai Veteriner Lampung. Pengamatan menggunakan Mikroskop Binokuler Leica DM500® yang terhubung dengan layar dan komputer untuk menghitung berbagai parameter histomorfometri pada usus halus broiler. Data hasil pengukuran rata-rata dianalisis menggunakan one way ANOVA yang dilanjutkan dengan uji Tukey. Hasil penelitian yaitu P3 mempunyai pengaruh yang signifikan (P<0,05) terhadap peningkatan ukuran kedalaman kripta duodenum; P2 berpengaruh nyata (P<0,05) terhadap peningkatan ukuran lebar apex vili ileum; P1 berpengaruh signifikan (P<0,05) terhadap peningkatan kedalaman kripta ileum. Kesimpulan penelitian ini bahwa suplementasi Vitamin E, Selenium dan Zinc pada air minum dapat meningkatkan ukuran kedalaman kripta duodenum dan ileum serta lebar vili apex ileum broiler. Kata kunci: Histomorfometri, Selenium, Usus Halus Broiler, Vitamin E, Zinc.
