Abstract

Aim of this study was to determine the effect of pre-treatment of mangosteen rind juice on the pharmacokinetics of rifampicin on white male Wistar rats. Rats (10) devided into 2 groups, each group consisted of 5 rats. Group I rats were given a dose of rifampicin 50 mg/kg body weight as the control group. Group II rats were given pre-treatment  mangosteen rind juice 200 mg/kg for 7 days and then on day 8 were given rifampicin 50 mg/kg and ma ngosteen rind juice 200 mg/kg as the treatment group. On the hour of 0,25; 0,5; 1; 1,5; 2; 3; 4; 6; 8; 10; 12 and 24  blood sample was taken and then deter-mined the rifampicin plasma levels by HPLC. The results obtained produced curve rifampicin levels in the blood versus time and pharmacokinetic parame-ters of rifampicin such AUC0-∞ , AUMC0-∞, and MRT and MRT can be determined.The results showed that pre-treatment mangosteen rind juice did not have a signiϔicant effect on the pharmacokine tics of rifampicin based parameters AUC0-∞, AUMC0-∞, and MRT.Keywords: pharmacokinetics, mangosteen rind, rifampicin Penelitian ini bertujuan untuk mengetahui pengaruh praper-lakuan jus kulit manggis terhadap proϐil farmakokinetika rifampisin pada ti-kus Wistar jantan. Hasil yang diperoleh dibuat kurva kadar rifampisin dalam plasma terhadap waktu kemudian ditentukan nilai parameter farmakokinetik rifampisin seperti AUC0-∞, AUMC0-∞, dan MRT. Hasil penelitian menunjukkan bahwa pemberian praperlakuan jus kulit manggis tidak memberikan penga-ruh yang signiϐikan terhadap farmakokinetika rifampisin berdasarkan para-meter AUC0-∞, AUMC0-∞, dan MRT.Kata kunci: farmakokinetika, kulit manggis, rifampisin

Abstract

ABSTRACTSurgical site infection is one of surgical infection that frequently happened in hospital sothat the use of antibiotic prophylaxis must be safe. The research was conducted to know incidenceof adverse event at usage of antibiotic prophylaxis in surgery installation. We use concordancesheet of antibiotic prophylaxis either from accuracy aspect of the drug type, administration time,duration of usage. Result analysis in the form of calculation relative risk of usage of antibioticdissected pre prophylaxis to risk the happening of surgical site infections cases. Result thisresearch show that relative risk value of usage of antibiotic to case of infection of operation hurtevaluated from inaccurate of type antibiotic 1,12 ( CI 95 %: 0,138-9,1), inaccurate aspect of gift time5,8 ( CI 95 % : 1,024 - 32,86) and from inaccurate aspect of duration of gift 3,21 ( CI 95 % : 1,043 -9,9).Keywords: antibiotic prophylaxis, surgical site infections, relatives risk

Abstract

Rifampisin merupakan salah satu obat yang dipergunakan sebagai terapi lini pertama dalam pengobatan tuberkulosis. Tujuan dari penelitian ini untuk mengetahui pengaruh pra-perlakuan pemberian madu terhadap profil farmakokinetika fase eliminasi rifampisin pada tikus Wistar Jantan.  Dalam penelitian ini hewan uji dibagi menjadi 2 kelompok, yaitu kelompok kontrol dan perlakuan.  Setiap kelompok terdiri dari 5  ekor tikus.  Kelompok kontrol diberikan rifampisin dosis tunggal 50 mg/kg tikus sedangkan kelompok perlakuan diberikan madu 7,65 ml/kg secara oral sekali sehari selama 7 hari dan pada hari ke-8 diberikan rifampisin dosis 50 mg/kg BB tikus secara per oral. Sebanyak 0,2 ml sampel darah diambil dari vena lateralis ekor  pada 0.25; 0.5, 1.0, 1.5, 2.0, 3.0; 4.0; 6.0; 8, 0; 10.0; 12.0, dan 24.0 jam.  Penetapan kadar rifampisin dalam plasma dilakukan dengan metode HPLC pada panjang gelombang 244.6 nm. Parameter farmakokinetika fase eliminasi yang ditetapkan adalah k, t ½, dan ClT. Hasil penelitian menunjukkan bahwa pemberian pra perlakuan madu tidak mempengaruhi farmakokinetika fase eliminasi dari rifampisin.

Abstract

 Abstract Ascorbyl palmitate is a derivative of ascorbic acid which has a more lipophilic propertis. Ascorbic acid as a hydrophilic compound in topical formulation has low penetration through the skin. Ascorbyl palmitate has better penetration through the skin than that of ascorbic acid. Problem of ascorbic acid and its derivatives that it has a low stability in topical formulation. Microemulsion may increase stability of drugs and have better penetration through the skin. Microemulsion has high surfactant concentration in the formula which followed irritation. The aim of this research was to evaluate stability and primary irritation ascorbyl palmitate microemulsion. Ascorbyl palmitate microemulsion was made by low energy process. Physical stability of microemulsion was examined for viscosity, phase separation and homogeneities. Stress testing was also performed to the  drug for hydrolisis (0.1 N HCl, water and 0.1 N NaOH) and oxidation (3% H2O2). Primary irritation test conducted with techniques on rabbits skin patch test. Rabbits were divided into 2 groups on the incision (n = 5) and not in the incision (n=5). Each group consisted of distilled water control, base control and treatment groups with dose variations 0.25 ml / inch, 0.50 ml / inch, 1 ml / inch and 2 ml / inch. Ascorbyl palmitate microemulsion change in colour, viscosity and pH values during storage at temperature of 25°C and of 40°C. The stability of microemulsion, ascorbyl palmitate underwent degradation by route of hydrolysis and by oxidation. Primary irritation index values obtained respectively 1.275, 1.825, 1.875 and 1.350.  Ascorbyl palmitate microemulsion produce irritation index slightly irritating. Keyword:  Ascorbil palmitate, mikroemulsion, stability, surfactan,skin irritation   Intisari Vitamin C memiliki beberapa turunan, antara lain berupa askorbil palmitat yang  memiliki kepolaran yang rendah, salah satu kendala dalam membuat sediaan vitamin C topikal adalah rendahnya stabilitas dan penetrasi ke dalam kulit. Mikroemulsi memiliki keunggulan antara lain mempunyai kestabilan yang baik, serta dapat meningkatkan penetrasi obat ke dalam kulit. Penelitian ini bertujuan mengetahui kestabilan mikroemulsi askorbil palmitat dan tingkat toksisitas dengan mengukur indeks iritasi pada kulit kelinci. Mikroemulsi askorbil palmitat dibuat dengan teknik low energy, dilakukan uji stabilitas fisik meliputi pH, visikositas, pemisahan fase dan homogenitasnya. Uji stabilitas kimia dilakukan dengan prosedur uji hidrolisis (0,1 N HCl, air dan 0,1 N NaOH) dan oksidasi (H2O2 3%). Uji iritasi primer dilakukan dengan teknik uji tempel pada kulit kelinci yang terbagi menjadi 2 kelompok yang di insisi (n=5) dan tidak di insisi (n=5).  Tiap kelompok terdiri dari kontrol akuades, kontrol basis dan perlakuan dengan variasi dosis pengolesan 0,25ml/inci, 0,50 ml/inci, 1 ml/inci dan 2 ml/inci. Hasil stabilitas fisik dari mikroemulsi mengalami perubahan pH, visikositas, warna dan homogenitas. Stabilitas kimia menunjukan askorbil palmitat mengalami degradasi secara hidrolisis dan secara oksidasi. Bedasarkan uji iritasi primer skor eritma dan edema pada kulit yang di insisi lebih besar daripada kulit yang tidak diinsisi, sedangkan berdasarkan dosis pengolesan didapatkan nilai indeks iritasi primer berturut-turut 1,275; 1,825; 1,875 dan 1,350 sehingga dapat disimpulkan sediaan tersebut memiliki indeks iritasi dengan kategori sedikit mengiritasi. Kata kunci : Askorbil palmitat, mikroemulsi, stabilitas, surfaktan, iritasi kulit

Abstract

Honey and curcumin is widely consumed by children as food supplements. Honey is useful as an antioxidant and antibacterial, while curcumin  is useful as an antioxidant, antiinflammatory,  antibacterial,  antiviral. Both  health  supplements  are  not  infrequently used in conjunction with drugs, especially paracetamol. This study aims to determine the effect of curcumin and honey on the pharmacokinetic parameters of paracetamol in male rats.  In  this  study,  test  animals  were  divided  into  3  groups:  control,  treatment  I  and treatment  II.  In  the  control  group  mice  were  given  paracetamol  150mg  /  KgBW, treatment  group  I  rats  were  given  curcumin  18  mg  /  KgBW,  1  hour  and  then  given paracetamol 150mg  / KgBW whereas  the  treatment group  II  rats given a dose of honey 7.65 mL / KgBW along with giving paracetamol 150mg KgBB . The results showed that administration of curcumin on the pharmacokinetics of paracetamol in mice can decrease the  primary  parameters  of  the  elimination  phase  of  CLT  for  68.25%,  increasing  the secondary parameter t 1 / 2 at 193.72%, thus increasing the derivative parameters AUC0-inf for 88.36%, and MRT of 155.17% (p

Abstract

IbM aims to improve the management of hypertension and Diabetes mellitus (DM) through optimizing the Posbindu (Integrated Development Position) Position of Terong and Jatimulyo Villages. The target is the establishment of posbindu with adequate facilities and trained posbindu cadres. Other targets are the formation of IEC media on hypertension and DM and obtaining baseline clinical parameters data and after management for 3 months. Devotion is implemented within 8 months in fiscal year 2017 in cooperation with the government of Terong Village and Jatimulyo and coordinate with local health center that is Dlingo II Public Health Center. The first stage is cadre training and posbindu formation. The training was conducted by using the method of "Learning Active Cadre" (CBKA), cooperative learning, case-based learning with material on Posbindu implementation, risk factor measurement, early detection, hypertension and DM management, herbal treatment, and basic counseling techniques to play motivator healthy lifestyle. After that done early detection (baseline data) and management of hypertension and DM disease for 3 months in order to know the level of improvement of disease management and as a picture of program sustainability. To improve the knowledge of the community also provided IEC media about Posbindu and hypertension and DM. Until the end of October 2017, the Dedication team has completed several programs such as training candidate cadres posbindu, procurement of medical equipment, and the establishment of posbindu in the Village Terong and Jatimulyo. Based on clinical examination data at the opening of posbindu, monitoring and final measurement can be concluded that the implementation of Posbindu has been able to detect, monitor and improve the management of DM patients and hypertension in both villages in order to achieve controlled clinical condition.

Abstract

Hyperlipidemia is a pathological condition due to lipid metabolism abnormalities. Increased and oxidized LDL in hyperlipidemia will trigger an inflammatory response and produce proinflammatory cytokines, such as vascular cell adhesion molecule-1 (VCAM-1) and Interleukin-6 (IL-6). Curcumin can be used as antioxidants, anti-inflammatory and antihyperlipidemia, but as it is practically insoluble in water, formulation of curcumin nanosuspension is made to improve the effects of curcumin therapy. This study was conducted to determine the activities of nanocurcumin preparation as preventive measures for rats induced with hyperlipidemia. The test animals used were 49 male Wistar rats divided into 7 test groups: normal control group, negative control group, 80mg/kg BW/day curcumin control group, and nanosuspension curcumin dose of 80mg/kg BW/day, nanoemulsion curcumin dose of 80mg/kg BW/day, SNEDDS curcumin dose of 80mg/kg BW/day and solid lipid nanoparticles dose of 80mg/kg BW/day. Provision of preventive measure was performed on days 1-67. On day 11-67, the induction of lard and egg yolk (1:1) was administered with a volume of 2mL/200g of rat BW. On the 68th day, blood samples were taken for the determination of VCAM-1 and IL-6 parameters using Biotin-Streptavidin-Amplified Enzyme-Linked Immuno-sorbent Assay method. Data analysis of VCAM-1 and IL-6 levels between test groups were conducted by means of normality test and One-way ANOVA (p<0.05). Based on the data of VCAM-1 and IL-6 levels, nanocurcumin dose of 80mg/kg BW preparation was able to significantly improve the preventive activity of curcumin compared to curcumin suspension dose of 80mg/kg BW by reducing VCAM-1 levels in SNEDDS (53.260%), nanoemulsion (52.737%), nanosuspension (52.325%) and solid lipid nanoparticles (51.444%) and decreasing IL-6 levels in SNEDDS (33.030%), nanoemulsion (31.568%), nanosuspension (29.898%), and solid lipid nanoparticles (28.875%).

Abstract

This study was conducted to determine the potential of ethanolic extract of red spinach leaves (Amaranthus tricolor L.) with control quality as a complementary treatment of hyperlipidemic based on histopathology and activity of alanin aminotrasferase (ALT). A total of 24 male Wistar rats were randomized in 6 groups: normal group; positive control group were given the drug simvastatin; negative control group; 1st treatment group was given extract at dose of 400mg/kgBW rat, 2nd group was given extract at dose of 400mg/kgBW rat and simvastatin dose 0.18 mg/kgBW rat; 3rd treatment groups were given extract at dose of 800mg/kgBW rat and simvastatin dose 0.18 mg/kgBW rat. Induction hyperlipidemic using high fat diet and poloxamer. The data obtained were tested normality with the Shapiro-Wilk test. Statistical analysis using Oneway ANOVA and Post-Hoc Tukey HSD to determine the significance of differences between groups for ALT parameter. Result show the use of ethanolic extract of red spinach leaves can reduce fatty liver condition based on decreased level of ALT and liver histopathologic. It is concluded that the ethanolic extract of red spinach leaves dose 400mg/kgBW rat combine with simvastatin can reduce activity of ALT until 31.57 U1-1.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas ekstrak etanolik daun bayam (Amaranthus tricolor L.) terstandar sebagai upaya preventif steatosis berdasarkan aktivitas enzim ALT dan histopatologis hati. Hewan uji yang dipergunakan adalah tikus Sprague-Dawley jantan. Sebanyak 30 tikus dibagi ke dalam 6 kelompok yaitu kelompok normal, kontrol negatif, kontrol positif (simvastatin 0,9 mg/kgBB), perlakuan I (ekstrak bayam merah 200 mg/kgBB), perlakuan II (ekstrak bayam merah 400 mg/kgBB), dan perlakuan III (ekstrak bayam merah 800 mg/kgBB). Hari pertama sampai hari ke-67 seluruh kelompok kecuali kelompok normal dan kontrol negatif dilakukan pemberian terapi preventif, selanjutnya hari ke-8 sampai hari ke-67 dilakukan induksi diet tinggi lemak dua jam setelah pemberian terapi preventif. Penetapan kadar ALT dilakukan pada hari ke-0 dan 67. Pada hari ke-68 dilakukan pemeriksaan histopatologi hati. Analisis menggunakan One Way Anova yang dilanjutkan analisis Post-Hoc Tukey HSD. Hasil penelitian menunjukkan ekstrak etanolik daun bayam merah terstandar dosis 200 mg/kgBB, 400 mg/kgBB dan 800 mg/kgBB dapat mempertahankan kadar ALT dalam rentang normal dan memperlihatkan gambaran histopatologi hati yang normal. Disimpulkan bahwa ekstrak etanolik daun bayam merah terstandar berpotensi sebagai terapi preventif steatosis.
