Abstract

Penelitian uji daya hambat dan analisis KLT bioautografi hasil partisi ekstrak etanol herba bandotan (Ageratum conyzoides L.) terhadap Shigella dysentriae  telah dilakukan. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak tersebut dalam menghambat pertumbuhan S.dysentriae dan membandingkan daya hambat ekstrak dengan tingkat kepolaran yang berbeda yaitu pada ekstrak n-heksana, larut etil asetat, dan tidak larut etil asetat berdasarkan  pengukurandiameter hambatan yang terbentuk. Herba tersebut diekstraksi dengan etanol menggunakan metode maserasi, lalu dipartisi dengan pelarut n-heksana dan etil asetat. Pengukuran dilakukan dengan menggunakan metode difusi pada medium Miller Hinton Agar (MHA) dengan waktu inkubasi 24 jam pada suhu 37oC dan memberikan diameter daerah hambatan terbesar pada ekstrak tidak larut etil asetat herba bandotan yaitu 9,3 dan 10,3 mm. Pemisahan secara KLT pada ekstrak n-heksana, ekstrak etil asetat, dan ekstrak tidak larut etil asetat dengan cairan pengelusi berturut-turut, n-heksana:etil asetat (3:1), n-heksana:etil asetat (1:1), dan etil asetat:etanol (10:1) dengan jumlah bercak noda berturut-turut 4, 5, dan 2. Nilai Rf pada ekstrak n-heksana 0.25, 0.41, 0.52, dan 0.71 sedangkan pada ekstrak etil asetat 0.34, 0.53, 0.65, 0.81, dan 0.92 pada ekstrak tidak larut etil asetat 0.33 dan 0.64. Hasil KLT bioautografi diperoleh komponen antibakteri yang diidentifikasi ekstrak n-heksana adalah golongan steroid dan pada tidak larut etil asetat golongan polifenol ABSTRACTInhibition test research and TLC bioautographic bioassay method of the partition results of the ethanol extract of bandotan (Ageratum conyzoides L.) herb against Shigella dysentriae have been conducted. This study aims to determine the ability of these extracts to inhibit S.dysentriae growth and to compare the inhibition of extracts with different polarity levels, n-hexane soluble, ethyl acetate soluble, and ethyl acetate insoluble extracts based on the diameter measurement of the formed resistance. The herbs were extracted with ethanol using the maceration method, then partitioned with n-hexane and ethyl acetate solvents. Measurements were carried out using the diffusion method on Miller Hinton Agar (MHA) medium with an incubation time of 24 hours at 37oC and gave the largest diameter area of resistance to the ethyl acetate insoluble extract of bandotan herb, value 9.3 and 10.3 mm. Separation by TLC on n-hexane extract, ethyl acetate extract, and ethyl acetate insoluble extract with elusive liquid respectively, n-hexane: ethyl acetate (3: 1), n-hexane: ethyl acetate (1: 1), and ethyl acetate: ethanol (10: 1) with the number of stains 4, 5, and 2, respectively. 0.81, and 0.92 in ethyl acetate insoluble extracts 0.33 and 0.64. The results of the bioautography TLC bioassay method showed that the antibacterial component identified in the n-hexane extract was a steroid compound and ethyl acetate insoluble was a polyphenol compound.

Abstract

Burns is one of the incidents that can lead to death (mortality). One of the natural products that have potential to serve as an alternative treatment of burns is orange peel. Orange peel has a chemical composition such as ascorbic acid, vitamin E, vitamin A, and polyphenols as antioxidants that inhibit free radicals responsible in the pathogenesis of both acute and chronic inflammatory. In this study, formulation was made in the form of biocellulose which is the primary metabolism product of bacteria. The purpose of this research was to obtain the concentration of the extract of orange peel on bioselullose that have the effect of decreasing the burn wound in rats. Orange peel was extracted then fortified into biocellulose with a concentration of 3%, 6%, and 9%. After that, the wound healing was tested on animals in the form of decreasing the wound diameter. The results showed that the concentration of extract of orange peel 3% on the fortification of biocellulose showed the good percentage of burn wound decreasing i.e. 45.52% with diameter average of 18.35 mm. This result indicates the concentration of extract of orange peel 3% is better than others.

Abstract

Currently, the antibacterial research is increasingly promoted primarily from natural materials, due to the increasing number of pathogenic bacteria that have been resistant to existing antibiotics. One of the pathogenic bacteria that has been much resistant to antibiotics is Staphylococcus aureus. Rosella calyx (Hibiscus sabdariffa L.) and chitosan are known to have antibacterial activity. The aim of this study to find out the antibacterial synergy of the roselle calyx extract and chitosan against. S.aureus ATCC 33592. Rosella calyx was extracted by maceration using 80 % ethanol, while chitosan is obtained from deasetilation chitin of shrimp husk. Test antibacterial synergism using checkboard assay method by calculating the minimum inhibitory concentration (MIC) using microdilution assay. The results showed minimum inhibitory concentration (MIC) value of roselle calyx extract and chitosan were 1250 ppm and 50 ppm, respectively. MIC value of rosella calyx extract in the presence of chitosan was 625 ppm, while the value of MIC chitosan in the presence of rosella calyx extract was < 0.19 ppm. Fractional Inhibition Concentration Index (FICI) was < 0.5 which concluded that the combination of roselle calyx extract with chitosan has a synergistic antibacterial effect on S.aureus ATCC 33592.

Abstract

Noni leaves (MorindacitrifoliaL.) are known to provide antibacterial properties and can be used in ointment formulations to prevent or eliminate germs that might cause infections, such as Staphylococcus aureus. This research aimedto formulate an ointment containing noni leaf ethanol extract that fulfills the physical quality test standards and to determine the antibacterial activity of various concentrations of ethanol extract of noni leaves in ointment preparations against Staphylococcus aureus bacteria. The extraction technique employed was maceration with 96% ethanol. The extracted substance was formulated into ointments with concentrations of 2.5%, 5%, and 10%. The antibacterial activity of the ointment was evaluated using the agar diffusion method with wells. Organoleptic, homogeneity, pH, spreadability, and adhesion tests demonstrated that the ointment at the three concentrations of 2.5%, 5%, and 10% satisfied the physical quality test standards. The antibacterial activity test revealed that a 96% ethanol extract of noni leaves inhibited the growth of Staphylococcus aureus bacteria with the maximum diameter of the inhibition zone at a concentration of 10% (15.26 mm) and was classified as strong.

Abstract

Propionibacterium acnes is one of the gram-positive bacteria that can lead to the development of acne. One of the plants with the potential to be an anti-acne agent is the Petai Cina leaf (Leucaena leucocephala (Lam.) De Wit.). This study aims to formulate a peel-off gel mask preparation of the ethanol extract of Petai Cina leaf (Leucaena leucocephala (Lam.) De Wit.) that meets the physical quality test requirements and to determine the antiacne activity of the formulation. The extraction method used was maceration with 96% ethanol solvent. From the obtained extract, peel-off gel mask preparations were made and tested for their minimum inhibitory concentration (MIC): FI (2.5%), FII (5%), and FIII (10%), Medi-klin® (positive control), and gel base (negative control). The antibacterial activity was tested using the well diffusion method. The results of this study showed that the peel-off gel mask preparations FI (2.5%), FII (5%), FIII (10%), and the gel base (negative control) met the physical quality test requirements. The results of the anti-acne activity testing against the growth of Propionibacterium acnes bacteria obtained concentrations of extract FI (2.5%) 18.53 mm, FII (5%) 18.85 mm, FIII (10%) 22.27 mm, Medi-klin® 26.43 mm, and the negative control of Petai Cina leaf 6 mm. This indicates that the ethanol extract of Petai Cinaleaf has inhibitory effects on the growth of Propionibacterium acnes bacteria as an anti-acne agent. Salah satu tanaman yang berpotensi sebagai antijerawat adalah daun petai cina (Leucaena leucocephala (Lam.) De Wit.). Penelitian ini bertujuan untuk memformulasi sediaan masker gel peel-off ekstrak etanol daun petai cina (Leucaena leucocephala (Lam.) De Wit.) yang memenuhi syarat uji mutu fisik dan untuk mengetahui aktivitas antijerawat formulasi sediaan masker gel peel-off ekstrak etanol daun petai cina (Leucaena leucocephala (Lam.) De Wit.). Metode ekstraksi digunakan yaitu metode maserasi dengan pelarut etanol 96%. Ekstrak yang diperoleh dibuat sediaan masker gel peel-off yang telah di uji konsentrasi hambat minimumnya (KHM) dengan konsentrasi ekstrak FI (2,5%), FII (5%) dan FIII (10%), Medi-klin® (kontrol positif) dan basis gel (kontrol negatif). Pengujian aktivitas antibakteri menggunakan metode sumuran. Hasil penelitian sediaan masker gel peel-off FI (2,5%), FII (5%), FIII (10%) dan basis gel (kontrol negatif) memenuhi syarat uji mutu fisik. Hasil pengujian aktivitas antijerawat terhadap pertumbuhan bakteri Propionibacterium acnes diperoleh konsentrasi ekstrak FI (2,5%) 18,53 mm, FII (5%) 18,85 mm, FIII (10%) 22,27 mm, Medi-klin® 26,43 mm dan kontrol negatif daun petai cina 6 mm. Hal ini menunjukkan bahwa ekstrak etanol daun petai cina memiliki daya hambat terhadap pertumbuhan bakteri Propionibacterium acnes sebagai antijerawat.
