Abstract

The food industry is one of the commodities that always grows and develops in line with the increasing needs and demands of the market. The growth in demand for fermented foods in the market is due in part to the increased interest in healthy food and the awareness of consumers' perceptions that fermentation is a naturally beneficial process. The ferementation process is inseparable from the role of probiotic microorganisms, bioactive compounds, and micronutrients produced through microbial action. This literature review study aims to analyze the role of probiotics in the fermentation of typical foods in Java, which have the potential to act as an immune regulatory system. The results of a literature study on various kinds of fermented specialties in Java and the manufacturing process show that the microbes that play a role in food fermentation consist of mold and bacteria. These microbes have the potential as an immune regulatory system.

Abstract

This research aims to isolate and characterize the fcin enzyme from Ficus septica stem latex. Ficin from Ficus septica stem latex was isolated using column chromatography. Then enzyme activity was tested at different temperature (40oC, 50oC, 60oC, 70oC) and pH (6.0, 7.0, 8.0) levels. Ficin enzyme activity of joint treatment with variations in temperature and pH was analyzed using two-way ANOVA with a factorial pattern followed by Least Signifcant Difference (LSD) test. The results showed that temperature treatment signifcantly affects enzyme activity. However, the treatment of pH and the interaction between temperature and pH did not signifcantly affect the fcin enzyme activity. There was no signifcant difference in fcin activity at the incubation temperatures of 40oC and 50oC, as well as 60oC and 70oC. However, comparing the incubation temperatures of 40oC and 50oC with treatment 60°C and 70°C showed a signifcant difference in fcin enzyme activity. In the treatment of incubation at pH 6, 7 and 8 for fcin enzyme activity showed no signifcant difference. We concluded that the Ficus septica plant latex contained fcin enzyme with an optimum temperature of 60°C and optimum pH of 6, 7, and 8.

Abstract

The diversity of intestinal bacteria in geese correlates with environmental conditions, rearing methods, and consumed feeds. The intestinal bacteria composition is useful for the absorption of nutrition, improving the metabolism, and may be related to the immune system. This study was conducted to examine the intestinal bacteria composition and the diversity of maintained goose in aviaries and barns. This research was an observational exploratory. Five geese were taken purposively from local breeders in Gunungpati District, Semarang City. A total of 5 g of intestinal contents from each sample was used for microbial genome isolation. Then, the genome was amplified to collect 16S rRNA gene region V3-V4. The amplicons were then sequenced using the next generation sequencing (NGS) method (Illumina high-throughput sequencing; paired-end reads) and analyzed using QIIME2 to identify bacterial species. In addition, GC-MS was performed to identify and measure fatty acid contents in the intestinal. The results showed that both rearing and caged goose contained nine phyla of intestinal bacteria. The number of intestinal bacteria of barn geese (SU) reached 32,748 Operational Taxonomy Units (OTU); higher than aviary geese (SK), which was 11,646 OTU. The intestinal bacteria community in barn geese was approved by Phylum TM7 (Saccharibacteria candidate) (53.18%), followed by Firmicutes (32.51%) and Bacteriodetes (5.42%). Whereas on SK Firmicutes was compiled 49.3 4% of total OTU, TM7 (S. candidate) up to 21.17%, and Actinobacteria up to 15.99 %. The abundance of TM7 may contribute to high 9,12-octadecadienoic acid production, while Firmicutes was related to the high production of oleic acid. Based on these data, the reared geese had a more abundant diversity of bacteria than the caged one.

Abstract

Panti Asuhan Yafikhi memiliki lahan kosong yang berpotensi digunakan sebagai perkebunan. Tantangan dalam pengembangan pertanian hortikultura adalah keseimbangan penggunaan bahan organik dan anorganik. Tujuan dari kegiatan pengabdian ini adalah untuk memberdayakan warga panti asuhan Yafikhi dalam (1) merancang arsitektur landscape “Taman TOGA” sesuai dengan luas dan kondisi lahan, (2) mengumpulkan dan menanam toga sesuai dengan landscape yang telah dibuat, (3) Menerapkan bioteknologi dalam pemanfaatan limbah rumah tangga menjadi pupuk organik, (4) merawat dan mengelola kebun TOGA menggunakan pupuk organik, (5) memanfaatkan tanaman obat untuk menjaga kesehatan. Metode yang digunakan adalah workshop, praktik, dan pendampingan. Hasil pengabdian menunjukkan peningkatan pemahaman warga mitra (Panti Asuhan Yafikhi Semarang) tentang pentingnya tanaman obat untuk menjaga kesehatan, terampil merancang dan mewujudkan Taman TOGA, terampil mengelola sampah rumah tangga menjadi kompos padat dan pupuk organik cair, dan terampil menggunakan pupuk organik untuk memelihara taman TOGA.

Abstract

Isu pangan merupakan persoalan krusial bagi seluruh lapisan masyarakat termasuk masyarakat perkotaan. Pemenuhan kebutuhan pangan dapat dilakukan dengan bertanam atau berternak. Urban farming atau pertanian perkotaan merupakan salah satu kunci pemberdayaan untuk ketahanan pangan berkelanjutan. Budidaya akuaponik merupakan sistem terintegrasi antara akuakultur dan hidroponik, limbah budidaya ikan berupa sisa metabolisme dan sisa pakan dimanfatakan untuk pupuk tanaman. Akuaponik dapat dilakukan dalam skala besar ataupun kecil. Keterbatasan lahan di Kelurahan Kalisegoro Kecamatan Gunungpati, Kota Semarang menyulitkan masyarakat melakukan kegiatan bercocok tanam secara konvensional. Tujuan dari kegiatan pengabdian ini adalah untuk memberdayakan warga Kalisegoro dalam memahami konsep urban farming, khususnya akuaponik, dan terampil menerapkan teknologi akuaponik. Metode yang digunakan adalah (1) participatory urban appraisal, (2) participatory technology development, yaitu workshop pemanfaatan teknologi tepat guna berintegrasi ilmu pengetahuan dan kearifan lokal yaitu akuaponik, (3) pendekatan sosialisasi melalui pendampingan dalam membuat dan merawat akuaponik sampai berhasil. Hasil pengabdian menunjukkan peningkatan pemahaman peserta pengabdian terkait konsep hidroponik dan akuaponik, terampil menyemai bibit tanaman hidroponik, terampil mendesain instalasi akuaponik, terampil menghitung nutrisi yang diperlukan dan terampil merawat akuaponik.

Abstract

Cinnamon bark (Cinnamomum verum) contains various bioactive compounds that can be used in drug development, one of which is anti-inflammatory. Inflammation is the body's defense response to the occurrence of an injury from objects unknown to the body, such as bacterial infections, trauma, autoimmune, viruses and toxins. The purpose of this study was to determine the content of bioactive compounds in cinnamon bark. This research method was carried out using the Dr. Duke's Phytochemical database. Duke's Phytochemical database to collect bioactive compounds in cinnamon bark, PubChem to download the ligand structure of bioactive compounds, PASS (Prediction of Activity Spectra for Substance) Online for screening the anti-inflammatory activity value of bioactive compounds, SEA and Swiss Target Prediction are used to determine the target protein, STRING is used to analyze the interaction between target proteins, Cytoscape is used to predict tissue topology, Pyrex is used for molecular docking, and Biovia Discovery Studio is used to visualize docking results. There are 16 cinnamon bark compounds that have a Pa value > 0.5. Based on the docking results, compounds that have a Pa value> 0.5. Based on docking results, beta caryopyhllene, caryopyhllene, fatty acid, and isocaryopyhllene compounds have PPARA activator mechanism with binding affinity of -7.4 kcal/mol, -7.9 kcal/mol, -7.8 kcal/mol, and -5.6 kcal/mol. Caffeic acid compounds have an MMP-9 activator mechanism with a binding affinity of -5.7 kcal/mol. Visualization of docking results shows that isocaryopyhllene and caffeic acid compounds have similar interactions and amino acid residues with control drugs so that they can be used as anti-inflammatory agents.

Abstract

cancer is a disease of excessive cell growth and is one of the cases of disease that has increased every year. cancer treatment therapy in the form of chemotherapy drugs has various side effects so that safer long-term treatment is needed, namely using herbal medicines such as sembukan plants (Paederia scandens). This research was conducted using experimental methods computationally through in silico methods as a potential prediction test as a reference for further studies. isolate compounds are carried out molecular tethering against progesterone, estrogen, HER2, and NfkB receptors using PyRx 0.8 software. Then the isolated compounds were tested for the feasibility of chemical and pharmacokinetic properties and toxicity using the Lipinski Rules of Five data base and ADMETLab 3.0. Furthermore, visualization was carried out to see the bonding interaction between ligand and receptor protein using Biovia Discovery Studio 2024 software. Based on the results of molecular tethering compounds Kaempferol 3-rutinoside-7-glucoside, Cosmetin, Quercetin 3-o-rutinoside-7-o-glucoside, Daidzein and Linarin have the potential as inhibitors respectively against progesterone, HER2, Nf-kB, estrogen and overall average receptors with each binding affinity value of -9. 6 kcal/mol, -9.8 kcal/mol, -9.8 kcal/mol, -10.8 kcal/mol, -8.9 kcal/mol and -9.0 kcal/mol respectively. The best physico-chemical properties were obtained for Daidzein and Linarin compounds. Visualization data generated that Kaempferol 3-rutinoside-7-glucoside, Cosmetin, Quercetin 3-o-rutinoside-7-o-glucoside, Daidzein and Linarin compounds have similar interactions and amino acid residues with control drugs so that they are predicted to have similar pharmacological effects as breast anticancer.
