Abstract

 In vitro propagation method for vetiver (Vetiveria zizanioides L.) had been effectively developed in this study. Several variations of media were used for shoot initiation and multiplication. Shoot formation was initiated from crown explant cultured on MS media with the addition of 2 mgL-1 of growth regulator Benzyladenine (BA).  Whereas for shoot multiplication, 3 mgL-1 of BA was evidently effective with the average shoot number was 126 shoots per explant. The application of high BA concentration (3-5 mgL-1) in multiplication media was capable of inducing more shoots, but the shoots resulted were shorter. In contrast, multiplication media supplemented with low BA concentration (1-2 mgL-1) yielded less shoots, but the shoots were longer. Plantlet regeneration was accomplished by inducing roots in the shoots yielded on MS media containing 1 mgL-1 growth regulator NAA. In vitro plants of vetiver had been successfully grown on soil media under greenhouse condition. By using foregoing method, it is possible to conduct mass propagation of vetiver through tissue culture technique.

Abstract

The objective of this research was to evaluate the effect of gamma irradiation on shoot growth and multiplication of Patchouli (Pogostemon cablin Benth.) Two weeks-old in vitro shoots were irradiated gamma-ray, at doses of 0, 15, 30, 45, 60, and 75 Gy. The control shoot was not irradiated. The irradiated shoots were cultured on Murashige and Skoog (MS) medium supplemented with 0.1 mg.L-1 NAA and 0.3 mg.L-1 BA and incubated in a growth room for eight weeks at a temperature of 25±2°C. The results showed that the gamma irradiation inhibited the growth and multiplication of shoots. Explants irradiated with high-dose gamma-ray (45-75 Gy) had not formed shoot in four weeks of culture, while 58.3-83.3% of the explants without irradiation or irradiated at low doses 15 and 30 Gy formed shoots. The higher irradiation doses increased percentage of browning explants and reduced the percentage of forming shoots. Within the eight weeks of culture, explant without irradiation was able to form shoots at the percentage of 100% with 24 shoots per explant, while explants irradiated at 15-45 Gy were able to grow form shoots at the percentage of 77.7-95.5%. The high doses-irradiated explants (60 and 75 Gy) were only able to form shoots less than 13-20%, with 2-3 shoots per explant. Keywords: Gamma rays (Gy), in vitro shoot, Pogostemon cablin Benth.

Abstract

This study aims to analyze the structure and germination of pollen in porang. Pollen structure and germination were observed microscopically. Pollen germination observations were carried out three days before anthesis (M-3), two days before anthesis (M-2), one day before anthesis (M-1), anthesis (M) to four days after anthesis (M+4). Pollen in porang has a dimorphic structure (X and Y structures: monad and compound units). Structure X with axis length (P)= 30-50 µm, equatorial diameter (E)= 16.67-41.67 µm, and index P/E= 1.19-1.79, while structure Y with P= 50-84 µm, E= 34-67 µm, and P/E index= 1.25-1.40. Porang pollen was subspheroidal and prolate in shape; the length of the axes of the media and magna types; monocolpate aperture type. The highest viability of the X structure was at M (31.58%), while the Y structure was at M+1 (21.76%). M+3 viability begins to decline, so porang artificial pollination is appropriate when pollen viability is high (M or not more than M+3).

Abstract

Penelitian ini bertujuan untuk mengetahui peta persebaran tanaman buah, karakter populasi tanaman buah, serta persepsi masyarakat pemilik tanaman buah di sepanjang jalur wisata Desa Kemiren, Tamansuruh, dan Kampunganyar, Kabupaten Banyuwangi. Metode yang dilakukan meliputi survei pemetaan pertanian tanaman buah (mangga, rambutan, manggis, durian, jambu air dan jambu biji) dengan merekam titik koordinat dari GPS untuk setiap tanaman buah. Penentuan karakter populasi tanaman buah dilakukan dengan mengamati morfologi tanaman terkait vitalitas dan periodisitas. Persepsi masyarakat dilakukan dengan wawancara dan kuisioner. Analisis data dilakukan dengan mengolah data koordinat dan data pengamatan karakter populasi tanaman buah ke dalam peta dasar melalui aplikasi GIS. Pemetaan persepsi masyarakat diperoleh dengan wawancara dan kuisioner yang dihitung dengan skala Likert kemudian dipetakan sebaran spasialnya dengan aplikasi GIS. Hasil penelitian menunjukkan bahwa persebaran tanaman buah yang ada di pekarangan rumah banyak tersebar di sepanjang jalur desa wisata dengan perbandingan jumlah buah yang ditemukan di Desa Kemiren 76 pohon, Tamansuruh 53 pohon, dan Kampunganyar 40 pohon. Kondisi tanaman buah dalam keadaan tumbuh dengan baik, bertunas, berbunga dan berbuah, hal ini dikarenakan pada saat penelitian waktunya tanaman buah memasuki masa berbuah dan masa panen. Antusiasme masyarakat tinggi untuk menjadikan tanaman buah yang ada di sepanjang jalur desa wisata sebagai daya tarik wisatawan. Abstract

Penelitian ini bertujuan untuk mengetahui persepsi masyarakat, mengetahui jenis-jenis tanaman yang digunakan untuk upacara Kasada serta mengetahui peran serta masyarakat Tengger di Desa Ngadas dalam mengkonservasi tanaman yang digunakan upacara Kasada. Metode penelitian yang digunakan adalah metode survei, observasi dan wawancara semi terstruktur dengan menggunakan 50 responden. Analisis penggunaan tumbuhan dengan menggunakan rumus indeks konsensus / fidelity level. Hasil penelitian ini menunjukkan persepsi masyarakat sangat tinggi terhadap pelaksanaan upacara Kasada serta penyerahan hasil bumi. Tanaman yang digunakan untuk upacara Kasada meliputi 16 jenis tanaman. Nilai penggunaan tanaman tertinggi adalah edelweis (Anaphalis longifolia) 96%, padi (Oryza sativa) 94%, kentang (Solanum tuberosum) 90%, bawang prei (Allium fistulosum) 86%, putihan (Buddleja asiatica) 84%, kubis (Brassica oleraceae) 80%, antinganting (Fuchsia magellanica) 78%, pisang raja (Musa paradisiaca) 74%, telotok (Curculigo latifolia) 70%, kenikir/gumitir (Cosmos caudatus) 68%, pinang (Areca catechu) dan beringin (Ficus benjamina) 46%, danglu (Engelhardia spicata) 40%, janur daun kelapa (Cocos  nucifera) 30%, sirih (Piper betle) 28%, dan jagung (Zea mays) 24%. Upaya konservasi keanekaragaman hayati yang dilakukan oleh masyarakat Tengger telah berkembang sejak lama, khususnya pada masyarakat yang memiliki pengetahuan lokal. Konservasi dilakukan dengan menanam flora tersebut di ladang, pekarangan dan jalan-jalan sekitar desa. Konsep pengelolaannya mengacu pada pemanfaatan berkelanjutan untuk memperoleh dinamika ekosistem yang selaras dan seimbang bagi kehidupan masyarakat Tengger.
