Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh auksin dan sitokinin terhadap induksi kalus dan regenerasi tanaman bawang putih (Allium sativum L.) secara in vitro. Medium Murashige and Skoog (MS) dengan penambahan kombinasi 2,4-D dan kinetin digunakan untuk induksi kalus dari eksplan meristem basal,  sedangkan NAA dan BAP digunakan untuk regenerasi planlet dari eksplan kalus. Pertumbuhan kalus pada medium kultur yang mengandung 2,4-D dan kinetin pada konsentrasi rendah lebih baik daripada konsentrasi tinggi. Kalus yang dikulturkan pada medium kultur yang mengandung NAA 0.1 ppm dikombinasikan dengan BAP 0.5-2 ppm mampu membentuk tunas. Jumlah tunas meningkat seiring dengan peningkatan konsentrai BAP. Di antara berbagai macam konsentrasi NAA dan BAP, jumlah tunas terbanyak diperoleh pada medium kultur yang mengandung NAA 0.1 ppm dan BAP 2 ppm. Tunas-tunas dipindahkan ke medium kultur dengan penambahan BAP 10 ppm untuk menginduksi pemanjangan tunas. Planlet bawang putih dihasilkan dari tunas yang dikulturkan pada medium kultur dengan penambahan IBA 2 ppm. Kata kunci: bawang putih, kalus, meristem basal, regenerasi in vitro

Abstract

Light is one of external stimulations affecting the plant’s movements and causing different responses. The time of opening and closing of plant organs caused by light stimulation are different. The aim of this research was to determine the opening and closing time of Portulaca grandiflora’s flower, Sesbania grandiflora’s and Cassia alata’s leaves. The study was conducted in Madiun and Trenggalek, East Java in November 2020 using field survey and observation. The survey was carried out to obtain sample plants, while observations were to observe the opening and closing time of leaves or flower in the sample plants. The observations were carried out at 04.00 to 19.00 every 15 minutes to analyze the time range of opening and closing of flower and leaves. The opening time of Portulaca grandiflora’s flower and the Sesbania grandiflora’s leaves were almost the same, around 09.00 to 10.30. Meanwhile, the Portulaca grandiflora’s flower began to close at almost the same time as the Cassia alata’s leaves, which only 45 minutes of difference (10.45 and 10.00 respectively). The opening and closing of plant organs were influenced by light stimulation with variation time.

Abstract

 In vitro propagation method for vetiver (Vetiveria zizanioides L.) had been effectively developed in this study. Several variations of media were used for shoot initiation and multiplication. Shoot formation was initiated from crown explant cultured on MS media with the addition of 2 mgL-1 of growth regulator Benzyladenine (BA).  Whereas for shoot multiplication, 3 mgL-1 of BA was evidently effective with the average shoot number was 126 shoots per explant. The application of high BA concentration (3-5 mgL-1) in multiplication media was capable of inducing more shoots, but the shoots resulted were shorter. In contrast, multiplication media supplemented with low BA concentration (1-2 mgL-1) yielded less shoots, but the shoots were longer. Plantlet regeneration was accomplished by inducing roots in the shoots yielded on MS media containing 1 mgL-1 growth regulator NAA. In vitro plants of vetiver had been successfully grown on soil media under greenhouse condition. By using foregoing method, it is possible to conduct mass propagation of vetiver through tissue culture technique.

Abstract

The effective method in making chromosome prepararation was able to count the number of chromosome easier, especially for analysis the ploidy level in plants. Garlic (Allium sativum L.) is one of the plants that the chromosome is difficult to be counted because their chromosomes are long and difficult to spread during the squashing. This research was to find out the effect of hydroxiquinoline on making chromosome preparation of root and callus in garlic. The steps of making chromosome preparation were fixation in Farmer’s fluid, put in hydroxiquinoline for an hour, hydrolysis in 1N HCl for two minutes at 60°C, put in Carnoy for 30 minutes, and stained in 1% aceto orcein for 20 minutes. The treatment were gift hydroxiquinoline for an hour after put in fixative fluid and the control were making preparation without gift hydroxiquinoline. The result of this research suggest that the method of making preparation by treatment with hydroxiquinoline for an hour after fixation produced the chromosomes that the stain were sharp and contrast, and also it produced well spread chromosomes that were clearly defined of both callus and root tip in garlic.

Abstract

Abstract: The aim of this study was to evaluate the perceptions of IAIN Tulungagung students on the implementation of the internship program at MTSN 2 Blitar. This study was conducted on March to November 2020. This study used a qualitative approach and secondary data was analyzed descriptively and exploratively. The informants in this study were students of the Tarbiyah and Teacher Training Faculty (FTIK) IAIN Tulungagung who carried out the Internship program 1 and 2 at MTsN 2 Blitar, East Java. Data were collected using questionnaires, observation, documentation, and interviews. As much as 92% of students stated that there was preparation or provision of apprenticeship program at IAIN Tulungagung before the students went to the field. At the same percentage, students claimed to be happy in carrying out an internship program. This was indicated by the existence of special preparations every day before entering the class which was stated by 84% of students. All students (100%) had high enthusiasm for teamwork. According to 84% of students, this was triggered by the atmosphere in the partner institutions to support the implementation of internships. Thus, all students (100%) can conduct socialization in a flexible manner with the school. Even though the implementation of internship was well, there were 30% of students who stated that there were obstacles in its implementation. The condition caused the dissatisfaction was felt by 23% of students. As many as 76% of students stated that there were administrative tasks and as many as 69% of students stated that there were several other tasks given by the school during the internship. Abstract

Abstract Education is one of the main things that can make a significant contribution to the development and progress of a nation. This fact is evidenced by the existence of a generation of people who are quality-produced from educational institutions in Indonesia, if the young generation of this nation does not attend education, perhaps the development and progress of the times will not be able to be like today. Education, especially in Muslims, one of the foundations and guidelines for which is the Koran, through some of the verses it contains, there are some instructions and knowledge that are very extraordinary if we dig into the meaning for the meaning. So it is important for Muslims to make Al-Qur'an as a guide in processing their studies. Education and knowledge are two main things that are interrelated, they are like two sides of a coin that cannot be separated, even always in pairs. In the context of this article, the concept of knowledge and education from the perspective of the Koran will be discussed, namely taking one of the verses in Surah al-Mujadilah, verse 11. The verse explains the various things it contains, then explains Asbabun Nuzul, describes the verse, Menafsiri is then analyzed, hopefully, what is discussed here becomes a study that can increase wealth, both theoretical wealth and scientific wealth practically. Keywords: Science, Education, Al-Mujadilah. Abstrak Pendidikan merupakan salah satu perkara pokok yang mampu memberikan kontribusi signifikan terhadap perkembangan dan kemajuan suatu bangsa. Fakta ini dibuktikan dengan adanya generasi bangsa yang secara kualitas dihasilkan dari bangku lembaga pendidikan yang ada di Indonesia, jika generasi muda bangsa ini tidak mengenyam bangku pendidikan, mungkin perkembangan dan kemajuan zaman tidak mampu seperti sekarang ini. Pendidikan khususnya dalam umat Muslim, salah satu landasan dan yang menjadi pedomannya adalah al-Qur’an, melalui beberapa ayat yang dikandungnya, terdapat beberapa petunjuk dan ilmu yang sangat luar jika diselami makna demi maknanya. Maka menjadi penting bagi umat Muslim menjadikan al-Qur’an sebagai pegangan dalam berproses di dalam menuntut ilmu. Pendidikan dan ilmu merupakan dua hal pokok yang berkaitkelindan, keduanya ibarat dua sisi mata uang logam yang tak mampu dipisahkan, bahkan selalu berpasangan. Dalam konteks artikel ini, maka akan di bahas kosep ilmu dan pendidikan dari persfektif al-Qur’an, yaitu mengambil salah satu ayatnya dalam surat al-Mujadilah ayat 11. Ayat tersebut dijelaskan berbagai hal yang dikandungnya, kemudian dijelaskan asbabun nuzul, mendeskripsikan ayat, menafsiri kemudian dianalisis, semoga apa yang dibahas di sini menjadi sesuatu kajian yang mampu menambah kekayaan, baik kekayaan secara teoritis maupun kekayaan keilmuan secara praktis. Abstract

The aim of this study was to identifiy the stomata morphological characters of Pteridophyta in the ​​Parangkikis Pagerwojo Waterfall area, Tulungagung. The first step of stomata observation was preparation of the abaxial leaf slice. The preparation was carried out by the replica method. Stomata character studied include types and size of stomata, the number of stomata and epidermis cells, and value of the stomatal index. The result of this study showed that stomata types of Pteridophyta were polocytic and anomocytic. Of the 15 Pteridophyta species observed, the all of stomata type were polocytic, except Selaginella which had type stomata anomocytic. Stomata oval was found in Selaginella intermedia and Phymatosorus sp., slightly oval (kidney) was found in Asplenium apogamum, Dryopteris sp., Asplenium normale, Nephrolepis bisserata, Nephrolepis davallioides, Asplenium nidus, and Pteris longipinnula sp., spherical was found in Dicranopteris linearis, Cyclosorus arida, Goniophlebium percussom, and Goniophlebium manmiense, and nonconcave was found in Coniogramme fraxinea. Stomata size affected the number of stomata. If the size of the stomata was small, the number of stomata was increasing. The highest number of stomata was found in D. linearis, which was 362, while the least number of stomata was S. intermedia, which was 18. Data on the number of stomata and epidermal cells were used to determine the stomatal index. The highest stomata index was found in D. linearis, which was 22.05% and the lowest was C. fraxinea, which was 5.44 %. Keywords: Anomocytic, Parangkikis, polocytic, Pteridophyta, stomata

Abstract

Praktikum merupakan bagian penting dalam proses pembelajaran, khususnya dalam perkuliahan Biologi. Praktikum yang baik perlu dilengkapi dengan komponen penunjang, salah satunya adalah buku petunjuk praktikum. Penelitian ini bertujuan untuk mendeskripsikan substansi, kelayakan, dan respon penggunaan buku petunjuk praktikum Biologi Umum Program Studi Tadris Biologi UIN Sayyid Ali Rahmatullah Tulungagung. Jenis penelitian ini adalah R D dengan model pengembangan ADDIE. Hasil penelitian menunjukkan bahwa substansi buku petunjuk praktikum Biologi Umum prodi Tadris Biologi UIN Sayyid Ali Rahmatullah Tulungagung terdiri dari tata tertib, jadwal praktikum, topik, tujuan, dasar teori, alat dan bahan, cara kerja, dan format lembar pengamatan sementara (LPS). Dilihat dari segi teoritis, desain, akurasi, dan relevansinya dengan materi perkuliahan, buku petunjuk praktikum Biologi Umum dinyatakan layak sebagai penunjang praktikum dengan kriteria sangat valid berdasarkan penilaian oleh ahli materi dan media. Penggunaan buku petunjuk praktikum ini mendapatkan respon positif dengan persentase penilaian yang tinggi. Respon positif ditunjukkan dengan pendapat responden yang menyatakan bahwa buku petunjuk praktikum mudah diperoleh, mudah dipahami, tersaji dengan lengkap, langkah-langkah praktikum dipaparkan dengan jelas sehingga dapat membantu dalam pelaksanaan praktikum.  Practicum is an important part of the learning process, especially in Biology lectures. A good practicum needs to be equipped with supporting components, one of which is a practical handbook. The aims of this study were to describe the substance, feasibility, and response to the use of the General Biology practical handbook for the Tadris Biology Study Program in UIN Sayyid Ali Rahmatullah Tulungagung. The type of research was R D with the ADDIE development model. The results showed that the substance of the General Biology practical handbook for the Tadris Biology Study Program in UIN Sayyid Ali Rahmatullah Tulungagung consisted of rules, practicum schedules, topics, objectives, theoretical basis, tools and materials, working methods, and temporary observation sheet (LPS) format. In terms of theoretical, design, accuracy, and relevance to lecture material, the General Biology practical handbook was declared feasible as a practicum supporter with very valid criteria based on assessments by material and media experts. The use of this practical handbook received a positive response with a high percentage of assessment. The positive response was indicated by the opinion of the respondents who stated that the practical handbook were easy to obtain, easy to understand, fully presented, the practicum steps were explained clearly so that it could assist in practicum implementation.

Abstract

ABSTRACTThe campus area of UIN Sayyid Ali Rahmatullah Tulungagung is an environment that has a high diversity of plants. Some students do not have information about plant morphology, especially the Fabaceae family in this area and have difficulty learning it. The aims of this research were to develop a catalog of the Fabaceae family as well as to determine the level of validity. The type of research was R & D with the ADDIE development model. The results of material expert validation obtained a percentage of 79% (valid) and media experts obtained a percentage of 98% (very valid). In the field trial stage, the average posttest score was higher than the pretest score, 83.5 and 66.5, respectively, with the t-test (t-test) results showing a significance value of 0.000 (< 0.005). Then, the average product readability test results in the form of a Fabaceae family catalog obtained a percentage of 88% (very valid). Thus, the Fabaceae family catalog was declared worthy as a media and additional learning resource so that it was give the significant effect on increasing student understanding and learning outcomes. Keywords:Catalog, Fabaceae, Learning Media ABSTRAKKawasan kampus UIN Sayyid Ali Rahmatullah Tulungagung merupakan lingkungan yang memiliki kenanekaragam tumbuhan cukup tinggi. Beberapa mahasiswa belum memiliki informasi mengenai morfologi tumbuhan khususnya Famili Fabaceae di kawasan tersebut dan mengalami kesulitan dalam mempelajarinya. Tujuan penelitian ini adalah mengembangkan katalog Famili Fabaceae sekaligus mengetahui tingkat validitasnya. Jenis penelitian ini adalah R & D dengan model pengembangan ADDIE. Hasil validasi ahli materi diperoleh persentase sebesar 79% (valid) dan ahli media diperoleh persentase sebesar 98% (sangat valid). Pada tahap uji coba lapangan diperoleh rata-rata nilai posttest yang lebih tinggi dibandingkan dengan nilai pretest, masing-masing 83.5 dan 66.5 dengan hasil uji-t (t-test) menunjukkan nilai signifikansi sebesar 0.000 (< 0.005). Kemudian, rata-rata hasil uji keterbacaan produk berupa katalog Famili Fabaceae diperoleh persentase sebesar 88% (sangat valid). Dengan demikian, katalog Famili Fabaceae dinyatakan layak sebagai media dan sumber belajar tambahan sehingga dapat memberikan pengaruh yang signifikan terhadap peningkatan pemahaman dan hasil belajar mahasiswa. Kata kunci:Katalog, Fabaceae, Media Pembelajaran
