Abstract

Indonesia biodiversity has the second highest number of indigenous medical plants after the amazon rain forest, meanwhile Indonesian people usually used plants as a medicine source and some medicine plants have been developed as modern drugs. One of medicine plant in Indonesia that usually used as a refreshment was grass jelly (Stephania capitata (Blume) Spreng). This plant frequently found in Sumatra, Borneo and Java island and the leaf of grass jelly has been reported as a medicine against stomach complaint (diarrhea) and fevers. This research was designed to investigate the secondary metabolites and the efficacy of leaf extract of grass jelly as anti diarrhea. The leaves extraction was done by using maceration with ethanol solvent at room temperature. Meyer reagent, dagendroff reagent, methanol, magnesium band, hydrochloric acid, iron (III) chloride, anhydrous acetic acid, sulfuric acid, chloroform were used for identification of secondary metabolites from grass jelly leaves. For potency anti diarrhea, animals were divided into six group: diarrhea group, treatment group with loperamid HCl and treatment group of leaf extract of grass jelly. The results showed that the leaf extract of grass jelly contain saponin, phenol, sterol and triterpen and the treatment with leaf extract of grass jelly significantly decreasing time duration of diarrhea.

Abstract

Online game is a game that can be accessed by the community using internet access. Online games are known to have a fun and addictive effect on their use. Indonesia is the 16th most online game user in the world. This study aims to provide an overview and the impact of the habit of playing online games on Pharmacy students in Samarinda. The method used in this study was using questionnaires distributed to at least 57 respondents of Pharmacy students in Samarinda. The results show that at least 80% of students have played online games with an average playing time of 1-2 hours in one day. The results of the questionnaire also showed an impact in the form of decreasing students' sensitivity to the surrounding environment and the tendency to speak harshly due to playing online games. Based on the results, it can be concluded that the habit of playing online games can reduce students' sensitivity to their environment and can encourage negative behavior, especially in verbal matters. This research can be used as a preliminary description of behavior changes towards the negative of students due to the habit of playing online games.

Abstract

Breast cancer with Human Epidermal Growth Factor Receptor (HER)2 overexpression increases tumor progession and lead to metastasis, which is primarily cause of mortality in breast cancer. Pentagamaboronon-0 Sorbitol (PGB-0-So) is an aquoeous formulation of curcumin analog, PGB-0, with sorbitol. This compound has been developed as an anti-cancer chemotherapeutic agent and a boron carrying pharmaceutical for boron neutron capture therapy (BNCT). The aims of this study are to investigate antimetastatic activities of PGB-0-So against HER2-overexpressed MCF-7 breast cancer (MCF-7/HER2) cells. The MTT cytotoxicity assay of PGB-0-So exhibited cytotoxic effect with an IC50 value of 35 μM. The testing of anti-migration activity using the scratch wound healing assay demonstrated that PGB-0-So inhibited the closure of the wound on MCF-7/HER2 cells compare to the control. Furthermore, PGB-0-So was able to suppress matrix metalloproteinase (MMP)-9 activities, based on the gelatin zymography assay. In conclusion, PGB-0-So has potency to be developed as an anti-cancer agent against metastatic breast cancer.Keywords : PGB-0-So, anti-metastatsis, cell migration, MMP-9, MCF-7/HER2

Abstract

The incidence of Breast Cancer Metastasis (MBC) can be categorized in stage IV as well as being the leading cause of death in cases of breast cancer. MBC prognosis is known to be weak, frequent recurrent, and MBC patients have only a survival rate of about 5 years. One of the proteins that causes breast cancer metastasis is Human Epidermal Growth Factor 2 (HER2). Pentagamaboron-0 (PGB-0), a newly curcumin analogue performed cytotoxic effect on HER2-positive breast cancer cells but it is practically water-insoluble. The aims of this study are to determine anti-metastatic activity of a more soluble form of PGB-0 namely PGB-0 fructose complex (PGB-0-F) toward HER2 positive cancer (MCF-7/HER2) cells. PGB-0-F was obtained from Cancer Chemoprevention Research Centre Faculty of Pharmacy Universitas Gadjah Mada. Based on scratch wound healing assay result, PGB-0-F inhibited cell migration especially in combination with doxorubicin at the concentration 15 μM. Under gelatin zymography assay, PGB-0-F in combination with doxorubicin decreased Matrix Metalloproteinases 9 (MMP-9) expression compare to the doxorubicin. Hence, PGB-0-F has a potency to be developed as anti-metastatic agent on HER2 overexpression breast cancer.Keywords : HER2, MCF-7/HER2, PGB-0-F, Metastasis

Abstract

Cancer is the highest cause of death in the world. This disease is known to have different regulations from other diseases. The development of drugs today is to target anticancer drugs on cancer regulation sites at Hallmark of Cancer. Lime (Citrus aurantifolia) is one type of Citrus (orange) which contains beneficial chemical compounds and active as anticancer agents. Lime peel is known to have bioactive content in the form of essential oils and flavonoids such as hesperidin and hesperitin. The content of this compound is closely related to anticancer activity. This study aims to identify the content of compounds in lime peel oil and the study of molecular docking silico in these compounds in protein that play a role in the regulation of cancer cells. Lime peel oil is obtained using the steam distillation method then analyzed using GC-MS to determine its constituent components to be analyzed in silico then using YASARA 10.1.8, MarvinSketch 15.4.20, and PLANTS software. The results showed that the main components of lime peel oil were d-limonen and beta-pinen with a percentage of 59.71% and 36.81%. In silico analysis shows that each of these compounds has an affinity greater than the native ligand on caspase-8 proteins (protein regulation of apoptosis). Based on these results it can be concluded, that the lime peel oil has the potential to be developed as an anticancer agent.

Abstract

Pulasan (Nephelium mutabile Blume) merupakan tanaman yang banyak tumbuh di Kalimatan. Tanaman ini diketahui memiliki aktivitas antioksidan yang tinggi tertutama pada bagian kulitnya akan tetapi pemanfaatan aktivitas ini belum banyak dilakukan. Hal ini dibuktikan dengan kurangnya penelitian mengenai formulasi buah pulasan menjadi sediaan farmasi yang mudah dan menyenangkan untuk digunakan. Penelitian ini bertujuan untuk mendapatkan formula optimum effervescent ekstrak kulit buah pulasan yang dapat diterima dan mudah digunakan setelah itu dikaji aktivitas antioksidan dengan metode 2,2-diphenyl-1-picrylhydrazyl (DPPH) menggunakan spektrofotometer UV-Vis. Pembuatan serbuk effervescent ekstrak kulit buah pulasan dilakukan menggunakan empat formula dengan konsentrasi ekstrak yang berbeda, dilanjutkan dengan uji kestabilan fisik meliputi uji pH, organoleptis, moisture content, waktu alir dan sudut diam. Hasil penelitian menunjukkan tidak ada yang signifikan antara hasil uji organoleptis, pH dan waktu melarut pada ke empat formula yaitu masuk dalam kategori memenuhi syarat. Namun pada uji moisture content dan sudut diam ke empat formula masuk dalam kategori tidak memenuhi syarat. Hasil optimum pada pengujian  antioksidan formula effervescent ekstrak kulit buah pulasan ditunjukkan oleh formula IV ditentukan oleh nilai IC50 826,45 (μg/mL). Hasil penelitian ini menunjukkan ekstrak pulasan dapat diformulasikan dalam bentuk effervescent, tetapi pemilihan konsentrasi ekstrak kulit buah pulasan perlu ditinjau ulang agar didapatkan nilai IC50 yang optimum. Kata Kunci: Effervescent, Pulasan, Antioksidan, DPPH Pulasan (Nephelium mutabile Blume) is a plant that generally grows in Kalimantan. This plant is known to have high antioxidant activity, especially in its peels, but the utilization of this activity has not been widely used. This has been proven by the lack of research on the formulation of this fruit into pharmaceutical preparations that are easy and pleasant to use. The aim of this study was to obtain the optimum effervescent formula for the peel extract of pulasan that acceptable and easy to us and also the antioxidant activity of the formula was assessed using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method using a UV-Vis spectrophotometer. The making of the effervescent powder of the fruit peel extract was carried out using four formulas with different extract concentrations, followed by physical stability tests including pH, organoleptic, moisture content, flow time and angle of repose tests. The results showed that there was no significant difference between the results of the organoleptic test, pH and dissolving time in the four formulas, which were included in the eligible category. However, in the test of moisture content and angle of repose the four formulas are in the category of not meeting the requirements. The optimum results in the antioxidant test of the effervescent formula of the outward skin extract were shown by formula IV determined by the IC50 value of 826.45 (μg/mL). The results of this study indicate that the pulasan peels extract can be formulated in the form of effervescent, but the selection of the concentration of the pulasan peels extract needs to be reviewed in order to obtain the optimum IC50 value.

Abstract

Free radicals are molecules that are inconsistent and reactive because they have a portion of the electron that does not have a mate. Free radicals in the body that exceed their capacity will result in oxidative stress. Oxidative Stress can induce damage to various instruments in cells, such as carbohydrates, proteins, DNA, and fat. If oxidational stress is not handled properly, then it will cause degenerative disease. Free radicals can be overcome with antioxidant compounds. Dayak onions (Eleutherine americana (L.) Merr) are suspected to contain flavonoids and phenolic substances that can be antioxidants. The study aims to determine the content of secondary metabolites, total phenolic levels, as well as the flavonoids of the water extract and the ethanol extract of Dayak onions (Eleutherine americana (L.) Merr) obtained from Tenggarong Seberang (L3), East Kalimantan. The method used to determine total phenolic levels using the Folin-Ciocalteu method, the determination of total flavonoid levels using the colorimetric method, as well as phytochemical screening tests using the tube method. Results obtained at the total phenolic content of dayak onions water extract 0.45±0.07 mg GAE/g and dayak onions ethanol extract 1.21±02.02 mg GAE/g, as well as the total level of flavonoids in dayak onions aquatic extracts 1.17±0.66 mg QE/g and dayak onions etanol extraction 1.33±0.41 mg QE/g, from such results indicate higher total levels of phenolics and flavanoids found in dayak onions ethanol extracted.
