Abstract

Diabetes mellitus is a disease characterized by hyperglycemia or an increase in blood glucose levels. The state of hyperglycemia is one of the supporting factors for oral candidiasis infection. The purpose of this study was to determine the correlation of hyperglycemia with the incidence of Candida albicans infection in patients with Diabetes mellitus. this study was conducted on 30 respondents. Blood glucose levels and Candida albicans culture were measured using the germ tube test method. Data analysis to see the correlation using the Chi-Square test obtained Sig. (0.015 0.00.050) and it can be stated there is a correlation between variables. From the results of data analysis, it can be concluded that there is a correlation between hyperglycemia and Candida albicans infection which causes oral candidiasis in DM patients.

Abstract

Many biscuits circulating in the community are not labeled and do not include the value of water and protein content in accordance with SNI. Therefore, it is necessary to determine the water content and protein content in the biscuits. Determination of water content and protein content in biscuits was carried out on 2 biscuit samples taken at the Banjarbaru Market. The water content test on the sample was carried out by the gravimetric method and the protein content test was carried out by the Kjeldahl method. The results obtained were the test of water content of 2 biscuit samples respectively at 4.43% and 3.84%. While the results of the protein content test from 2 samples are 5.31% and 6.89%, respectively. According to SNI 01-2973-2011 the water content limit does not exceed 5% while the protein content limit is a minimum of 5%. The results of 2 samples showed the water content and protein content in accordance with SNI.

Abstract

Green grass jelly leaf (Cyclea barbata Miers) is a plant that has many benefits with the content of green grass jelly leaves (Cyclea barbata Miers) which are carbohydrates, fats, proteins and other compounds such as polyphenols, flavonoids and minerals such as calcium, phosphorus, vitamin A, and vitamin B. This study aims to determine the comparison of the results of the phytochemical screening of infusion and green grass jelly leaf extract (Cyclea barbata Miers). This research is a type of experimental research conducted at the Laboratory of Pharmaceutical Analysis STIKES Muhammadiyah Klaten. The research stages included sample preparation, extraction process, and phytochemical screening. The data analysis of this research is in the form of qualitative, the data is presented in tabular form and then the results are described. The results of phytochemical screening on green grass jelly leaves showed that the maceration and infusion processes of green grass jelly leaves contain flavonoids, alkaloids, saponins, and tannins. The difference is in the color change that occurs from the results of determining the flavonoid compounds, the results show that the color of the maceration extract is brownish yellow, while the color in the infusion is yellow. The conclusion of this study shows that green grass jelly leaf extract contains flavonoids, alkaloids, saponins, and tannins.

Abstract

Ekstrak buah naga merah (Hylocereus polyrhizus) dan ekstrak bawang putih (Allium sativum) diketahui mampu menurunkan kadar kolesterol total darah dalam studi in vivo. Penambatan molekul dapat dilakukan dalam memprediksi senyawa yang bertanggung jawab berdasarkan nilai docking. Tujuan dari penelitian ini adalah mengetahui zat aktif potensial dari H. polyrhizus dan A. sativum yang berperan penting dalam penurunan kadar kolesterol darah melalui jalur inhibisi enzim HMG-CoA reduktase dalam uji in silico. Reseptor yang digunakan adalah enzim HMG-CoA reduktase (PDB ID: 1HW9) dengan ligan natif simvastatin yang dipreparasi menggunakan YASARA. Struktur senyawa aktif sebanyak 19 senyawa dari buah H. polyrhizus 17 dari A. sativum digambar menggunakan MarvinSketch pada pH 7,4 dengan 10 bentuk konformasi. Proses penambatan molekul dilakukan menggunakan program PLANTS dan divisualisasi dengan Discovery Studio Visualizer. Ligan natif menunjukkan nilai RMSD sebesar 1,5265 Å dengan nilai docking -79,1320. Senyawa 4, dan 17 pada buah H. polyrhizus serta senyawa 36 pada A. sativum menunjukkan kedekatan relative dengan ligan natif lebih dari 95%. Senyawa ini diprediksi mampu menurunkan kadar kolesterol total darah dengan penghambatan enzim HMG-CoA reduktase secara penambatan molekul. Kata Kunci: Buah Naga Merah, Bawang Putih, Vitamin E, Antilipidemia, Kolesterol Total  The red dragon fruit extract (Hylocereus polyrhizus) and garlic extract (Allium sativum) are able to decrease the total blood cholesterol levels in in vivo study. The molecular docking can predict the responsible compound based on the docking value. The purpose of this study is to determine the potential active substances of H. polyrhizus and A. sativum which play an important role in reducing blood cholesterol levels through the HMG-CoA reductase enzyme inhibition pathway in the in silico study. The receptor in this study was the enzyme HMG-CoA reductase (PDB ID: 1HW9) with the native ligand simvastatin and prepared using YASARA. The structure of the active compound (19 compounds from H. polyrhizus and 17 compouns from A. sativum) were drawn using MarvinSketch at pH 7.4 with 10 conformations. The molecular docking process was carried out using the PLANTS program and visualized with the Discovery Studio Visualizer. The native ligand showed RMSD value of 1.5265 Å with a docking value of -79.1320. The compounds 4 and 17 in H. polyrhizus fruit and compounds 36 in A. sativum showed relative closeness with native ligands of more than 95% of the docking score. Based on molecular docking, these compounds are to be able to decrease the total blood cholesterol levels by inhibiting the HMG-CoA reductase enzyme.

Abstract

Isolation and identification of compounds and pharmacological activity of the Micromelum minutum grown in some countries has been done, but the Indonesian M. minutum has not been studied, either phytochemically or pharmacologically. This study aimed to determine the cytotoxic activity of Indonesian M. minutum leave extracts toward MCF-7 and 4T1 breast cancer cell lines. The leaves were obtained from M. minutum grow in Bantimurung National Park, Bulusaraung, South Sulawesi, and then were macerated gradually in hexane, ethyl acetate, and methanol. The cytotoxic activity of obtained extracts was determined by MTT assay. The extraction yielded hexane (HEM), ethyl acetate (EEM), and methanol (MEM) extracts of 2.65, 6.12, and 6.49%, respectively. HEM was the most potent extract with IC50 values of 148 and 87 µg/mL on MCF-7 and 4T1 cells, respectively, followed by EEM (185 and 170 µg/mL). MEM possessed a weak potency with an IC50 value of 384 µg/mL on MCF-7 cells and was not toxic toward 4T1 cells. Therefore, HEM is important to be further investigated for its active constituents.

Abstract

Micromelum minutum is used widely in traditional folk medicine. Although this species has been investigated extensively and several bioactive compounds have been isolated, little work has been done on Indonesian M. minutum. This research aimed to study the chemical constituents and biological activities of M. minutum cultivated in Bantimurung Bulusaraung National Park, South Sulawesi, Indonesia. The isolated compounds were assessed for their cytotoxicity towards MCF-7 and 4T1 cell lines by MTT method. The dried ground leaves of M. minutum were sequentially macerated with n-hexane, ethyl acetate, and methanol. The n-hexane and ethyl acetate extracts contained a flavonoid 5,7-dihydroxy-3,4',8-trimethoxyflavone (1) which inhibited MCF-7 and 4T1 cell viability by 50% at concentrations of 369±8 and 227±5 µM, respectively. Further separation of the ethyl acetate extract by column chromatography yielded acetyldihydromicromelin A (2) and a mixture of dihydromicromelin A (3) and dihydromicromelin B (4), which were not active toward MCF-7 and 4T1 cells.

Abstract

Cancer is the highest cause of death in the world. This disease is known to have different regulations from other diseases. The development of drugs today is to target anticancer drugs on cancer regulation sites at Hallmark of Cancer. Lime (Citrus aurantifolia) is one type of Citrus (orange) which contains beneficial chemical compounds and active as anticancer agents. Lime peel is known to have bioactive content in the form of essential oils and flavonoids such as hesperidin and hesperitin. The content of this compound is closely related to anticancer activity. This study aims to identify the content of compounds in lime peel oil and the study of molecular docking silico in these compounds in protein that play a role in the regulation of cancer cells. Lime peel oil is obtained using the steam distillation method then analyzed using GC-MS to determine its constituent components to be analyzed in silico then using YASARA 10.1.8, MarvinSketch 15.4.20, and PLANTS software. The results showed that the main components of lime peel oil were d-limonen and beta-pinen with a percentage of 59.71% and 36.81%. In silico analysis shows that each of these compounds has an affinity greater than the native ligand on caspase-8 proteins (protein regulation of apoptosis). Based on these results it can be concluded, that the lime peel oil has the potential to be developed as an anticancer agent.

Abstract

Ultraviolet rays cause wrinkles and premature ageing. The bad effects of sun exposure can be avoided by using sunscreen. Artocarpus integer is empirically used as a black spot remover and cold powder. Total flavonoid content is an extract quality parameter that is related to the consistency of the pharmacological effects produced. The purpose of this study was to determine the total flavonoid content and the value of Sun Protection Factor (SPF) from the dried extract Artocarpus integer. The study began with a drying of fresh leaves using an oven, making simplicia powder, extraction process by maceration, and drying extracts using a drying cabinet. The dry extract was measured for total flavonoid content using a spectrophotometer UV-Vis with quercetin as a marker. The dry extract also determined the SPF value at concentrations of 200, 400, 600, 800, and 1000 ppm. The results showed that the total flavonoid content of the dry extract of cempedak leaves was 1,283 ± 0.032 %w/w equivalent to quercetin. The SPF value of dry extract of cempedak leaves at concentrations of 200, 400, 600, 800, and 1000 ppm respectively, namely 2,923 ± 0.009, 5.990 ± 0.002, 8.846 ± 0.001, 13,181 ± 0.008 and 15.856 ± 0.1533

Abstract

Hypertension is a disease that often causes death because many people do not realize it. People have hypertension if the blood pressure above 140/90 mmHg after regular checking. In Sungai Ulin Public health center, hypertension is the second-highest disease. The study aimed to determine the characteristics of hypertensive patients and the profile of prescribing the use of anti-hypertensive drugs in outpatients in the period October 2020 at Sungai Ulin Public health center. This research is an observational descriptive study using retrospective data with the inclusion criteria. A population of all prescription anti-hypertensive drugs given to outpatients in the period October 2020 in Sungai Ulin Public health center (87 prescriptions). The results showed the percentage of prescription use of anti-hypertensive drugs, namely women who received more anti-hypertensive prescriptions as many as 59 (65,02%) prescriptions, while the age criteria from > 65 years was (26,43%). The majority of drugs were given in Puskesmas Sungai Ulin was amlodipine with a percentage of 72,41%.

Abstract

Daun Cempedak (Artocarpus integer) merupakan salah satu tumbuhan yang diketahui memiliki kandungan senyawa flavonoid dan berpotensi digunakan sebagai tabir surya. Penelitian ini bertujuan untuk menentukan kadar flavonoid total dan aktivitas tabir surya fraksi ekstrak etanol daun A. integer dengan menggunakan instrumen spektrofotometer UV-Vis. Daun A. integer segar dilakukan proses ekstraksi secara maserasi dengan pelarut etanol 96 %. Ekstrak kering difraksinasi dengan n-heksan dan etil asetat serta fraksi diuapkan hingga kental. Penentuan kadar flavonoid dilakukan dengan metode kolorimetri AlCl3, sedangkan uji Sun Protection Factor (SPF) dilakukan pada konsentrasi 200, 400, 600, 800, dan 1000 ppm menggunakan instrumen spektrofotometer UV-Vis. Hasil menunjukkan kadar flavonoid total fraksi ekstrak etanol daun A.integer yaitu fraksi n-heksan 1,565 ± 0,007 % b/b, fraksi etil asetat 2,443 ± 0,035 % b/b, dan fraksi air 0,560 ± 0,002 % b/b ekivalen kuersetin. Aktivitas tabir surya fraksi ekstrak etanol daun A. integer yang paling baik terdapat pada konsentrasi 1000 ppm yaitu fraksi etil asetat termasuk kategori kuat dengan nilai SPF 8,511; fraksi n-heksan termasuk kategori sedang dengan nilai SPF 4,786; dan  fraksi air termasuk kategori sedang dengan nilai SPF 4,322.
