Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan kuning telur angsa berbagai konsentrasi pada media sitrat terhadap kualitas spermatozoa sapi aceh pasca pembekuan. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 4 kelompok perlakuan pengencer sitrat kuning telur angsa 5% (K1), pengencer sitrat kuning telur angsa 10% (K3), pengencer sitrat kuning telur angsa 15% (K3), dan pengencer sitrat kuning telur angsa 20% (K4). Masing-masing kelompok diulangi sebanyak 5 kali. Sampel semen dikoleksi dari 1 ekor sapi aceh jantan sehat berumur 3 tahun menggunakan vagina buatan satu kali dalam seminggu selama lima minggu. Semen yang berkualitas baik diencerkan dengan media sitrat kuning telur sesuai perlakuan dan dikemas dalam mini straw. Selanjutnya diequilibrasi selama 4 jam pada suhu 50C dalam cool top. Kemudian dilakukan pembekuan di atas uap nitrogen cair selama 12 menit dan disimpan di dalam kontainer berisi nitrogen cair (-196ºC). Setelah penyimpanan selama 1 minggu, masing-masing semen beku perlakuan thawing untuk di evaluasi kualitasnya. Data kualitas spermatozoa dianalisis dengan ANOVA dan dilanjutkan dengan uji Duncan. Rata-rata persentase motilitas, spermatozoa hidup dan membran plasma utuh spermatozoa setelah pembekuan pada kelompok pengencer sitrat kuning telur angsa 15% (K3); lebih tinggi dari pada sitrat kuning telur angsa 5% (K1); 10% (K2) dan 20% (K4). Simpulan penggunaan kuning telur angsa 15% dalam pengencer sitrat dapat mempertahankan motilitas, spermatozoa hidup dan membran plasma utuh spermatozoa sapi aceh setelah pembekuan yang lebih baik dibanding kuning telur angsa 5%, 10% dan 20%. This research aims to know the effect of using goose yolk various concentrations in citrate medium to the quality of post-freeze aceh cattle spermatozoa. This research used complete randomized design (CRD)  with 5 groups of 20% chicken yolk citrate dilution treatment (K0); 5% diluent of goose yolk citrate (K1); 10% diluent of goose yolk citrate (K3); 15% diluent of goose yolk citrate (K4). Each groups is repeated 5 times. Semen samples were collected from one 3-years-old male aceh cattle using artificial vagina once a week for five weeks. The good quality semen is diluted by the yolk citrate appropiate to treatment and packed in a mini straw. Afterwards equilibrated for 4 hours at 5°C in cool top. Then freezing on liquid nitrogen vapor for 12 minutes and stored in containers of liquid nitrogen at a temperature of -196°C. After storaging for 7 days, each frozen semen thawing treatment for the quality evaluation. The spermatozoa quality data were obtained in the ANOVA analysis and obtained by Duncan tests. Average motility percentage, alive spermatozoa and spermatozoa MPU after freezing in 15% diluent of goose yolk (K3); higher than 5% goose yolk (K1); 10% (K2) and 20% (K4) but not different with 20% chicken yolk citrate (K0). In conclusion,  using 15% goose yolk in citrate diluent can maintain mortality, alive spermatozoa and  spermatozoa MPU Aceh cattle after better freezing than 5%, 10% and 20% goose yolk.

Abstract

Effect of Supplementation of ENERVON-C and SANTA-e in Feed on Sperm Motility of Local CocksABSTRACT. This study aimed to observe the effect of supplementation of ENERVON-C and SANTA-e in feed on sperm motility of local cocks. The research was conducted at Reproduction Laboratory, Faculty of Veterinary Medicine, Syiah Kuala University, Banda Aceh. The research used 12 local cocks divided into 4 treatment groups : P0 (feed of 521), P1 (feed of 521 + 200 mg ENERVON-C/kg of feed), P2 (feed of 521 + 200 mg SANTA-e/kg feed) and P3 (feed of 521 + 100 mg ENERVON-C/kg of feed and 100 mg SANTA-e/kg of feed). During the research, all cocks were given 100 g of 521 feed/day. Feed with ENERVON-C and SANTA-e was given for 2 weeks before semen collections. Drink was given ad libitum. Data were analysed with analysis of variance and if there are effects among the treatments, analysis will be continued with Duncan Test. Results shows that supplementation of ENERVON-C or SANTA-e and their combination in feed can increase the percentage of sperm motility. There are significant differences (p0,05) between control and treatment groups. To sum up, supplementation of ENERVON-C or SANTA-e in feed can improve the quality of sperm (motility) of local cocks. The sperm quality is better after giving of ENERVON-C and SANTA-e together than single supplementation.
