Abstract

ABSTRACTThe research of Methanol Extract Bioactivity Rf0.5 of Mangrove Leaf Rhizophora mucronata L. (Rhizophoracea) for Bacteria Resistent Escherichia coli had been done at Marine Chemistry Laboratory, Faculty of Marine and Fisheries and Microbiology Laboratory, Medical Faculty, Syiah Kuala University in January 2018. This study aims to determine the bioactivity of the compound from Rhizophora mucronata leaf extract Rf0.5 against Escherichia coli resistant bacteria. The bioactive compound was isolated based on bioassay-guided separation with thin layer chromatography method. The phytochemical test of Rf0.5 compound indicates active metabolite as the secondary metabolite of alkaloid group in the hydrocarbon system. The result of active metabolite showed diameter of inhibitory zone at  7.10 -8.25 mm as a potential antibiotic toward resisten Escherichia coli. Keywords : R. mucronata leaf, E. coli, alkaloid, inhibit zone. ABSTRAKPenelitian yang berjudul Bioaktivitas Rf0,5 Ekstrak Metanol Daun Mangrove Rhizophora mucronata L. (Rhizophoracea) terhadap Bakteri Escherichia coli telah selesai dilakukan di Laboratorium Kimia Laut, Fakultas Kelautan dan Perikanan dan Laboratorium Mikrobiologi, Fakultas Kedokteran, Universitas Syiah Kuala pada bulan Januari 2018. Penelitian ini bertujuan untuk mengetahui bioaktivitas senyawa dari ekstrak daun Rhizophora mucronata terhadap bakteri resisten Escherichia coli. Senyawa bioaktif diisolasi berdasarkan bioaktivitas melalui tahap kromatografi lapis tipis. Hasil uji fitokimia senyawa Rf0,5 mengindikasikan metabolit aktif memiliki struktur bangun hidrokarbon yang mempunyai gugus alkaloid. Hasil uji bioaktivitas menunjukkan diameter zona hambat pada 7,10-8,25 mm yang menunjukkan bahwa metabolit sekunder Rhizophora mucronata berpotensi sebagai antibiotik terhadap bakteri resisten Escherichia coli.Kata Kunci : Daun R. mucronata, E. coli, alkaloid dan zona hambat.

Abstract

Research on percentage of the coral cover in ulee kareung coastal waters of  subdistrict of simpang mamplam district of bireuen has been done on August, 2016 with the purpose to find out the percentage of coral cover in Ulee Kareung coastal areas. Determinationof location of research using Sistematic Sampling Methods and monitoring the data of coral reef using LIT (Line Intersect Transect) Methods. The results showed that The coral reef condition in ulee kareung beach subdistrict of simpang mamplam district of bireuenhas average value 28,55% in shallow areas and 23,92% in deep areas and there were 19 genus of coral with the highest coral percentage arePorites13.40%.Based on the coral cover percentages in Ulee Kareng beach indicated as medium category.The result of this study are expected to be used as supporting data and used as a reference in consideration of maintainaceulee kareung beach subdistrict of simpang mamplam district of bireuenPenelitian tentang persentase tutupan karang di pantai Ulee Kareng Kecamatan Simpang Mamplam Kabupaten Bireuen telah selesai dilakukan pada bulan agustus 2016 yang bertujuan mengetahui persentase tutupan karang di kawasan Pantai Ulee Kareung. Penentuan lokasi penelitian menggunakan Metode Sistematic Sampling dan memonitoring data terumbu karang menggunakan Metode LIT (Line Intersect Transect). Hasil pengamatan menunjukkan bahwa kondisi rata - rata terumbu karang pada Pantai Ulee Kareung Kecamatan Simpang Mamplam Kabupaten Bireuen dikedalaman dangkal (shallow) sebanyak 28,55% dan kedalaman dalam (deep) sebanyak 23,92% serta terdapat 19 genus karang dengan persentase karang tertinggi yaitu genus Porites (13,40%). Berdasarkan hasil rata – rata persentase tutupan di pantai Ulee Kareung sebanyak 26,23% mengindikasikan bahwa persentase tutupan karang termasuk kategori sedang.Hasil penelitian ini diharapkan dapat dijadikan data pendukung serta digunakan sebagai acuan dalam pertimbangan menjaga kawasan pantai Ulee Kareung, Kecamatan Simpang Mamplam, Kabupaten Bireuen.

Abstract

      The study of the isolation of bioactive compound from Oscillatoria sp. as antibacterial Escherichia coli has been done in December 2017. Bioactive metabolite was isolated based on bioassay-guided separation with several steps of chromatography. The maceration results showed the potency of antibacterial activity of 8.75 mm diameter, while the positive control of chloramphenicol is 8.70 mm diameter. Furthermore, isolation using the partitioning technique showed the potential activity of the K1B15 fraction with a diameter of 8.80 mm, while the positive control was 6.75 mm. The result of thin layer chromatography showed that the active metabolite compound had two different Rf values. Based on the results of photochemical tests indicate that the active metabolite is the alkaloid compound. Bioactivity to Escherichia coli bacteria showed activity at a dose of 100 μg/mL, with the result that Oscillatoria sp. has a potent activity against Mycobacterium Escherichia coli.       Penelitian mengenai isolasi senyawa bioaktif dari Oscillatoria sp. sebagai antibakteriEscherichia coli telah selesai dilakukan pada bulan Desember 2017. Metabolit bioaktif diisolasi berdasarkan bioaktivitas melalui beberapa tahapan kromatografi. Hasil maserasi menunjukkan adanya potensi aktivitas antibakteri berdiameter 8,75 mm, sedangkan kontrol positif chloramphenicol berdiameter 8,70 mm. Isolasi lebih lanjut menggunakan teknik partisi menunjukkan adanya potensi aktivitas dari fraksi K1B15 yang berdiameter 8,80 mm, sedangkan kontrol positif 6,75 mm. Hasil kromatografi lapis tipis menunjukkan senyawa metabolit aktif memiliki dua nilai Rf yang bervariasi. Berdasarkan hasil uji fitokimia mengindikasikan bahwa metabolit aktif termasuk golongan senyawa alkaloid. Bioaktivitas terhadap bakteri Escherichia colimenunjukkan aktivitas pada dosis 100 µg/mL sehingga memiliki potensi sebagai antibakteri Escherichia coli.

Abstract

Research on the distribution and abundance of fish species Apogon Kiensis, from Apogonidae family, was conducted on September 2017 on seagrassAhmad Rhang Manyang bay, Lamreh, Aceh Besar District. Distribution data collection and abundance using systematic sampling method and belt transect technique. Fish datas collect starts fromat 1 to 5 meter with 3 repetitions. The result of observation Apogonidae family fish distribution showed that the index value of morisita ranged from 0.91 - 1 with the random and uniform category. The results of observation of fish family abundance Apogonidae at every depth of 1 meter to 5 meters ranged between 7 ind / 100 m^2 until 16,67 ind / 100 m^2and known relative abundance ranged between 100%, relative frequency and frequency ranged between 100%.  Based on the results of depth comparison, apogonidae fish abundance along with depth increases the amount of fish is also increasing and fish distribution pattern is randomly caused by the environmental condition is homongent, while fish are uniformly influenced by high natural selectivity so that there is a competition that pushes for the same spatial distribution. This spread may be suspected due to food competition.Keywords: Distribution and abundance of Apogon Kiensis fish, Apogonidae family, Ahmad Rhang Manyang Bay.       Penelitian tentang distribusi dan kelimpahan ikan spesies Apogon Kiensis, Famili Apogonidae telah selesai dilakukan pada tanggal 25 September 2017pada kawasan padang lamun di Perairan Teluk Ahmad Rhang Manyang, Lamreh, Kabupaten Aceh Besar. Pengambilan data distribusi dan kelimpahan menggunakan metode sistematik sampling serta tehnik metode belt transect. Pengambilan data ikan dilakukan dimulai pada kedalaman 1 sampai 5 meter serta dilakukan 3 kali pengulangan. Hasil pengamatan distribusi ikan famili Apogonidae menunjukan nilai indeks morisita berkisar antara 0,91 - 1 dengan kategori acak dan seragam. Hasil pengamatan kelimpahan ikan famili Apogonidae pada setiap kedalaman 1 meter sampai 5 meter berkisar antara 7 ind/100m2 sampai 16,67 ind/100 m2dan diketahui kelimpahan relatif berkisar antara 100%, frekuensi dan frekuensi relatif berkisar antara 100%. Berdasarkan hasil perbandingan kedalaman, kelimpahan ikan apogonidae seiring pertambahan kedalaman jumlah ikan semakin meningkat dan pola distibusi ikan secara acak disebabkan terjadi kondisi lingkungan bersifat homongen, sedangkan ikan secara seragam dipengaruhi selektifitas alam yang tinggi sehingga timbul kompetisi yang mendorong untuk pembagian ruang hidup yang samapenyebaran ini dapat diduga karena persaingan makanan.Kata Kunci : Distribusi dan kelimpahan ikan Apogon Kiensis, Famili Apogonidae, Teluk Ahmad Rhang Manyang.

Abstract

ABSTRACTThe research of bioactivity test Rf0,5 methanol extract of mangrove leaf Rhizophora mucronata L. (Rhizohporaceae) for bacteria resistent Escherichia coli was conducted in January 2018. Secondary metabolite isolation was carried out at the Marine Chemical Laboratory, Faculty of Marine and Fisheries, and bioactivity test was conducted at Microbiology Laboratory, Medical Faculty, Syiah Kuala University. The purpose of this research was to study themethanol extract bioactivity Rf0.5 of mangrove leaf Rhizophora mucronata against Escherichia coli. The bioactive compound was isolated based on bioactivity through thin layer chromatography. The results of the phytochemical test was indicated the presence of an active component of the alkaloids group. Abstract

Research on prediction of carbon uptake in mangrove vegetation has been done in mangrove area Gampong Iboih, Sukakarya sub-district, Sabang city. Mangrove forests have a similar role to other forests to absorb carbon dioxide (CO2) so that it can help in the prevention of climate change. The purpose of this study was to estimate carbon uptake (C) in Rhizophora sp. and Bruguiera sp. in Gampong Iboih, Sukakarya sub-district, Sabang city. This research was conducted in June 2017. The method used in this research was purposive sampling method to determine two observation station, the sample was taken once without repetition. The results of this study indicate that mangroves in Rhizophora sp. has a role in carbon uptake in amount 21,344,73 g/trees. The most carbon-absorbing part of the tree is the stem part in amount 9.729 g/tree. While Bruguiera sp. absorbing carbon dioxide of 2,890.12 g/tree. The part of the tree that absorbs the most carbon is the root part of 774.37 g/tree. The larger the biomass, the greater the potential for carbon uptake.       Telah dilakukan penelitian tentang dugaan serapan karbon pada vegetasi mangrove di kawasan mangrove Gampong Iboih, Kecamatan Sukakarya, Kota Sabang. Hutan mangrove memiliki peran yang sama dengan hutan yang lainya untuk menyerap karbon dioksida (CO2) sehingga dapat membantu dalam pencegahan perubahan iklim. Tujuan penelitian ini adalah untuk menduga serapan karbon (C) pada vegetasi mangrove jenis Rhizophora sp. dan Bruguiera sp. di kawasan mangrove Gampong Iboih, Kecamatan Sukakarya, Kota Sabang. Penelitian ini dilakukan pada bulan Juni 2017. Metode yang digunakan dalam penelitian ini adalah Metode Purposive sampling untuk menentukan dua stasiun pengamatan, sampel diambil sekali tanpa pengulangan. Hasil dari penelitian ini menunjukkan bahwa mangrove pada jenis Rhizophora sp. memiliki peran dalam penyerapan karbon sebesar 21.344,73 g/pohon. Bagian pohon yang paling banyak menyerap karbon adalah bagian batang sebesar 9.729 g/pohon. Sedangkan Bruguiera sp. menyerap karbon dioksida sebesar 2.890,12 g/pohon. Bagian pohon yang paling banyak menyerap karbon adalah bagian akar sebesar 774,37 g/pohon. Semakin besar biomassa, maka akan semakin besar pula potensi serapan karbonnya.

Abstract

     The study of chlorophyll-a concentration and phosphate content was conducted in the estuary of Panga river, Aceh Jaya district on August 2017. The objective of this research was to figure out the correlation between these parameters in this area. Samples were collected using the purposive sampling method. The result showed that the chlorophyll-a concentration was positively related to the phosphate content with the value of the correlation coefficient, r= 0,66 and determination coefficient, R2= 0,44. It was obtained that chlorophyll-a concentrations were ranged from 0,9-1,23 mg/L and phosphate contents were ranged from 60-70 μg/L.       Kajian mengenai konsentrasi klorofil-a dan kadar fosfat dilakukan di muara sungai Panga, Kabupaten Aceh Jaya pada Bulan Agustus 2017. Tujuan dari penelitian ini adalah untuk mengetahui hubungan kedua parameter tersebut di daerah ini. Pengambilan sampel dilakukan dengan metode purpossive sampling. Hasil penelitian menunjukkan bahwa konsentrasi klorofil-a berkolerasi positif dengan kadar fosfat, dengan nilai koefisien kolerasi, r= 0,66 dan koefisien determinasi, R2= 0,44. Diperoleh konsentrasi klorofil-a di muara Sungai Panga ini berkisar antara 0,9-1,23 mg/L dan kadar fosfat berkisar antara 60-70 μg/L.

Abstract

The research of structure of the seagrass community at with variation of depth was conducted on September 2017 at Ahmad Rhang Manyang Bay, Aceh Besar District. The aims of study was analyze the structure community with variate depth. This research line transect methods with quadratic frame techniques. The seagrass observations using include species, density, frequency, cover area, dominance index and seagrass roots. The result from Observations showed that there are two species of seagrass at Ahmad Rhang Manyang Bay, Halophila ovalis and Halodule pinifolia. The highest density of seagrass species was found at depth of 4 meters with an average value of 558 roots/m² for Halodule pinifolia species, while for Halophila ovalis species was found at depths of 6 meters to 9 meters with an average value of 252.7 roots/m² . The highest seagrass cover percentages was found in all stations at 4 meters depth with 75% Halodule pinifolia species, while for Halophila ovalis species with 68.68%. Halophila ovalis species have the highest frequency values in all stations, furthermore The dominance index of seagrass species Halophila ovalis and Halodule pinifolia at Ahmad Rhang Manyang Bay is low. Based on observations of high depth factors affect the different types of seagrasses found in each depth.Keyword : Halophila Ovalis, Halodule Pinifolia, Seagrass Cover, Ahmad Rhang Manyang Bay.       Penelitian tentang struktur komunitas padang lamun pada kedalaman yang berbeda telah dilaksanakan pada bulan September 2017 di Teluk Ahmad Rhang Manyang, Kabupaten Aceh Besar, dengan tujuan untuk mengkaji struktur komunitas padang lamun pada kedalaman berbeda. Penelitian ini menggunakan metode transek garis (line transect) dengan teknik bingkai kuadrat. Pengamatan lamun di lapangan meliputi jenis lamun, kerapatan, frekuensi, luas tutupan, indeks dominansi dan tinggi tegakan lamun. Hasil pengamatan menunjukkan bahwa terdapat dua spesies lamun di Teluk Ahmad Rhang Manyang yaitu Halophila ovalis dan Halodule pinifolia. Kerapatan jenis lamun tertinggi untuk spesies Halodule pinifolia terdapat pada kedalaman 4 meter dengan nilai rata-rata 558 tegakan/m2, sedangkan untuk spesies Halophila ovalis terdapat pada kedalaman 6 meter sampai 9 meter dengan nilai rata-rata 252,7 tegakan/m2. Persentase penutupan lamun tertinggi terdapat di semua stasiun pada kedalaman 4 meter dengan persentase spesies Halodule pinifolia 75% dan spesies Halophila ovalis dengan persentase 68,68%. Spesies Halophila ovalis mempunyai nilai frekuensi paling tinggi di semua stasiun, selanjutnya dominansi lamun spesies Halophila ovalis dan Halodule pinifolia di Teluk Ahmad Rhang Manyang tergolong rendah. Berdasarkan hasil pengamatan faktor kedalaman yang tinggi mempengaruhi perbedaan jenis lamun yang ditemukan pada setiap kedalaman.Kata kunci : Halophila ovalis, Halodule Pinifolia, Luas Tutupan Lamun, Teluk Ahmad Rhang Manyang.

Abstract

Analysis of BOD and COD has been done at estuary of Cut River in Alue Naga Village from March to April 2017. The purpose of this research was to analysis the water quality in Krueng Cut estuarial area, which is compared to the Government Regulation No. 82/2001. The method used in this research is field survey by employing purposive sampling method and laboratory analysis to the samples obtained in the field.The BOD, COD, DO, nitrate, nitrate was tested at Laboratory of Chemistry, Faculty of Marine and Fishery of Syiah Kuala University by applyingspectrophotometric method using Filterfotometer PF-11. The results showed that the highest concentration of BOD and COD was at station 3 where the mixing region occurred because the freshwater from river uphill meets with salt-water from the ocean, and the lowest BOD and COD concentration was at station 7 whichis the mouth of the estuary. The results obtained that water quality in Krueng Cut waters based on Government Regulation of water quality no. 82/2001 on the water quality categorized as a class III water quality, which is the water that can be used only for the function of fishery cultivation.Analisa BOD dan COD telah dilakukan di perairan Krueng Cut Desa Alue Naga yang dilakukan pada bulan Maret sampai dengan April 2017. Tujuan dari penelitian ini adalah untuk mengetahui kualitas air di perairan Krueng Cut Desa Alue Nagayang dibandingkan dengan Peraturan Pemerintah (PP) nomor 82 Tahun 2001.Metoda yang dilakukan dalam penelitian ini adalah survey lapangan dengan metode purposive sampling dan analisa laboratorium terhadap sampel yang diperoleh di lapangan untuk uji BOD, COD, DO, nitrat, nitrit di Laboratorium Kimia Laut Fakultas Kelautan dan Perikanan Universitas Syiah Kualadengan metoda Spektofotometri Filterfotometer PF-11. Hasil penelitian menunjukkan bahwa konsentrasi BOD dan COD tertinggi berada pada stasiun 3 yaitu daerah sirkulasi arus (pertemuan antara arus air tawar dan air asin), dan konsentrasi BOD dan COD terendah berada di stasiun 7 yang merupakan daerah mulut estuari. Hasil penelitian diperoleh bahwa kualitas air di perairan Krueng Cut berdasarkanPeraturan Pemerintah No. 82 Tahun 2001 tentang kualitas air baku mutu dikatagorikan sebagai kualitas air kelas III yaitu air yang dapat digunakan hanya untuk fungsi pembudidayaan perikanan.

Abstract

The bioactive compound from the Indonesian Marine SpongeCallyspongia sp.isolated based on bioassay-guided separation with several steps of chromatography has been done. There are 4 major compound known as isocopalane diterpene derivatives together with a plausible new compound, isocopalanol, a polyoxygenated isocopalane diterpene. Isocopalanol has molecular weight [M+H]+ 412.576 m/z indicate that the compound has molecular formula C24H44O5 determined by LCMS-ESI. This compound has bioactivity IC50 = 0.1 μg / mL against human pancreatic cancer cell.Â
