Abstract

The present research aimed to study the characteristics of frozen-thawed semen in beef bulls ex-import in Ungaran, Indonesia. 5 heads Simmental and 5 heads Limousin of 5.5 years old were used in this research. The research was done in Ungaran Artificial Insemination Center, Central Java, Indonesia and Laboratory of Reproduction and Obstetry, Faculty of Veterinary Medicine, Gadjah Mada University, Yogyakarta, Indonesia. Frozen semen was obtained from this AI center. Results of the research indicated that the average individual motility of Simmental and Limousin is 39.00±5.48% and 36.00±2.24%. The average percentage of live spermatozoa is 54.00±10.75% for Simmental and 48.20±9.78% for Limousin while  the average proportion of abnormal spermatozoa is 9.60±3.36% and 6.80±4.15% for both Simmental and Limousin, respectively. On average, Simmental frozen-thawed semen have the higher mean proportion of sperm motility, live sperm, and abnormal sperm than that of Limousin frozen-thawed semen

Abstract

Bovine viral diarrhea virus (BVDV) is a common pathogen that causes major economic losses to dairyindustry worldwide. Amplification of 5’-UTR regions of BVDV genome from blood samples usingreversetranscriptase polymerase chain reaction (RT-PCR) is one of sensitive and accurate techniques forBVDV detection. A common limitation for molecular detection of virus is the ability to obtain high qualityof nucleic acid in the samples. Inappropriate storage conditions will result in a false negative test. FiveEDTA-blood samples collected from BVDV persistently infected cattle were used in the present study.Ten repetitions were done for each sample. The blood were then dripped in the middle part of the FlindersTechnology AssociatesTM (FTA) cards and let it dried for approximately 2 hours. The effectiveness of theFTATMcards for storage and retrieval of BVDV RNA from blood samples molecularly using RT-PCRtechnique were tested and evaluated. The results proved that FTATM cards are effective and safe for thestorage of BVDV genome for a long period at room temperature. Based on the result of the present study,it can be concluded that in veterinary medicine field, the application of FTATM technology needs to bedeveloped to various type of samples and should be used routinely for collecting samples containinginfectious agents.

Abstract

A study was carried out to determine the serum estradiol and progesterone concentration, vulvaappearance, and vaginal cytology during estrous cycle in bligon goat. Four female non pregnant bligongoats (Indonesian indigenous) were used in this research. They were clinically healthy and showed normalestrous cycle. Estrous state was examined by using biological method. When the animals showed acceptanceto be mounted by a buck, they were confirmed as the first day of estrous cycle (estrous phase). Collectionof blood sample, preparation of vaginal smears, and measurement of some physical genital parameterswere conducted by standard veterinary clinical methods at day 11, day between 3-5, day between 6-16, andday between 17-19 of estrous cycle. Enzyme linked immunosorbent assay (ELISA) was used to determineserum progesterone and estradiol concentrations. Mean difference of each parameter was analyzed byanalyses of variance (Anova), followed by Tuckey HSD. The serum estradiol concentration ranged between211.25-247.77 pg/ml. Peak of the estradiol was occurred at the first day of estrous phase, namely 247.77pg/ml and to decrease at day 3-16. The serum progesterone concentration was at the level of 0.21-0.70 ng/ml and showed a significant increase at day 6-16 (p<0.05). The specific physical genital changes (swellingvulva, reddish mucous membrane of vulva, and excretion of viscous transparent vaginal discharge) wereidentified as the animals were at the estrous phase. Composition of exfoliative vaginal cytology did notshow any specific and consistent changes throughout the estrous cycle. Superficial and intermediate cellswere very dominant (>60%) during estrous cycle. In conclusion, serum estradiol, progesterone, the vulvaappearance combined with sexual receptivity could be a good estrous predictor for female cycling bligongoat.

Abstract

The objective of this research was to evaluate metabolism of inorganic phosphat (Pi) and calcium (Ca) in kacang goats subjected to 60-d feed restriction and 60-d full feeding (refeeding). Nine adult male goats were divided into three groups used in this study. Restrictions include 40% (group I), 50% (group II), and 60% (group III) of the ration of dry fodder. To obtain the baseline all animals from all groups were fullfeeding from the start of the study (day 0) to day 60 then followed by restriction stage on day 61 to day 120. The next stage is refeeding from day 121 to 180. Pi and Ca levels were analyzed on days 60, 120, and 180, respectively. The results showed that restriction followed by refeeding decreased Ca levels in all three groups and Pi of animals in groups II and III was below the baseline. The level of Pi reach above the baseline were seen only in animals in group I following restriction and refeeding, although there was a tendency to decrease after refeeding. Based on the results of the study it is concluded that the restriction followed by refeeding decreases the level of Pi and Ca in adult male kacang goats.
