Abstract

Liver disease is a disease caused by various factors that damage the liver, such as viruses, obesity and alcohol consumption. Many studies have been done to find natural remedies for this disease, one of which is Rosella. In this research, the effective dose of ethanol extract of rosella calyx (EERC) was conducted, in high-fat feed (HFF)-induced rats. The aim of this study was to determine the effective dose of EEKR based on parameters:liver histopathological, Alanine Transferase (ALT) and Gamma Glutamyl Transferase (GGT) level. The study was conducted on 30 male Wistar rats, which were divided into five treatment groups. As results, histopathological observations showed a significant decrease in cloudy swelling scores at EERC 200 mg/KgBW/d, decreased steatosis scores at doses of 200, 400, 600 mg; whereas ballooning degeneration, lobular inflammation, and fibrosis scores did not decrease. EERC 200, 400, 600 mg/KgBW/d significantly decreased ALT (p ? 0.01); while doses of 400 and 600 mg reduce GGT. Conclusions, effective dose of EEKR for liver disease based on histopathological features of liver, ALT and GGT in HFF- induced Wistar rats is dose of 200 - 400 mg /KgBW/day. Abstract

Salah satu pilihan pengelolaan obesitas adalah memperlambat absorbsi asam lemak dengan cara menginhibisi enzim lipase dalam saluran pencernaan. Inhibitor lipase pankreas dilaporkan telah berhasil dalam pengelolaan terapi obesitas. Senyawa aktif dalam Jati Belanda dan kedelai dapat berfungsi sebagai inhibitor lipase. Tujuan penelitian ini adalah mengetahui aktifitas lipase dari ekstrak etanol kedelai Detam 1 (EEKD), daun Jati Belanda (EEJB), kombinasi serta hasil fraksionasinya menggunakan metode standar dan modifikasi. Uji aktifitas lipase terhadap EEKD, EEJB, kombinasinya serta fraksi etil asetat, air dan n-heksana menggunakan kit Lipase Liquicolor (HUMAN®) dengan prinsip kolorimetri, menggunakan spektrofotometer 580 nm. Metode modifikasi menggunakan 96 well plate, microplate reader 630 nm. Penelitian ini menyimpulkan bahwa aktifitas inhibitor lipase tertinggi didapatkan pada sampel kombinasi EEKD:EEJB perbandingan 1:2, baik menggunakan metode standar maupun metode modifikasi; fraksi yang mengandung aktifitas inhibitor lipase paling tinggi adalah fraksi air kedelai Detam 1 (KD-1) dan air Jati Belanda (JB) pada 100 ppm, serta aktifitas lipase dalam fraksi air JB lebih tinggi dari fraksi air KD-1.

Abstract

Subchronic toxicity test is performed to evaluate the eff ect and monitor the toxicity eff ects of a compound. The use of combination of ethanol extract of Detam 1 soybean (EEDS) and Jati Belanda (EEJB) as an antiobesity therapy will be consumed for a long time, therefore this test need to be done. The purpose of this research is to know the subchronic toxic eff ect of the test material on Wistar rat hematology. The study was conducted in accordance with the toxicity test stipulated by the Food and Drug Supervisory Agency of Indonesia, on 120 Wistar rats (60 male, 60 females) for 90 days and 120 days (satellite). Rats were divided into 6 groups. Parameters measured were leukocyte, hemoglobin, MCH, MCHC, MCV, hematocrit, RBC, and platelets using the Hematology Analyzer with the principle of colorimetry. The results showed that the eff ect of the test material on all hematologic parameters was not diff erent from the control group (p <0.005), except for the leukocyte, MCH and platelet levels, but the results could be negligible. Conclusion: subcronic toxicity

Abstract

Latar Belakang Biji kedelai Detam 1 dan daun jati Belanda berefek menghambat kenaikan berat badan, akan tetapi dikhawatirkan mempengaruhi organ hepar. Tujuan penelitian Tujuan penelitian untuk mengetahui efek ekstrak etanol biji kedelai Detam 1 (EEKD), ekstrak etanol daun jati Belanda (EEJB) dan kombinasinya terhadap penghambatan kenaikan berat badan dan gambaran histopatologis hepar pada tikus Wistar yang diberi pakan tinggi lemak (PTL). Metode penelitian Penelitian merupakan eksperimental laboratorium dengan rancangan acak lengkap bersifat komparatif. Sebanyak 40 ekor tikus Wistar jantan dibagi secara acak menjadi 8 kelompok perlakuan, masing masing terdiri dari 5 ekor. Selanjutnya diberi perlakuan selama 28 hari, semua kelompok kecuali kelompok kontrol negatif (KN), tetap diberi PTL. Pada hari ke-29, seluruh tikus dikorbankan dan semua hepar tikus, kecuali kelompok Orlistat (K6), dibuat sediaan histopatologis dengan pewarnaan Haematoxylin Eosin (HE). Hasil Penghambatan kenaikan berat badan terjadi pada semua kelompok perlakuan, kelompok K3 (EEKD 10 mg : EEJB 20 mg) menunjukkan penghambatan kenaikan berat badan yang paling baik dan potensinya setara dengan kontrol positip (KP) atau Orlistat. Pada semua kelompok perlakuan (K1, K2, K3, K4 dan K5) terjadi perubahan struktur arsitektur dan inflamasi di daerah portal namun tidak menyebabkan bengkak keruh dan degenerasi lemak. Kesimpulan: Pemberian kombinasi EEKD 10 mg : EEJB 20 mg menunjukkan penghambatan kenaikan berat badan yang paling baik. EEJB sediaan tunggal menyebabkan perubahan gambaran histopatologis hepar paling buruk pada tikus Wistar jantan yang diinduksi pakan tinggi lemak. Abstract

Massive tubule fibrosis is a histopathology hallmark of chronic renal failure (CRF). The previous study of protein hydrolysate of green pea (Pisum sativum) bromelain (PHGPB) showed promising results as an antifibrosis in glucose-induced renal mesangial culture cells, by decreasing their TGF-β levels. In this experiment the anti-inflammatory, anti-fibrotic effect of PHGPB in Cisplatin-induced chronic renal failure (CRF) rats was measured. The purpose of this study is to evaluate the anti-inflammatory and antifibrosis effect of PHGPB in Cisplatin-induced chronic renal failure rats. Five groups consisted of five rats: negative control, Cisplatin control, and three groups of Cisplatin+PHGPB (dose of 100, 200, and 400 mg/kg BW/day) treated for 56 days. The examined parameters are Beta-2-microglobulin (β2MG), hs-CRP, histopathological observations of Masson's trichrome stain, and IHC. The level of β2MG and hs-CRP in the Cisplatin+PHGPB group lowered and was highly different from Cisplatin control (p<0.05) depending on doses. In histopathology, the Cisplatin+PHGPB 400 mg/kg BW group showed less fibrosis and no significant difference with the negative control in Masson trichome staining. While in IHC histopathology, PHGPB treatment slightly ameliorated TGF-β expression and intensity of TGF-β values convincingly. In conclusion, PHGPB can relieve kidney fibrosis in cisplatin-induced nephrotoxicity rats: Emphasis on anti-inflammatory activities.

Abstract

Background: The incidence of chronic kidney disease (CKD) continues to increase from year to year. There was an increase in CKD rates in the 2013 and 2018 basic health research or Riset kesehatan dasar (Riskesdas). Several provinces in Indonesia show a high incidence of CKD and require hemodialysis. As the incidence of CKD increases, it is important to investigate the influencing factors.Objective: To identify the influencing factors of CKD incidence in Indonesia.Methods: Data from 11 provinces with the highest incidence of CKD and hemodialysis were obtained from the Indonesian Ministry of Health through the Riskesdas 2018 survey. For comparison, the incidence of hemodialysis patients in 2020 at Immanuel Hospital Bandung was included in the investigation. All data were analysed using a Python software program, and a decision tree was determined. The results of the decision tree were analysed using Chi-square. Subject profiles were descriptively analysed for some Riskesdas 2018 data and medical records at Immanuel Hospital. Results: The total data from Riskesdas was 130,787 subjects, among those, there were 610 people with CKD/hemodialysis, meanwhile data of 79 people with hemodialysis were obtained from Immanuel Hospital. The odds ratios of diabetes mellitus was 4.54 (p=0.000), hypertension was 3.00 (p=0.000), salty food was 2.26 (p=0.000), waist circumference (WC) was 1.35 (p=0.025), and body mass index (BMI) was 1.06 (p=0.605). Conclusion: Diabetes, hypertension, salty foods, WC and BMI are the five most important factors influencing the incidence of CKD in Indonesia. These variables need to be managed properly to reduce the incidence of CKD.
