Abstract

Post thawed frozen semen viability is one of the most important keys in artificial inseminationdepended on two major cell death mechanism, apoptosis and necrosis. It has been known that good fertilitydiary bull’s seminal plasma contains high concentration of osteopontin (OPN). Osteopontin also known ascell survival protein via inhibition to apoptotic cell death, and B-cell CLL/Lymphoma-2 (Bcl-2)expressionmostly related to the ability of cell survival. The aim of this study was to investigate the influence ofadditional OPN into frozen semen diluter on post thawed sperm Bcl-2 expression. Fresh semen collectedfrom ±4 year Friesian Holstein bull. Treatment group divided into four groups i.e.: control group (P0:without OPN supplementation), (P1: fresh semen with OPN supplementation 5 ?g/5.107 spermatozoa), (P2:fresh semen with OPN supplementation 10 ?g/5.107 spermatozoa), (P3: fresh semen with OPNsupplementation 20 ?g/5.107spermatozoa). Bcl-2 sperm expression was determined usingimunocytochemistry. The result of this study showed that there was no significant difference betweengroup P0 and P1 (p>0,05), but both group P2 and group P3 showed a significant difference with P0 (p<0,05).Nevertheless, there was no significant difference between group P2to group P3 on post thawed FriesianHolstein sperm Bcl-2 expression (p>0,05). The conclusion of this study is osteopontin supplement in frozensemen diluter is capable to increase post thawed Friesian Holstein sperm Bcl-2 expression.

Abstract

eCG (equine Chorionic Gonadotropin) atau PMSG (Pregnant Mare Serum Gonadotropin) merupakan hormon gonadotropin yang beredar di pasaran yang saat ini masih diproduksi menggunakan bahan baku serum Thoroughbred bunting. eCG sangat dibutuhkan untuk mengobati kasus hipofungsi ovarium (inactive ovaries/post partum acyclicity) pada sapi, tetapi harganya mahal dan di Indonesia saat ini sulit memperolehnya. Sudah dibuktikan melalui penelitian sebelumnya bahwa kadar eCG pada kuda CBG4 (cross bred antara kuda Sandel dengan Thoroughbred hingga generasi keempat) tidak berbeda dengan Thoroughbred bunting 3,5 bulan. Serum kuda bunting di samping mengandung eCG, terdapat juga didalamnya E2, P4 (progesteron) dan prolaktin. Sephadex G-75 merupakan produk polimer polidekstran berbentuk gel yang biasa digunakan untuk memisahkan molekul protein dengan kisaran massa molekul antara 10.000–75.000 Dalton (1075 kDa). Tujuan penelitian ini adalah memisahkan P4 dari serum enam ekor CBG4 bunting 3,5 bulan dengan teknik kromatografi filtrasi gel menggunakan sephadex G-75. Model pemisahan dilakukan melalui penampungan sembilan fraksi eluat (fraksi ketiga sampai kesebelas). Selanjutnya, dengan teknik RIA fase padat akan ditentukan kadar P4 tiap-tiap fraksi. Hasil penelitian menunjukkan bahwa kadar P4 (ng/ml) terendah 1,34 ± 1,32 terdapat di dalam fraksi eluat ketiga yang berbeda secara bermakna dengan fraksi eluat kesebelas 18,69 ± 2,99. Di samping itu, terdapat kecenderungan bahwa kadar P4 semakin meningkat seiring dengan semakin mengarahnya eluat ke fraksi akhir. Melalui model pemisahan atas kesembilan fraksi eluat tersebut nantinya digambarkan karakteristik kandungan urutan fraksi eluat untuk setiap komponen protein di dalam serum CBG4 bunting 3,5 bulan. Untuk menunjang maksud tersebut, disarankan untuk meneruskan penelitian terhadap upaya pemisahan eCG dari serum CBG4 bunting 3,5 bulan.

Abstract

Vascular injury caused by Fogarty balloon cathetering lead on endothelial dysfunction and trigger the development of foam cell, early atherosclerosis plaque. L-Arginine, as a nitric oxide precursor, has shown to decrease foam cell development. The aim of this study is to prove the effect of L-Arginine on decreasing the amount of foam cell post surgical embolectomy with Fogarty balloon catheter on iliac arteries of rabbit. Sixteen male rabbits were randomized divided into two group of treatment (n=8). After surgical embolectomy in right iliac artery has done, rabbits were fed standard ration (T0) and standard ration with oral L-Arginine (T1). At 28 days after surgical embolectomy, necropsy was done and foam cells were counted. Vascular injury by Fogarty balloon catheter resulted in the development of foam cells both in T0 (13.38 ±14.64) and T1(1.49 ± 1.20). Unpaired t test result showed not significant different (p0.05). The conclusion of this research is L-Arginine reduces the amount of foam cell post surgical embolectomy with Fogarty balloon catheter on iliac arteries of rabbit, however statistical test did not show any difference.
