Abstract

Penelitian ini dilakukan dengan tujuan untuk isolasi dan karakterisasi streptokokus pada manusia yang diperkirakan mempunyai potensi terinfeksi Streptococcus equi subsp. zooepidemicus di wilayah Bali. Swab tonsil dilakukan terhadap 15 orang yang bekerja di Hutan Wisata Alam (HWA) Alas Kedaton, Bali. Dalam penelitian ini digunakan pula 3 Streptococcus equi subsp. zooepidemicus isolat kera sakit Min 1 isolat babi sakit yang diisolasi pada seat wabah pada tahun 1994 dan 1 isolat babi subklinis yang diisolasi pada tahun 1998. Hasil isolasi swab tonsil dapat diidentifikasi Streptococcus equi subsp. zooepidemicus pada 2 orang pekerja pemandu wisata HWA. Hasil karakterisasi terhadap 7 kultur Streptococcus equi subsp. zooepidemicus isolat manusia, kera dan babi terlihat bahwa 4 kultur (3 kera dan 1 babi) tumbuh pada media padat dengan koloni besar, mukoid dan tampak pertumbuhan yang keruh pada media cair. Tiga kultur yang lain (2 manusia dan 1 babi), tampak koloni kecil, bersifat nonmukoid pada media padat dan tumbuh jernih pada media cair. Berdasar uji hidrofobisitas dan uji hemaglutinasi diketahui bahwa 4 kultur mukoid bersifat hidrofil dan tidak mengaglutinasi eritrosit, sedangkan 3 kultur nonmukoid bersifat hidrofob dan dapat mengaglutinasi eritrosit.

Abstract

Tengger’s horse (Equus caballus) is a local Indonesian horse an originated from its ancestor in Java.As the population of Tengger’s horse is almost extinct it is important to conserve and increase the horsepopulation by in situ or ex situ conservation.The objective of this research was to study the moleculargenetic of partial D-loop of Tengger’s horse. Sequencing of PCR product, showed that the D-loop consistedof 319 nucleotides. The DNA was isolated from whole blood and amplified and sequenced using a publishedprimer sets. The sequence was aligned and compared with horse D-loop sequences available in Genebankusing Clustal W method in MEGA program version 4.0.2. Ten different nucleotide sites were found inTengger horse from (nucleotide no. 9, 52, 64, 69, 102, 117, 133, 170, 187 and 293). The genetic distanceanalised using Kimura 2-parameter model ranged between 0,0% and 3,2%, with the average of 1,7%. Thephylogenetic tree using neighbor joining method based on the sequence of nucleotide partial D-loop couldnot be used to differentiate among horse from Tengger and E. caballus.

Abstract

Covid-19 pandemic is consider as global pandemic since February 2020 by World Health Organization (WHO), it forces education system shifted into online learning. E-learning madrasah as a tool for online teaching and learning process issued by Ministry of Religion Affairs are considered very effective and useful as online learning and teaching during Covid-19 pandemic. This study investigated students’ and teachers’ perceptions, and the effectiveness of e-learning madrasah in English teaching and learning. A mixed qualitative and quantitative methods was used to reveal 362 students’ and 13 teachers’ perceptions and the effectiveness of e-learning madrasah at Islamic state junior high schools (MTSN) in Lamongan regency, East Java, Indonesia. The finding revealed that e-learning madrasah as a tool in teaching and learning process is considered useful during Covid-19 pandemic outbreak, students are motivated to join their English class, and students perceived that this learning tool is ease to utilize as well. In term of teachers’ perceptions indicates the same way as students’. They perceived that this e-learning system as a tool of teaching and learning was very useful and ease to use. Also, they perceived that the system of this e-learning madrasah in term of system was not difficult to be implemented in teaching and learning process.
