Abstract

Thyroxine hormone (T4) was one of the hormones related to metabolism rate. Animal transportation could be stressor interfering metabolism. Sargassum crasifoium was one of brown algae species that contained some important nutrients and also flavonoids. The study aimed at finding out the effect of the Sargassum on the decrase in thyroxine hormone and body weight loss. It used 9 Javanese Randu goats that were classified into 3 treatment groups. Group I served as control, Group II was treated with 450 mg/kg BW of Sargassum estract and Group III was treated with vitamin B1. All of the goats were transported for 12 hours using pickup truck at the speed of 60 km/hour. Blood samples were drawn before the transportation, during loading, 6 hours after the transportation and at the end of the transportation, while the body weight of the goats was weighed before and after the transportation. The results of the study showed that the T4 hormone decreased in the Group I and increased in the Group III during the transportation, while it was stable in the Group II. The biggest percentage of the body weight loss took place to the Group III (0.13%), while the smallest percentage took place to the Group II (0.063%). Statistical analysis showed that the Sargassum extract did not have any significant effect (p>0.05) on both the thyroxine hormone and the body weight loss during the transportation. Based on the results of the study it could be concluded that the Sargassum extract could stabilize the thyroxine hormone during the transportation so that it could prevent the body weight loss

Abstract

The aim of this research was to know the heart activity of rats (Rattus norvegicus) at some environmental temperature. Ten rats (Rattus norvegicus), male, 2 months old, the average weight of 200 gr purchased at experiment Animal Care Unit Gadjah Mada University, Yogyakarta. Allf rats were adapted in a cage with food and water ad libitum. Furthermore, each tail rat was measured heart activity is heart rate frequences (beats per minute), mean blood pressure (mmHg), systolic blood pressure (mmHg) and diastolic blood pressure (mmHg) at the environmental temperature of 30o C, 32o C, 34o C, 36o C, 37o C, 38o C, and 40o C  measured with a blood pressure cuff BP-98A at coccygea vein for 3 minutes at each temperature. The measurement was repeated 3 times. The data of the  heart activities obtained were analyzed by using one way analysis of variance. It was concluded that the environmental temperature of 30o C was optimal to the heart activities of the rats, including  heart rate frequences , blood pressure, systolic and diastolic blood pressures.Keywords:    heart rate frequences, blood pressure, systolic and diastolic, environment temperature, Rattus norvegicus

Abstract

Digestive dysfunction is a common symptom of diabetes. Diabetes can lead to the loss of enteric neurons as well as nerve dysfunction and gastrointestinal complications. There has been no complete report of mienteric nitrergic of small intestines on hyperglycaemia condition, so the purpose of this study was to assess changes of nitrergic neural profile of small intestines on hyperglycemic Wistar strain rats (Rattus norvegicus). Twelve rats were adapted for 3 days, then they were randomly divided into 3 groups namely group I (K-1, as control), group II (K-2), and group III (K-3). Before treatment all rats were fasted for 12 hours, rats K-1 were induced with 1.0 ml of buffer citrate single peritoneal dose; rats K-2 were induced with 40 mg streptozotocin (STZ)/ kg body weight of single peritoneal dose; and rats K-3 were induced with 45 mg STZ /kg body weight of single peritoneal dose. On the 1st and 5th days after STZ induction, blood was collected through the coccigea vein to measure blood glucose levels and weigh the weight. On the day 5th the rats were uthanated with a high dose of ketamine, dissected and taken small intestines (duodenum, jejunum, and ileum) for staining with Nicotinamide Adenine Dinucleotide Phosphate-diaphorase (NADPH-d). Morphometric changes of nitrergic neurons of the small intestine segments were observed. The results showed that on the 5th day increased blood glucose levels of the K-2 and K-3 groups were significantly higher than K-1 (P <0.01). The weight of three groups did not show any significant improvement difference (P> 0.05). The number of neurons in the duodenum, jejunum and ileum of K-2 group increased higher than that of K-1 and K-3 groups. These results indicate that in hyperglycaemic states, the number of nitrergic neurons increases (K-2 group), and there is a significant decrease in the number of neurons (group K-3) compared with K-1 and K-2 groups (P <0.01). The results of this study concluded, that the STZ induction treatment causes hyperglycemia. Hyperglycemia does not affect to the body weight and the number of  nitrergic neurons of small intestines in Wistar strain rats (Rattus norvegicus).

Abstract

Gonadotropine Releasing Hormone (GnRH) is one of the recommended hormones to overcome ovulation problems and it can increase pregnancy rate so that it is used in government programs to increase cattle population in Indonesia, although the results are not yet optimal. The purpose of this study is to compare the composition of bovine GnRH receptor nucleotides with the GenBank reference so that the level of genetic diversity of bovine receptors in Indonesia is known. PCR product sequencing using Promoter F and Exon 1 R primers were further aligned with the reference sequences of Bos Taurus GnRHR mRNA GenBank using the MEGA X program. The results of the analysis found the presence of Single Nucleotide Polymorphism (SNP) in the coding region  of 1st cow  position 38(A> T), 261(C > T), 342(C >T), 411(C >T) and 495(C > T) and 2nd cow positions 261(C >T). Changes in amino acids were also detected in 1st cow position 13 (H> L). The presence of SNP was found to indicate genomic variation between individuals at cattle receptors in Indonesia.

Abstract

Protein is very important for the growth, development and activity in rats. The impact of lack protein intake to gastrointestinal, especially in the small intestine, the number of nitrergic neurons, and changes in body weight in Wistar rats has not been studied. Therefore the aims of this study to examine the effect of a low-protein diet for 4 weeks on the number of duodenal, jejunal, ilealnitrergicneuron and changes in body weight in Wistar rats. Nine male Wistar (Rattusnorvegicus) rats, aged 1 month, with an average body weight of 120 grams used in this study. The rats were adapted for 7 days in individual cages with a normal diet (containing 24% protein) and drinking ad libitum. After being adapted all rats were divided into 3 groups (K-24, K-14, and K-4), consist of 3 rats. Then all rats fasted for 12-hours. Each group received treatment as follows: K-24 group (as a control) was given a diet containing 24% of protein, K-14 group was given a diet containing 14% of protein, and K-4 group was given a diet containing 4% of protein . Diet and drinking water are given ad libitum. Along of treatment for 4 weeks.body weights and blood glucose were measured every weeks, then all rats were euthanized and sample tissues of duodenum, jejunum, and ileum were then were obtained for histopathological preparations using the NADPH-d technique. Data on the number of nitrergicmyenteric neurons in the duodenum, jejunum, and ileum, body weight gain, and blood glucose levels were statistically analyzed by single factor design Anova. The results of this study concluded that a low protein diet (14% and 4% protein) for 4-weeks in the duodenum, jejunum have been increased the number of nitrergic neurons, on the other hand, in the ileum have been decreased the number of nitrergic neurons, reduced body weight, and did not increase blood glucose levels in Wistar rats.

Abstract

  This study examines the prolactin hormone profile in Etaawa, Sapera and Saanen lactating goats, adult females, pregnant does, and yearlings. This study used a total of 80 randomly selected goats with details of 26 Ettawa Crossbreed goats (eight lactating, six adult females, six pregnant, and six heifers), 32 Sapera goats (eight lactating, six adult females, twelve pregnant, and six yearlings) and 22 Saanen goats (five lactating, four adult females, six pregnant and yearling). All goats were declared clinically healthy. Blood sampling was performed through the jugular vein in each animal; then, serum was analyzed with an Enzyme-linked immunosorbent assay (ELISA) to prolactin concentration. High prolactin concentrations were found in Saanen goats in pregnant physiological status compared to Sapera and Ettawa crossbreed goats.

Abstract

Penelitian ini bertujuan memperoleh profil fluktuasi kadar hormon ovari sepanjang siklus estrus. Sampel darah yang berasal dari sepuluh ekor tikus betina diambil selama tiga siklus estrus dan dianalisis menggunakan perangkat inframerah. Kadar progesteron selama tiga siklus memiliki kisaran 6-11 ng/ml pada masa estrus dan meningkat menjadi 32-49 ng/ml pada saat non-estrus.
