Abstract

Boesenbergia flava merupakan tanaman yang berasal dari famili zingiberaceae. Sebagianbesar, ditemukan dalam area sangat lembab, area teduh dan biasanya dekat dengan sungai ataudalam kondisi berawa, yang keberadaannya masih susah ditemukan karena tanaman ini langka,genus dari tumbuh-tumbuhan rhizomatus kecil, sebagian besar tumbuh di hutan. Zingiberaceaemerupakan tanaman herba tropis yang digunakan sebagai obat-obatan, bahan kosmetik maupunbumbu masak. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi BAP dan NAAterhadap pertumbuhan akar Boesenbergia flava secara in vitro. Penelitian ini dilaksanakan diLaboratorium Kultur Jaringan Tanaman, Fakultas Pertanian Universitas Sumatera Utara, Medanyang dimulai pada bulan Maret sampai dengan Mei 2012. Rancangan yang digunakan adalahRancangan Acak Lengkap (RAL) dengan dua faktor perlakuan. Faktor pertama adalah konsentrasiNAA yang terdiri dari 4 taraf yaitu 0 mg/l; 1 mg/l; 2 mg/l; 3 mg/l. Faktor kedua adalah konsentrasiBAP meliputi 0 mg/l; 1.5 mg/l; 3 mg/l; 4.5 mg/l. Hasil penelitian menunjukkan bahwa konsentrasiNAA dan BAP tidak berpengaruh nyata terhadap jumlah akar dan persentase eksplan yang hidup,tetapi interaksi antara NAA dan BAP berpengaruh nyata terhadap jumlah akar. Pada konsentrasiNAA, BAP dan interaksinya berpengaruh nyata terhadap panjang akar.Kata kunci

Abstract

ABSTRACTOne of the problems encountered in the development of the sugar palm seedling there is no fertilizer issuitable for palm nursery. Based on this, the objective of this research was to determine the effect of liquidorganic fertilizer on the growth of sugar palm seedling. The research was conducted at area University ofSumatra Utara, Medan with ± 25 m height above sea level which is held in August to December 2012randomized block design was used. The treatment was a liquid organic fertilizer with some types anddossages of different fertilizers. The treatment was used four replication. The type and dosage of fertilizerwas used in control (without organic liquid fertilizer), organic liquid fertilizer N: <2%, P2O5: <2%, K2O:<2% (50 cc / seeds), organic liquid fertilizer N: <2%, P2O5: <2%, K2O: <2% (75 cc / seeds), organic liquidfertilizer P2O5: <2%, K2O: <2% (4.5 cc / seeds), organic liquid fertilizer P2O5: <2 %, K2O: <2%(7cc/bibit), liquid organic fertilizer N 0.011%, P 6.26 mg/100 ml, 72.13 K mg/100ml, auxin IAA 0.066 g / l,GA3 gibberellins 0.093 g / l (6cc/bibit), liquid organic fertilizer N 0.011%, P 6.26 mg/100 ml, 72.13 Kmg/100ml, auxin IAA 0.066 g / l, GA3 gibberellins 0.093 g / l (8.5 cc / seeds). Parameters measured werepalnt height, leaves number, stem diameter, total leaves area, shoot weight, root weight, shoot dry weight androot dry weight. The results showed that the liquid organic fertilizer had not been significantly affected forall parameters were palnt height, leaves number, stem diameter, total leaves area, shoot weight, root weight,shoot dry weight and root dry weight.Keywords: iquid organic fertilizer, palm

Abstract

Rice is an important commodity in Indonesia. As time went on the narrowing of fertile irrigatedland due to conversion of non-agricultural land for the benefit of making an effort to do that is theuse of improved varieties of upland rice using proper spacing. This study aims to determine themost appropriate spacing and varieties that will enhance the growth and production of uplandrice. The research was carried out on community lahan masyarakat 1 Tanjung Sari, District MedanMedan Selayang Municipal ± 25 m altitude above sea level in the month of June to October2012. This study uses a randomized block design (RBD) with treatment consisting of five varietiesof varieties namely, varieties Situ Bagendit, Situ Patenggang, Towuti, Batutegi and Cirata, andtreatment spacing of three that is 20x20 cm, 20x25 cm and 20x30 cm and the treatment wasrepeated three times.The data obtained were analyzed using analysis of variance, followed byDuncan's Multiple Range Test (DMRT). Results of data analysis showed that the different varietiesof real observations on variables plant height, number of tillers, number of panicles, number ofempty grains, the amount of productive grain, grain weight per sample, 1000 grain weight, and theweight per sample. Spacing is not significantly different on all variables observations. Interactionsbetween plant spacing and different varieties of real variables only on observations of productionper plot.Key words: varieties, spacing, upland rice.

Abstract

The aimed of this reaserch was to determine the respon of growth and production of mustard(Brassica rapa L.) on the application of organic fertilizers. The research was conducted on the farmfield with ± 25 meters above sea level from Mei to June 2013. Randomized Block Design was usedwith 2 factors, ie: varieties (Green Pakchoy and White Pakchoy) and organic fertilizers (0, 200, 300,400, 500 (g/polybag). The parameters observed were plant height, The number of leaf, leaf area, thebiomass weight of plant, the fresh weight of selling and the harvest index.The result of theobservations showed that varieties have significantly affected of plant height and the number ofleaf. while the organik fertilizer give significant effect to all of the parameters observed.Key word : mustard, variety, organic fertilizers

Abstract

The aim of the research was to analyze the genetic diversity of the endemic andaliman inNorth Sumatera based on RAPD markers. A total of 30 accessions andaliman originated fromthe various region in North Sumatera, i.e.: Dairi, Karo Highland, Simalungun. The RAPDanalyse was used with 3 random primer: OPH-06, OPC-12 and OPN-03. These three primersshowed 32 bands, on which 31 were polymorphic (95.23%). Further genetic diversitycoefficient and the filogenetic dendogram were obtained using the Darwin 5.05 software. Theresults showed that 30 accessions of andaliman were clustered in three groups. Each groupconsisted of the accessions of three regions from different altitudes. There were 30 accessionsof three regions studied showed a high genetic diversity.Keywords: Andaliman, genetic diversity, random amplified polymorphic DNA

Abstract

The aims of this research was to evaluate the response of the growth and yield of some mutants (M2) mungbean to water supply of 40% of field capacity. The research was carried out in greenhouse of Agricultural Faculty, University of North Sumatera, Medan, start from December 2013 till March 2014 with eight genotypes, A: is genotype of (F2R0C0), B: is genotype of (M2R1C0),C: is genotype of (M2R1C3), D: is genotype of (M2R2C0), E: is genotype of (M2R2C2), F: is genotype of (M2R3C0), G: is genotype of (M2R3C1), H: is genotype of (M2R3C2), at 40% of Field Capacity by using randomized block design. The treatment was repeated twice. Data were analyzed with ANOVA and continued with LSD. The result of research showed that the mutant genotype B (M2R1C0), E (M2R2C2), and F (M2R3C0) were significantly difference on seed weight per plant, genotype B (M2R1C0) and E (M2R2C2) were significantly difference on 100-seed weight, and genotype E (M2R2C2) was significantly difference on number of pods containingKeywords: Mung bean, mutant (M2), 40% of field capacity.

Abstract

Penelitian ini bertujuan untuk mengetahui keragaan fenotipe M3 kacang hijau pada pemberian air 40% kapasitas lapang. Penelitian ini dilaksanakan di Rumah Kaca Fakultas Pertanian, Universitas Sumatera Utara Medan, yang dimulai sejak Februari 2015 sampai dengan April 2015 dengan perlakuan 4 genotipe yaitu A: merupakan genotipe F3R0C0 (varietas Vima-1 sebagai kontrol), D: merupakan genotipe M3R1C3 (turunan ketiga (M3) varietas Vima-1 pada 10 krad pada 40% Kapasitas Lapang), F:  merupakan genotipe M3R2C1 (turunan ketiga (M3) varietas Vima-1 pada 20 krad pada 80% Kapasitas Lapang), I: merupakan genotipe M3R3C0 (turunan ketiga (M3) varietas Vima-1 pada 30 krad pada 100% Kapasitas Lapang) dengan menggunakan Rancangan Acak Kelompok (RAK). Data yang diperoleh dianalisis dengan sidik ragam dan dilanjutkan dengan Uji Beda Nyata Terkecil (BNT). Parameter yang diamati adalah tinggi tanaman, jumlah cabang produktif, kandungan klorofil daun, luas daun, umur berbunga, umur panen, jumlah polong berisi, bobot biji, bobot 100 biji, volume akar, bobot kering akar, bobot kering tajuk, dan nisbah bobot kering tajuk-akar. Hasil penelitian menunjukkan bahwa genotipe F (M3R2C1) berbeda nyata terhadap parameter tinggi tanaman 2 MST, 3 MST, 4 MST, dan jumlah cabang produktif dan genotipe I (M3R3C0) berbeda nyata terhadap parameter luas daun, volume akar dan umur berbunga sedangkan genotipe A (F3R0C0) berbeda nyata terhadap parameter umur panen.

Abstract

Penelitian ini bertujuan untuk menganalisis keragaman genetik DNA kelapa dengan menggunakan primer spesifik untuk beta karoten. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Tanaman, Fakultas Pertanian, Universitas Sumatera Utara dan Laboratorium Bio Molekuler SSPL (Socfindo Seed Production and Laboratories) Bangun Bandar, Dolok Masihul, Serdang Bedagai-Sumatera Utara, dimulai dari bulan Februari hingga Agustus 2016. Sebanyak 38 sampel stok DNA dari tanaman kelapa sawit yang dikeluarkan oleh PT. Socfin Indonesia diidentifikasi menggunakan marka molekuler primer spesifik Beta untuk karakter beta karoten. Hasil penelitian menunjukkan analisis filogenik dendogram menggunakan DARwin 6.0.13 membagi 38 sampel menjadi tiga kelompok utama.

Abstract

The  aim of the research was to analyze molecular diversity of oil palm MTG variety (Moderate Tahan Gano) with SSR primer. This research was conducted in Plant Tissue Culture Laboratory of  Agriculture, University of  Sumatera Utara and Bio Molecular Laboratory of SSPL (Socfindo Seed Production and Laboratories) Bangun Bandar, Dolok Masihul, Serdang Bedagai-  Sumatera Utara, from February to  September 2016. Individuals were observed as many as 27 samples of oil palm crop varieties MTG issued by PT. Socfin Indonesia using SSR primer. Dendogram formation program conducted with the help of Darwin 6.0.13. The results showed that the percentage of polymorphic 100% and analysis of phylogenetic dendodgram divided into 3 main groups consisting of two sub-groups of 27 samples analyzed.   Keywords: molecular diversity, MTG variety, oil palm, SSR primer.

Abstract

MTG variety is “moderate resistant” oil palm variety to ganoderma disease, these variety is not totally resistant to ganoderma disease, it means can be attacked and has the same morphological with other variety, so need to analyzed with moleculer marker to see genetic differences. The aim of this study is to know the pattern of DNA bands of oil palm based on RAPD marker OPA-13, OPD-13, OPH-13. This study was conducted in Plant Tissue Culture Laboratory, Faculty of Agriculture, Universitas Sumatera Utara, Medan, from March 2017 to February 2018. Analyzed genetic materials were 26 DNA stocks of oil palm of commercial MTG variety from PT. Socfindo and resulted 100% of polymorphic percentage. The size of the DNA that produced a varied range between 424 to 2572 bp. Only 19 DNA samples that could be processed in software from the 26 DNA samples that analyzed, because there some samples that were not able to be amplified, so they did not meet the standardized precentage. Genetic distance measurement and dendrogram formation were analyzed using DARwin 6.0 software. The results of the analysis showed that 19 individuals were divided into three clusters with a total molecular diversity 49.87%. Coefficient of dissimilarity distance was 0 to 0,6.Key words: genetic diversity, RAPD markers, oil palm of MTG variety
