Abstract

The effect of addition cytokinins and modification of sucrose concentration on growth and alkaloid canthinoneproduction in cell suspension cultures of Eurycoma longifilia Jack were studied. The additions of cytokines, BAand kinetin, show effect on the production of biomass and alkaloid in cell suspension of E. longifilia Jack. Theoptimum totals of two-alkaloids were obtained on addition 4.44 μM BAP and without kinetin, respectively. Theaddition of 4.44 μM BA (6-benzyladenine) into TAM medium stimulated increased total of 9-hydroxycanthine-6-one,but decreased total of 9-methoxycanthin-6-one. While the addition of 2.32 - 9.29 μM kinetin (6-furfurylaminopurine)into TAM medium decreased total of two alkaloids (from 0.582 mg to 0.461 - 0.257 mg per 25 ml medium). Whensucrose concentration in TAM medium was increased from 3% to 5%, production of biomass would increase from0.374 g to 0.585 g dry weight per 25 ml medium. While total of two-alkaloids increase from 0.328 mg to 0.441 mgper 25 ml medium when concentration of sucrose in TAM medium was increased from 3% to 4% sucrose.

Abstract

Growth and Production of soybean varieties of Bradyrhizobium inoculation. Demand of soybean as a protein source continues to increase, but the increasing of domestic soybean production is still low so it must import from abroad. This is due to the decline in soil fertility and agricultural land conversion to residential. The solution, soybean planting can be done by optimizing the existing land commonly called agricultural intensification. In this research, agricultural intensification is done by adding Bradyrhizobium as biofertilizer. symbiosis between soybean plants with these bacteria can directly fix the nitrogen (N) from the air and can save in the N fertilizer of inorganic form. The objective of the research was to know the responses of the soybean varieties toward Bradyrhizobium inoculation. The split plot design with three replication was used, Bradyrhizobium as main plot and soybean varieties (Anjasmoro, Burangrang, Mitani, Grobogan, Tanggamus, Detam 1, and Detam 2) as sub plot. The result showed that the Bradyrhizobium were not significant affected on all of parameters. The varieties significantly affected on plant height at 5 weeks after planted, flower initiation, the time of harvest, the seed weights per plant, and 100 seeds weight. Interaction between Bradyrhizobium inoculation and soybean varieties were not significant on all of parameters .   Keyword : growth, soybean, inoculation, bradyrhizobium, variety

This research was conducted at the Sunge Putih Research Centre with altitude ± 80 m above sea level, in August until November 2014. The research was using a block randomized design with two factors and three replications. The first factor was variety IPB 1, Wilis, Anjasmoro and the second factor was Rhizobium (Density 109g or ml/plant) then without Rhizobium. The results showed that the differences in varieties had significantly effect on plant height of age 2-5 (week after plant), age 2-3 (week after plant), summarize of leaf area, mean number of productive branches, root fresh weight, root dry weight, and weight of 100 seeds per plot. Inoculation rhizobium significantly effect on summarize of leaf aera and mean number of productive branches. Abstract

The effect of various macronutrients, micronutrients and sucrose on growth and canthinone alkaloid production in cell suspension cultures of Pasak Bumi (Eurycoma longifolia Jack) was investigated. The optimum macronutrients and micronutrients content for the high alkaloid production of E. longifolia Jack was different to that found in the Murashige and Skoog (MS) medium. The highest amount of alkaloids, 9-hydroxycanthin-6-one and 9-methoxycanthin-6-one, could be obtained from E. longifolia Jack cells cultured in modified MS liquid medium that containing macronutrients: 21.50 mM NH4NO3, 14.25 mM KNO3, 7.50 mM CaCl2•2H2O, 2.50 mM MgSO4•7H2O, 1.45 mM KH2PO4, while content of micronutrients was 0.233 mM FeNa-EDTA, 0.215 mM MnSO4•4H2O and without CuSO4•5H2O. Increased sucrose concentration to 4.00% (w/v) in modified MS liquid medium could increase total of two-alkaloid. The modification of macronutrients and micronutrients concentration based the optimum production of biomass was obtained MSBs medium that producing high biomass but also increasing the production of 9-hydroxycanthin-6-one. The modification of macronutrients or macronutrients and micronutrients based the optimum total of two-alkaloid was obtained MSC and MSD medium that producing low fresh weight but producing the high 9-hydroxycanthin-6-one. Abstract

One of the main obstacles in providing quality potato seed tubers is the long dormancy period of the potato seeds. A study that explores this has been carried out to elucidate the response of dormancy release and initiation of potato sprouts to the differences in tuber weight and storage duration before immersion in cytokinin (6-benzylaminopurine). The study was conducted at the seed warehouse UPT Horticulture Parent Seed Kutagadung Berastagi, Department of Food and Horticulture of North Sumatra Province, from November 2018 to January 2019. The research was carried out with a factorial completely randomized design with two factors, namely (i) tuber weight, consisting of 4 levels [25-35 g, 55-65 g, 85-95g, and 115-125 g]; and (ii) tuber storage duration before immersion in 6-benzylaminopurine (BAP) solution, consisting of 5 levels [control (without immersion), 1, 15, 30, and 45 days after harvest]. Tuber weight, storage duration before immersion, and the interaction of the two factors showed significant effects in dormancy breaking time, germination percentage, germination rate, number of sprouts, and length of sprouts. The number and length of the sprouts increased with the increasing tuber weight and storage duration before immersion in BAP. Keywords: cytokinin, germination, Solanum tuberosum L., tuber seeds

Abstract

To fulfill the demand of soybeans, the expansion of the planting area can be one of the choices through the conversion of marginal land by utilizing suboptimal land, such as saline soil. One of the plant breeding techniques to obtain tolerant plants to salinity is the use of chemical mutations, such as colchicine. The study aimed to reveal the effect of salinity on the characters of soybean plants (mutated with colchicine) and the effect of colchicine on the number of chromosomes and ploidy level. This study was conducted from January to March 2020 and carried out in the plastic house of Faculty of Agriculture, Universitas Sumatera Utara. The experiment was arranged in a Randomized Block Design with salinity levels (0 dS.m-1, 2 dS.m-1, 4 dS.m-1, 6 dS.m-1) as treatments and consisted of ten replications in each treatment. The result showed that the soybean plants were not successfully induced by colchicine at a concentration of 0.04 % with soaking duration of 10 hours, as indicated by the number of chromosomes and analysis of ploidy level. The soybeans that had been induced were planted in saline soil. There were significant effects observed on the productive branch, number of filled pods, and the weight of seeds per plant. Colchicine treatment did not successfully affect the number of chromosomes of soybeans but had an impact on the chromosome pattern. In addition, the salinity treatment of 4 dS.m-1 significantly affected the number of filled pods compared to the treatment of 6 dS.m-1.

Abstract

Tujuan dari penelitian ini yaitu menganalisis pengaruh varietas unggul, kompos, dan dosis pupuk K terhadap pertumbuhan ubi jalar.Penelitian dilaksanakan di Kebun Percobaan Sekolah Tinggi Penyuluhan Pertanian Medan, Sumatera Utara pada bulan Januari 2010 sampai dengan Agustus 2010. Metode penelitian yang digunakan adalah rancangan petak-petak terpisah terdiri atas 3 (tiga) faktor dan 3 (tiga) ulangan. Petak utama adalah varietas ubi jalar (V) terdiri atas 2 taraf, yaitu: Varietas Sari dan Varietas Beta 2. Anak petak adalah Kompos terdiri atas 3 taraf, yaitu :  Tanpa Kompos, kompos jerami 12 ton/ha dan kompos Tandan Kosong Kelapa Sawit (TKKS) 10 ton/ha. Anak-anak petak adalah Dosis Pupuk K terdiri atas 4 taraf, yaitu : 0 kg/ha KCl, 75 kg/ha KCl, 150 kg/ha KCl, dan 225 kg/ha KCl. Hasil penelitian menunjukkan pemberian 225 kg/ha KCl menghasilkan pertumbuhan yang paling baik. Kompos Tandan Kosong Kelapa Sawit (TKKS) dapat meningkatkan kadar C organik, dan Kdd dalam tanah. Kompos ini juga dapat meningkatkan bobot kering dan luas daun, tetapi tidak berbeda nyata dengan kompos jerami. Pertumbuhan, Varietas Sari lebih baik dibandingkan dengan Varietas Beta 2. Pertumbuhan yang terbaik adalah pada interaksi Varietas Sari, kompos Tandan Kosong Kelapa Sawit (TKKS) dan 150 kg/ha KCl. Kata kunci: ubi jalar, kompos, pupuk K

Abstract

Suatu kajian yang bertujuan untuk mengetahui kemampuan beberapa varietas tomat (Lycopersicum esculentum Mill.) untuk beradaptasi pada lingkungan salinitas tinggi. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) faktorial, dengan faktor pertama terdiri dari 3 (tiga) varietas tanaman tomat, yaitu varietas Permata, Monica dan Jelita. Faktor kedua terdiri dari konsentrasi garam NaCl, yaitu 150, 300, 450, 600 dan 750 ppm. Parameter yang diamati adalah tinggi tanaman, jumlah cabang, umur berbunga, jumlah buah per tandan, jumlah  buah per tanaman, produksi per sample, produksi per plot, berat rata-rata buah dan panjang akar. Hasil penelitian menunjukkan bahwa perbedaan varietas berbeda berpengaruh nyata terhadap peubah amatan tinggi tanaman (2, 3, 4, 5 minggu setelah tanam), sedangkan pemberian garam NaCl berpengaruh nyata terhadap peubah amatan tinggi tanaman (4 dan 5 minggu setelah tanam), jumlah buah per tandan, jumlah buah per tanaman, produksi per sampel (g) dan produks per plot (g). Interaksi dari perlakuan varietas dan pemberian garam NaCl berpengaruh tidak nyata terhadap semua peubah amatan yang diamati. Kata kunci: Lycopersicum esculentum Mill., salinitas, varietas.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui pengaruh aplikasi jerami padi dan paket teknologi pemupukan terhadap pertumbuhan dan produksi padi pada pola penanaman intensif. Penelitian ini dilaksanakan pada bulan November 2009 hingga Februari 2010 di Kebun Percobaan BPTP Sumut, Pasar Miring, pada tanah inseptisol. Rancangan penelitian ini menggunakan Rancangan Petak Terpisah dengan tiga ulangan, dengan petak utama terdiri dari aplikasi tanpa jerami padi, tunggul tanpa dekomposer, tunggul + dekomposer, dan tunggul + jerami + dekomposer. Sedangkan subplot adalah paket pemupukan yang terdiri dari pemupukan petani setempat, pemupukan berdasarkan hasil analisis  N, P, K tanah, pemupukan berdasarkan Kepmentan No. 1/2006, pemupukan berdasarkan Bagan Warna Daun untuk N dan penggunaan PUTS untuk P dan K, dan pemupukan berdasarkan simulasi piranti lunak PuPS 1.0. Hasil penelitian menunjukkan bahwa jerami padi dapat meningkatkan produksi padi tetapi tidak berpengaruh terhadap pertumbuhan secara signifikan. Aplikasi dekomposer pada jerami padi memberikan efek yang lebih tinggi dibandingkan tanpa dekomposer. Paket pemupukan berpengaruh terhadap pertumbuhan dan produksi tanaman padi. Paket pemupukan dengan piranti lunak PuPS 1.0 dapat meningkatkan pertumbuhan dan produksi tanaman padi. Interaksi antara kombinasi jerami padi, dekomposer dan paket pemupukan dapat meningkatkan pertumbuhan tetapi tidak memberikan efek secara signifikan terhadap produksi tanaman padi. Abstract

ABSTRACT Response on Growth and Yield Some Varieties of Soybean (Glycine max (L.) Merrill) to Bokashi Fertilizer. Growth and productivity of plant is affected by some factors, among other things are varieties and fertilizing. The research had been conducted at experimental field of Balai Benih Induk Palawija, Tanjung Selamat, District of Deli Serdang (± 57 m asl) in March – June  2012. Experimental was conducted using by Randomize Block Design with double factors consist of four hybrid varieties, that were Anjamoro, Mutiara,  Tanggamus, and Wilis and bokashi fertilizer was consist of three degrees, were without bokashi, 200 g, 400 g, and 600 g, three replications was used to the treatments. Data were analyzed with ANOVA and continued with HSD. The results showed that varieties were significantly to all parameters except the number of empty pods, bokashi fertilizer addition were significantly to plant height at 2 and 6 weeks after planted, flowering time, number of pods contents per plant, dry weight of canopy, and weight seeds per plant. The combination between varieties and bokashi fertilizer were not significantly to whole parameters. High heritability value in growth characteristics (0.72 – 0.94) and productivity characteristics (0.52 – 0.99)  except Mutiara variety in dry weight of root (0.03). Pertumbuhan dan produksi tanaman dipengaruhi oleh beberapa faktor, diantaranya varietas dan pemupukan. Penelitian dilaksanakan di lahan Balai Benih Induk Palawija Tanjung Selamat, Deli Serdang (± 57 m dpl.) pada Maret – Juni 2012. Penelitian ini menggunakan Rancangan Acak Kelompok dengan faktor ganda yaitu 4 varietas, yaitu Anjasmoro, Mutiara, Tanggamus, dan Wilis dan pupuk bokashi 4 taraf, yaitu tanpa pupuk bokashi, 200 g, 400 g, dan 600 g, perlakuan diulang tiga kali. Data yang diperoleh dianalisis dengan menggunakan sidik ragam dan dilanjutkan dengan Uji Beda Nyata Jujur (BNJ). Hasil penelitian menunjukkan bahwa varietas berbeda nyata terhadap semua peubah amatan kecuali jumlah polong hampa per tanaman. Aplikasi pupuk bokashi berbeda nyata terhadap tinggi tanaman 3-6 MST, umur berbunga, jumlah polong berisi, bobot kering tajuk, dan bobot biji per tanaman. Interaksi antara varietas dan pupuk bokashi tidak berpengaruh nyata terhadap semua peubah amatan. Nilai heritabilitas yang tinggi terhadap karakter pertumbuhan (0.72 - 0.94) dan karakter produksi (0.52 – 0.99)  kecuali varietas Mutiara pada parameter bobot kering akar (0.03).
