Abstract

The use of antibiotics as feed additive improves performance in livestock. However, scientific data related to the use of antibiotics in feed merge spreading of bacterial resistance in animal and human bodies, therefore the usage of antibiotics in animal production is restricted. This condition raise the utilization of natural antibiotic as functional feed such as phytogenics (essential oil, flavonoid, saponin, and tannin), enzyme, probiotic, and prebiotic to improve the livestock’s performance, quality, and health. Functional feeds increase profitability in animal husbandry production and its use is feeds are expected to be functional foods that may have positive effects in human nutrition. Abstract

Quorum sensing is known as a communication mechanism among bacteria to control gene expression such as bioluminescence, pigmentation, and pathogenicity. Quorum quenching is known as inhibition of quorum sensing activity. In order to block quorum sensing activity, some bacteria produced enzymes which could degrade AHL, such as AHL-acylase, AHL-lactonase, and AHL-oxidase and reductase. In this study, soil bacteria were isolated and screened for their quorum quenching activity. These isolates were divided into Streptomyces and non-Streptomyces isolates. Detection were done by using Chromobacterium violaceum as an indicator bacteria for pigment inhibition by quorum quenching activity. About 10 out of 695 isolates were shown to possess quorum quenching activity. These isolates were further identified by biochemical tests, Gram staining, and identified by 16S rRNA sequencing. Four positive isolates showed similarity with Streptomyces sp., and 6 positive isolates were non-Streptomyces which showed similarity with Pseudomonas putida, Bacillus pumilus, Bacillus sp., Enterobacter ludwigii, and Enterobacter sp.

Abstract

AbstrakKKeterlibatan biofilm pada infeksi kronis dan pada permukaan peralatan medis selalu menjadi wacana penting bagi kesehatan umum di dunia. Biofilm bakteri berkaitan dengan tingkat resistensi terhadap antibiotik yang menjadikan infeksi sulit untuk diobati. Untuk mengatasi masalah ini, pengendalian yang efektif perlu diimplementasikan, seperti penerapan senyawa antibiofilm. Beberapa tahun terakhir, lingkungan akuatik menjadi salah satu sumber potensi penghasil senyawa bioaktif, termasuk senyawa antibiofilm. Tujuan dari penelitian ini yaitu menapis dan mengkarakterisasi bakteri asal air terjun dan laut yang diperoleh dari beberapa lokasi di Indonesia, sebagai penghasil aktivitas antibiofilm. Isolat dievaluasi berdasarkan kemampuan aktivitas antimikroba terhadap enam bakteri patogen dan diikuti dengan penapisan senyawa antibiofilm. Sebanyak 11 dari 65 isolat menunjukkan aktivitas quorum sensing atau quorum quenching, dan hanya terdapat satu isolat yang memiliki aktivitas keduanya. Supernatan kesebelas isolat menunjukkan penghambatan pembentukan biofilm setidaknya terhadap satu patogen dengan metode uji biofilm statis. Karakterisasi senyawa bioaktif dari lima isolat yang terpilih menunjukkan aktivitas senyawa yang berbeda, seperti karbohidrat, protein, dan asam nukleat. Sekuensing gen penyandi 16S rRNA menetapkan kelima isolat tersebut berada dalam dua genus yang berbeda, Vibrio (WK2.4, WK2.6, and WK2.3) dan Pseudomonas (S1.2 dan S1.3). Penelitian ini memberikan wawasan baru terhadap pencarian kandidat bakteri akuatik sebagai agen antibiofilm yang potensial. Abstract Biofilm involvement in chronic infections and on the surface of medical equipment have been considered as public health concern worldwide. Bacterial biofilm is related to antibiotic resistance, making the diseases difficult to treat. An effective control strategy should be implemented, for example, by applying antibiofilm agents. The use of aquatic environment as potential sources of bioactive compounds, including the antibiofilm compounds, is recently of concern. This study aimed to screen and characterize bacteria with antibiofilm activity that were isolated from waterfall and marine environment and obtained from several locations in Indonesia. The isolates were firstly evaluated for their antimicrobial activity against six bacterial pathogens and followed by antibiofilm screening. Eleven out of 65 isolates showed quorum sensing or quorum quenching activity, and one of them showed both activities. Supernatants of 11 isolates inhibited biofilm formation of at least one pathogen by using static biofilm assay. Bioactive compounds characterization of the selected five isolates revealed the presence of different compounds, such as carbohydrates, proteins, and nucleic acids. The 16S rRNA gene sequencing analysis classified five isolates into two different genera, namely Vibrio (WK2.4, WK2.6, and WK2.3) and Pseudomonas (S1.2 and S1.3). The present study provides insights into the discovery of aquatic bacteria candidates as antibiofilm agents.

Abstract

The use of biocontrol agent in aquaculture is being adapted as an effective alternative to antibiotics which can lead to the elaboration of antibiotic-resistant bacteria and confer unpleasant impacts to aquatic organisms. Aquatic bacteria have been discovered as biocontrol agents and potential probiotic candidates to improve the health of aquatic organisms, feed efficiency, and disease resistance to aquaculture pathogens. However, local isolate has not intensively been explored and used to increase aquaculture sector productivity. Therefore, this research aimed to determine minimum inhibitory concentrations of their antibacterial compounds against aquaculture pathogens and to characterize aquatic bacteria by their viability in the feed. Four isolates from several aquatic environments in Indonesia (Pseudomonas sp. S1.1, Pseudomonas sp. S1.2, Pseudomonas sp. SL1.1, and Bacillus subtilis KM16) were used to characterize of antibacterial compound and to determine the viability in feed. Ethyl acetate extracts from all isolates showed better antibacaterial activity against Aeromonas hydrophila and Vibrio vulnificus than chloroform and dichloromethane extracts, in which ethyl acetate extract from Bacillus subtilis KM16 showed the strongest antibacterial activity. Pseudomonas spp. were more effective against V. vulnificus (40 mg/mL) and Bacillus subtilis KM16 was more effective against A. hydrophila (20 mg/mL), as proved by the minimum inhibitory concentrations of their ethyl acetate extracts. In this research, Bacillus subtilis KM16 had stable viability in feed than Pseudomonas sp. isolates.

Abstract

Bacteria can interact with each or other microorganisms by releasing, sensing, and reacting to small diffusible chemical signal molecules to alter their community behavior. This process, termed quorum sensing, is influenced by density of other bacteria that present in the environment. One example as a result of this process is the formation of biofilm. Biofilm consists of bacterial communities that attach to a surface and envelope themselves in secreted polymers. This formation can be beneficial to pathogenic bacteria because they become highly resistant to antibiotics and human immunity. Thus, antibiofilm agents that can inhibit biofilm formation are needed. The objective of this study were to screen and evaluate bacteria from hot spring and crater lakes that have antibiofilm activity against pathogenic bacteria. In this study, 26 isolates were successfully obtained and tested for quorum sensing and quorum quenching activities. Based on the result, two isolates, which were KM16 and PAP26, were found to have quorum quenching activity. Further research showed that KM16 and PAP26 had antibiofilm activity against more than six pathogenic bacteria. From characterization of the bioactive compounds, it is known that different compound from KM16 and PAP26 have different activity against each pathogen. In molecular identification, isolates KM16 and PAP26 were identified as Bacillus subtilis and Pseudomonas sp. through molecular identification.

Abstract

Aquaculture is currently experiencing massive loss due to the outbreak of pathogenic bacteria. One of the outbreak causes is the development of pathogenic bacterial resistance to the antibacterial. The problem can be solved using microorganisms that can produce new antibacterial compounds. The purpose of this research was to obtain bacteria from fishpond water that could produce antibacterial compounds. About two out of 81 isolates could produce antibacterial compounds. Those two isolates were obtained from saltwater fishponds in North Jakarta (TS2) and Harapan Island (PHY). All fishpond water was grown in marine broth or Luria broth. Extraction of antibacterial compounds was performed using four types of solvents: chloroform, dichloromethane, ethyl acetate, and methanol. Each of the solvents showed a different result. The extraction can only be successfully performed using chloroform and dichloromethane. Extraction using dichloromethane showed a larger inhibitory clear zone than chloroform. Based on 16S rRNA gene sequencing, PHY isolate was identified as Bacillus sp. and TS2 as Acinetobacter sp. In conclusion, isolate TS2 and PHY, which produced antibacterial compounds, showed potential use as aquaculture probiotics.

Abstract

Selai buah pada umumnya memiliki gula tinggi yang dapat meningkatkan resiko penyakit diabetes mellitus. Oleh karena itu dilakukan formulasi selai buah dengan pemanis alternatif stevia. Penelitian ini bertujuan untuk mengetahui masa simpan selai apel rendah gula dengan penambahan stevia, menguji sifat fisikokimia, mengukur kadar gula reduksi, menganalisis penerimaan sensori, serta membuat informasi nilai gizi selai tersebut. Selai dibuat menggunakan buah apel Malang dengan pemanis sebanyak 18% dan substitusi stevia sebanyak 4 dan 8 g. Pengujian masa simpan dilakukan dengan mengamati perubahan sensori dan pH selama penyimpanan. Uji sifat fisikokimia dilakukan dengan pengukuran aw, total padatan terlarut, dan analisis warna, uji gula reduksi dilakukan dengan metode 3,5-dinitrosalicylic acid (DNS), uji penerimaan dan daya oles dilakukan menggunakan dengan uji sensori hedonik dan ranking, serta informasi nilai gizi berdasarkan analisis proksimat. Keseluruhan selai apel yang dihasilkan memiliki pH 3. Selai dengan substitusi stevia menghasilkan selai dengan warna yang lebih cerah dengan masa simpan selama 2 minggu pada suhu ruang. Nilai aw selai dengan stevia diketahui lebih rendah, serta total padatan terlarut yang lebih tinggi dibandingkan dengan selai kontrol. Kadar gula reduksi selai dengan stevia juga diketahui lebih rendah dari selai kontrol. Aftertaste dan daya oles dari selai kontrol lebih disukai dengan perbedaan yang tidak signifikan dibandingkan dengan selai substitusi 4 g stevia. Setiap 20 g selai substitusi 4 g stevia mengandung 8 g karbohidrat, protein dan lemak tidak terdeteksi. Kesimpulannya, substitusi stevia pada selai buah apel dapat dilakukan untuk menghasilkan selai dengan sifat fisikokimia yang lebih baik.Low-Calorie Malang Apple (Malus sylvestris Mill) Jam using Stevia as SweetenerAbstractFruit jam in general has high sugar content that can increase diabetes mellitus risk. Therefore, the formulation of fruit jam with an alternative sweetener such as stevia was carried out. This research aims to determine the shelf-life of low sugar apples jam, physicochemical properties, reducing sugar content, as well as sensory acceptability, and create the nutrition fact the jam. The jams were made from Malang apples using 18% sweetener with 4 and 8 g stevia substitution. The shelf life testing was carried out by observing the changes of sensory properties and pH during storage. The physicochemical properties test was carried out by measuring aw, total dissolved solids, and color analysis. The reducing sugar test was carried out by 3.5-dinitrosalicylic acid (DNS) method, the acceptance test and spreadability were examined using the hedonic and ranking sensory testing, the jam’s nutrition fact was made based on the proximate analysis. The results showed that the apple jams had a pH of 3. Jam with stevia substitution had lighter color with shelf life of 2 weeks at room temperature. Aw value of the jams with stevia was known to be lower and higher total dissolved solids compared to the control jam. Reducing sugar content of jam with stevia also known to be lower than control jam. The aftertaste and spreadability of control jam were preferable with insignificant difference compare to the 4 g stevia jam. Every 20 g of 4 g stevia jam contains 8 g carbohydrates and undetectable protein and fat. As conclusion, the substitution of stevia in jam might produced better physicochemical quality of apple jam.

Abstract

Tempe bungkus daun jati merupakan produk pangan fermentasi tradisional Indonesia yang kaya protein, serat, dan vitamin yang sangat popular di Kabupaten Blora, Jawa Tengah. Penelitian ini bertujuan mengetahui pengaruh penambahan daun kelor pada tahapan perendaman dan peragian dalam proses produksi tempe terhadap profil bakteri asam laktat (BAL) dan tingkat kesukaan. Daun kelor segar dan serbuk disuplementasikan dengan variasi konsentrasi pada proses produksi tempe kacang kedelai. Hasil penelitian menunjukkan bahwa daun kelor tidak menghambat aktivitas pertumbuhan dari kapang Rhizopus. Penambahan serbuk daun kelor sebanyak 1x dan 2x (b/b) dari total jumlah ragi pada proses peragian menghasilkan tekstur tempe yang kompak. Semakin tinggi konsentrasi air perasan daun kelor dalam proses perendaman, jumlah total bakteri asam laktat menurun hingga 103 CFU/g. Profil BAL dari tempe bungkus daun jati dengan ataupun tanpa suplementasi daun jati mengandung BAL dari genus Weissella. Tempe bungkus daun jati dengan suplementasi 1x (b/b) serbuk daun kelor sangat berpotensi untuk disosialisasikan sebagai pangan fungsional kepada masyarakat.Lactic Acid Bacteria Profile and Sensory Evaluation of Teak Leaf-Wrapped Tempeh Supplemented with Moringa LeavesAbstractTempeh wrapped in teak leaves is a traditional Indonesian fermented food product that is rich in protein, fiber, and vitamins which is very popular in Kabupaten Blora, Central Java. This study aimed to investigate the effect of Moringa leaves addition in the soaking and fermentation stages of tempeh production towards the profile of lactic acid bacteria (LAB) and the level of respondent acceptance. Fresh and powdered leaf was supplemented in various doses to soybean tempeh production. As results, Moringa leaves did not inhibit the growth of Rhizopus. The addition of Moringa leaf powder as much as 1x and 2x (w/w) of the total amount of yeast in the fermentation process produced a compact tempeh texture. The higher the concentration of Moringa leaf juice in the soaking process reduced total number of lactic acid bacteria to 103 CFU/g. LAB profile of teak leaf-wrapped tempeh with or without teak leaf powder supplementation contained LAB that belongs to the genus Weissella. Teak leaf-wrapped tempeh supplemented by 1x (w/w) Moringa leaf powder has a potential value to be socialized as functional food to the community.

Abstract

In this paper, we report a suitable method for extracellular synthesis of copper oxide nano particles by using Phormidium cyanobacterium. We hypothesize that synthesis of copper oxide nano particles is believed to occur by extracellular hydrolysis of the cationic copper by certain metal chelating anionic proteins/reductase secreted by bacteria under simple experimental conditions like aerobic environment, neutral pH and room temperature. Proteins not only reduce Cu (II) into copper oxide nano particles (CONPs) but also plays significant role in stabilization of formed nanoparticles at room temperature. Further TEM, SEM, XRD and FTIR analysis have confirmed the synthesis of nano particles through microbial route. Extracellular induction of metal chelating proteins/reductase was analyzed by SDS-PAGE.
