Abstract

Abstrak.Virus Dengue menginfeksi 50 sampai 100 juta orangper tahun, namun terapi yang spesifik atau obat antivirus yang efektif belum ditemukan. Kurkumin diketahui mampu melakukan penghambatan system ubiquitin-proteasome yang menyebabkan penurunan produksi salah satu jenis Flavivirus yaitu Japanese encephaitis. Modifikasi struktur kurkumin terbukti meningkatkan aktivitas biologisnya. Pentagamavunon-0 (PGV-0) diketahui memiliki aktifitas mirip atau bahkan lebih baik dari kurkumin. Tujuan dari penelitian ini adalah mengetahui pengaruh pemberian PGV-0 pada fase awal infeksi virus Dengue-2 (satu hari infeksi). Penelitian ini termasuk penelitian kuasi-eksperimental. Metode yang digunakan untuk deteksi virus Dengue-2 dalam penelitian ini adalah imunositokimia, setelah sebelumnya dilakukan uji sitotoksik PGV-0 terhadap sel vero. Hasil uji sitotoksik menunjukkan konsentrasi yang aman (tidak memberikan efek toksik) dari PGV-0 terhadap sel Vero adalah 4,44 μM. Hasil uji imunositokimia menunjukkan nilai rata-rata sel positif akibat infeksi Dengue-2 satu hari dengan perlakuan PGV-0 jauh lebih kecil (3.8 ± 3.89) dibandingkan kontrol positifnya (14.55 ± 7.25). Disimpulkan bahwa PGV-0 mampu menurunkan positive rate akibat infeksi Den-2 pada periode awal infeksi

Abstract

Curcumin has been known to reduce cholesterol levels in hyperlipidemic state. Faculty of Pharmacy Universitas Gadjah Mada (UGM) synthesized curcumin analogues namely PGV-0 and GVT-0. Like curcumin, both PGV-0 and GVT-0 exhibit anti-inflammatory activities. The aims of this study were to determine the effect and the mechanism of curcumin analogues PGV-0 and GVT-0 on regulation of cholesterol levels in serum. This study was aimed to examine whether PGV-0 and GVT-0 affected cholesterol levels through cholesterol absorption which is regulated by lipase enzyme. To determine the reduction in cholesterol levels, rats were feed with high fat diet (HFD) for 45 days. PGV-0 and GVT-0 were given on day 31-45 at doses of 40 mg/kg bw and 60 mg/kg bw consecutively. Total cholesterol and lipase activity in serum were measured and then statistically analyzed using ANOVA and t-test. The increase of cholesterol levels was markedly reduced by the treatment with both curcumin analogues. Furthermore, lipase activity was clearly inhibited by the treatment with PGV-0 and GVT-0, suggesting that these compounds inhibit cholesterol levels through the reduction of lipase enzyme activity
