Abstract

Data berupa kadar glukosa darah hewan uji versus waktu dianalisis menjadi Area Under Curve (AUC) kadar glukosa versus waktu (0-2 jam).

Abstract

Pacing (Costus speciosus) is an herbaceous plant that is native to Indonesia and it  can be used as a male contraceptive due to spermatogenesis inhibition. The purpose of this study is to find out the composition of optimum Avicel PH 200® as the filler-binder and amylum as the disintegration agent and to find out the variations on physical properties of the powder and tablet. The tablets were made by directly compression in 8 runs based on Simplex Lattice Design (SLD) from Design Expert 7.1.5, in which the test on physical properties of powder includes tapping index, water absorption, and moisture content, as well as on the physical properties of tablet, including hardness, friability, and disintegration time. The results showed that the variation in the composition between Avicel® PH 200 as the filler-binder and amylum as the disintegration agent had a significant effect on the friability of Costus speciosus (CS) extract tablet, in which the combination of both materials can increase the friability of the tablet. The optimum formula of CS tablet had a composition of Avicel® PH 200 by 462,5 mg and amylum by 37,5 mg contained in each tablet.

Abstract

Oxydative stress is known as a major trigger in cardiovascular dysfunction that leads to hypertension, atherosclerosis, and chronic heart failure. Polymethoxyflavone such as tangeretin and nobiletin has been reported to have biological activity including reducing cholesterol level and anti inflammation activity. The critical point that mediates atherosclerosis is the formation of stress oxidative which causes endothelial cells dysfunction. This study was conducted to explore the effect of tangeretin in protecting H2O2-induced oxidative stress on HUVEC. Cell viability was determined by using MTT assay. Cells were treated with different concentrations of tangeretin (12.5; 100; and 200 µM) for 24h before exposed to 600 µM H2O2 for 4h. Cells viability analyses showed that at low concentration (12.5 µM), tangeretin inhibited H2O2-induced cytotoxicity. Furthermore, it induced cells proliferation. However, at high concentration, it failed to protect HUVEC from H2O2-induced cytotoxicity and increased the cytotoxicity of H2O2. Based on this result, tangeretin in low concentration has a potential to reduce oxidative stress on HUVECs.

Abstract

The aim of the study was the determination of glycaemic index (GI) of local tubers such as Uwi (Dioscorea alata), Gadung (Dioscorea hispida), dan Talas (Colocasia esculenta). Tubers were prepared as raw flour which were administered orally to the rats. Blood were taken at 0; 0,5;  1 and 2 hour after tubers administration. The dose was 2,5 kg/kg body weight (BW) whereas glucose was administered as a standart treatment. Area Under Curve (AUC) was calculated after plotting the graph of time versus glucose levels. Glucose levels were determined using GOD-PAP kit. GI values were calculated as a ratio of AUC (0-2hr) of tubers to glucose. Porang was moderate while the other tubers were low GI.

Abstract

Curcumin has been known to reduce cholesterol levels in hyperlipidemic state. Faculty of Pharmacy Universitas Gadjah Mada (UGM) synthesized curcumin analogues namely PGV-0 and GVT-0. Like curcumin, both PGV-0 and GVT-0 exhibit anti-inflammatory activities. The aims of this study were to determine the effect and the mechanism of curcumin analogues PGV-0 and GVT-0 on regulation of cholesterol levels in serum. This study was aimed to examine whether PGV-0 and GVT-0 affected cholesterol levels through cholesterol absorption which is regulated by lipase enzyme. To determine the reduction in cholesterol levels, rats were feed with high fat diet (HFD) for 45 days. PGV-0 and GVT-0 were given on day 31-45 at doses of 40 mg/kg bw and 60 mg/kg bw consecutively. Total cholesterol and lipase activity in serum were measured and then statistically analyzed using ANOVA and t-test. The increase of cholesterol levels was markedly reduced by the treatment with both curcumin analogues. Furthermore, lipase activity was clearly inhibited by the treatment with PGV-0 and GVT-0, suggesting that these compounds inhibit cholesterol levels through the reduction of lipase enzyme activity

Abstract

Chronic inflammation is common in infectious diseases, rheumatoid arthritis, gout, and autoimmune diseases. However, using non-steroidal anti-inflammatory drugs (NSAIDs) is accompanied by dangerous side effects. Therefore, searching for safer alternative therapies without side effects is very important. A natural blend of ingredients produced by stingless bees from plants was potential as a remedy. Meanwhile, the potential of kelulut bee products from East Kalimantan as an anti-inflammatory is still unknown. This study aimed to compare the chemical composition of kelulut bee (Trigona apicalis) products and evaluate the anti-inflammatory effect of honey, propolis, and their combination. Propolis extract and honey were determined as secondary metabolites. An anti-inflammatory in vivo assay triggered the edema using carrageenan on male mice and measured its anti-inflammatory power value. Propolis extract and honey from T. apicalis have a promising anti-inflammatory effect and are significantly higher than the positive control. Meanwhile, combining propolis extract and honey did not enhance the anti-inflammatory effect. In addition, combining honey and propolis preparations with a ratio of 75 : 25 has a better effect on reducing edema volume than the other two combinations. Still, it is not better than the treatment with propolis extract or honey alone. The content of polyphenol compounds found in honey and propolis preparations is thought to have an important role in reducing edema volume.

Abstract

The standardization of raw materials from nature is essential before they are made into products. Until now, there is still limited information regarding the standardization of propolis from stingless bees. This research aims to determine the bioactivity with standardized specific and non-specific parameters of ethanol extracts from Homotrigona apicalis and Homotrigona fimbriata propolis. The methods include organoleptic tests, tests for water-soluble and ethanol-soluble compound content, drying shrinkage tests, specific gravity tests, water content tests, metal contamination tests, phytochemical screening, and determining total phenolic and flavonoid content. The free radical scavenging capacity was measured using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method, and antimicrobial activity against Candida albicans, Propionibacterium acnes, Streptococcus mutans, Salmonella typhi, and Escherichia coli was assessed using the agar well diffusion method. The results indicated that the standardization parameters tested on H. apicalis and H. fimbriata propolis met the standard criteria. Both species contain alkaloids, flavonoids, phenolics, tannins, and saponins. Generally, the propolis extracts from these two stingless bees exhibit potential antioxidant and antimicrobial effects. The total phenolic content for H. apicalis was 0.963 ± 0.041 mg GAE/g, while for H. fimbriata, it was 0.345 ± 0.004 mg GAE/g. The total flavonoid content for H. apicalis was 12.066 ± 0.465 mg QE/g, and for H. fimbriata, it was 9.325 ± 0.031 mg QE/g. All these results can be used as a reference for developing products derived from stingless bees.

Abstract

Background: Yellow wood (Coscinium fenestratum) is one of the typical forest plants of East Kalimantan. The hallmark of this plant is the roots, stems and the fruits pulp have a yellow color. There have been many studies on its use and potential in treating liver disease. However, there is limited research about the use of their fruits peel. Objectives: The purpose of this study was as an initial screening of the antioxidant and antibacterial activity from yellow wood (Coscinium fenestratum) fruits peel. Material and Methods: The yellow wood fruits peel was extracted using methanol solvent to obtain the MeOH extract of their fruits peel. The DPPH test was carried out to determined its antioxidant activity (25,50 and 100 ppm). The agar well difusion method was carried out to determined its antibacterial activity. Phytochemical tests are also carried out to determined the secondary metabolites of that fruits peel. Results: The results of the phytochemical test showed that the fruits peel contained alkaloids, flavonoids, carbohydrates, and tannins, but there were no terpenoids or steroids. The fruits peel has an antioxidant activity (58%) at 100 ppm concentration. While the antibacterial test showed strong inhibition at 100 ppm concentration on S. mutans, S. aureus, P. acne and E. coli bacteria. Conclusions: The fruits peel of the yellow wood has compounds that have strong antibacterial activity and medium antioxidant activity.
