Abstract

Contamination of mycotoxins produced by Aspergillus spp., Fusarium spp . and Penicillium s pp . i n agricultural products has been a concern regarding their effect to health and economic impact. Integrated control system should be based on Hazard Analysis and Critical Control Point (HACCP) approach involving Good Agricultural Practices (GAP) and Good Manufacturing Practices (GMP) . Prevention should be carried out through pre harvest, harvest, post harvest, as well as control quality at all stages of production. Pre harvest control is conducted through the selection of resistant varieties, insect control and weeds management mechanically or applying fungicides and herbicides, plant rotation, irrigation and soil management, as well as biological control . Harvesting at the right time using clean equipments from fungal contamination and insect infestation avoids the contamination of mycotoxins . Post harvest control by physical selection, washing and dilution, drying, storage, application of chemicals and binding agents, natural products, nutrients and vitamins, microbiological control, heating and radiation could also minimize mycotoxin in food and feed . Although chemicals can effectively reduce mycotoxin, the use of those on food/feed should be considered the safety . The addition of natural products, nutrition supplements and vitamins suppress the negative effect of mycotoxin on animals . The use of non-toxigenic fungi and other microbes as biological control is the effective and safe methods for food/feed . The implementation of integrated mycotoxin control system by utilizing the HACCP concept would meet the qualified and safe food/feed products .   Key words: Control, mycotoxins, HACCP

Abstract

Aflatoxicosis is a disease generated as the consequence of aflatoxin contamination in foods and feeds. Garlic (Allium sativum Linn.), a natural spice known to reduce the toxicity of aflatoxins in poultry. Twenty five laying hens were used to study the effectiveness of garlic extract in reducing aflatoxicosis. The animals were divided into 5 groups i.e (1) control group, (2) group treated with 0.4 mg/kg BH, (3) group treated with 0.4 mg/kg BH and 4% garlic extract in feed, (4) group treated with 5 mg/kg BH, and (5) group treated with 5.0 mg/kg BH and 4% garlic extract in feed. Body weight gains, eggs production, and the activity of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), and γ-glutamytransferase (γ- GT) enzymes were observed every week. The residue levels of the aflatoxin and metabolites were measured in the eggs using high performance liquid chromatography (HPLC). The results indicated that group treated with 0.4 mg AFB1/kg BH dan 5.0 mg AFB1/kg BH resulted in decreasing body weight gains, egg production, and increasing the level of GOT, GPT and γ-GT. The addition of 4% garlic extract in the feed was effective to improve the body weight gain and egg production only in the group of chicken treated with 0.4 mg AFB1/kg BH. However, it decreased the enzymes activities of the GOT, GPT, and γ-GT, as well as reduced the aflatoxin residues and metabolites in the groups at both aflatoxin levels (0.4 and 5.0 mg/kg BH). Aflatoxin residues decreased up to 42.2% for the group treated with the low dose of AFB1 (0.4 mg/kg) and 49.0% for the group treated with the high dose of AFB1 (5 mg/kg).   Key words: Garlic, aflatoxicosis, laying hens, aflatoxin residues

Abstract

Indonesia as a tropical country that very conducive for the growth of mycotoxigenic fungi. The climate in Indonesia as a tropical country is very condusive for the growth of mycotoxins producing fungi. Mycotoxins have properties as carcinogenic, mutagenic, teratogenic, estrogenic, neurotoxic and immunotoxic. Mycotoxins reduce performance, appetite, weight, and immunity. They also cause reproductive disorders and generate the residues in animal products that affect human health. These can be prevented by controlling mycotoxins contamination in agricultural products that used for feed ingredients through good management practices (during planting, harvesting, and storage). Mycotoxins contamination can also be minimized by physical, chemical and biological treatments as well as the application of mycotoxin binders. This review describes the use of mycotoxin binders in animal feed. They are used as feed additives, may be derived from organic, inorganic materials or their combination. Combination of organic and inorganic substances proven to be more effective and efficient in controlling mycotoxin contamination. Therefore, it is recommended to use mycotoxin binders to prevent animal health disorder and to decrease mycotoxin residues in animal products.

Abstract

Abstract    The quality of Indonesiam coffee andcocoa products has been declined due to the contamination of fungi producing ochratoxin, a serious human mycotoxin. Therefore, develop-ment of fast, accurate and simple method for early detection of ochratoxin contamination is required. As a part of research attempted to develop early detection technique of ochratoxin in severall commodities, especially agricultural products, the objective of this study was to produce immunochemical reagent for ochratoxin detectionusing immunoglobulin Y (IgY) based on immunochromatographic method. Results showed that OTA-OVA could be synthesized using active ester method with addition of N-hydroxy-succiimide and dicyclocarboimide. The inter-mediate compound produced showed C=O stretching vibrational band at 1600 cm-1 and C-O stretching vibrational band at 1300-1000 cm-1. Antibody-gold nanoparticle conjugate was optimally produced at pH 9 and with antibody dilution of 1:7.5 (v/v). There was 50 nm absorb-tion shift in visible absorbtion after the antibody was conjugated with gold nanoparticle. Even though the test strip did not show  clear visual-ization, the cut off of ochratoxin concentrationis obviously determined at 10 ppb. This results suggest thatthe technique could be used to detect ochratoxin contamination. Abstrak Komoditas kopi dan kakao Indonesia ter-kendala masalah mutu produk yang rendah akibat kontaminasi jamur yang menghasilkan okra-toksin. Okratoksin merupakan mikotoksin yang membahayakan kesehatan manusia. Oleh karena itu, cara deteksi dini kontaminasi okratoksin yang cepat, akurat dan mudah perlu dikembangkan. Sebagai bagian dari usaha untuk mengembangkan teknik deteksi dini okratoksin pada pada berbagai komoditas, khususnya produk pertanian, pene-litian ini bertujuan menghasilkan reagen imuno-kimia yang menggunakan antibodi immune-globulin Y (IgY) berdasarkan metode imunokro-matografik untuk deteksi okratoksin. Hasil penelitian menunjukkan bahwa OTA-OVA dapat disintesis dengan metode ester aktif dengan menambahkan N-hidroksisuksiimida dan disiklo-karboimida. Senyawa antara yang dihasilkan memiliki absorpsi pada frekuensi 1600 cm-1 yang menunjukkan adanya vibrasi ulur ikatan C=O dan adanya banyak absorpsi pada 1300-1000 cm-1 yang mengindikasikan adanya serapan ulur yang kuat ikatan C-O. Konjugat antibodi-nanopartikel emas dihasilkan optimum pada pH 9 dan dengan pengenceran antibodi 1:7,5 (v/v). Hasil pengujian spektrofotometer visible menunjukkan adanya pergeseran serapan sebesar 50 nm setelah anti-bodi dikonjugasikan pada nanopartikel emas. Meskipun secara visual tidak begitu jelas, tetapi hasil pengujian pada teststrip menunjukkan bahwa nilai cut off konsentrasi okratoksin yang terdeteksi adalah10 ppb. Hasil ini menunjukkan bahwa teknik yang dikembangkan dapat diguna-kan untuk deteksi kontaminasi okratoksin.

Abstract

Polyclonal antibodies of aflatoxin B1 were successfully produced from New Zealand White female rabbits after immunization by the hapten of aflatoxin B1-carboxymethyl hydroxylamine hemihydrochloride (AFB1-CMO) conjugated with bovine serum albumin (BSA) as the antigen. The hapten was synthesized using the carbodiimide method with CMO as a linker. Absorption peaks at 362, 264, and 218 nm were observed as a result of characterization with UV-Vis spectroscopy, while IR spectroscopy showed peaks at 3448 cm-1 and 1642 cm-1 attributable to the hydroxyl and nitrile groups, respectively. Furthermore, mass spectrometry showed fragmentation at the m/z of 386, 368.2, and 310, which confirms that the hapten of AFB1-CMO was successfully synthesized. The hapten was then conjugated with BSA to serve as an antigen of AFB1 when it was injected into the rabbits. The specificity of the antigen towards its antibody and the confirmation of hapten-BSA conjugation were characterized using the dot blot immunoassay, which showed a BSA concentration of 1.74 mg/mL. Two weeks after the primary immunization by its antigen, agar gel precipitation testing showed that the rabbit blood serum had positive results for polyclonal antibodiest against AFB1 with the highest concentration of antibodiest of 2.19 mg/mL.
