Abstract

ABSTRACT   The aim of this study was to determine the genetic diversity of potato Supejohn transgenic that had been twenty-five times subcultured with exploration  method. The genetic variability was calculated using molecular marker Polimorphic Random Amplified DNA (RAPD) analysis of ten samples from four weeks planlets of Supejohn transgenic plants using ten random primers. DNA isolation of 10 samples using CTAB buffer  then measurement of the DNA qualification and quantification,  afterwards amplification of DNA by PCR using 10 random primers followed by electrophoresis on a 1% agarose gel with TAE 1x electrode buffer solution. Visualisation  the results used UV Transluminator to see DNA bands and the data of  polymorphic bands had been analized used the NTSYS-pc version of 1.07 program to obtain the similarity coefficient  and the dendrogram.The results showed that five random primers produced polymorphic DNA bands with 0.39 - 0.88 similarity coefficient  and the average similarity coefficient is 0.63 (63%) or the genetic diversity of the samples as many as 37%. Dendrogram formed eight distinct clusters corresponding similarity coefficient, the formation of clusters means that there is genetic diversity among the DNA samples. Keywords: potato Supejohn transgenic, RAPD, genetic diversity ABSTRAK Penelitian ini bertujuan untuk  menentukan keragaman genetik kentang Supejohn transgenik yang telah dua puluh lima kali disubkultur dengan menggunakan metode penelitian eksplorasi dengan penanda molekuler  Random Amplified Polimorphic DNA (RAPD) terhadap 10 sampel tanaman Supejohn transgenik, berumur empat minggu dan  menggunakan  10 primer random.  Isolasi DNA dari 10 sampel menggunakan buffer CTAB sesudah itu dilakukan pengukuran kualitas dan kuantitas  DNA, kemudian dilanjutkan dengan amplifikasi DNA secara PCR menggunakan 10 primer random diikuti dengan  elektroforesis pada gel agarose 1% dengan larutan penyanggah elektroda TAE 1x. Visualisasi hasil elektroforesis  menggunakan UV Transluminator untuk melihat pita DNA  dan analisis  data pita polimorfik  menggunakan program NTSYS-pc versi 1.07 sehingga didapatkan koefisien kesamaan dan dendrogram. Hasil penelitian menunjukkan bahwa lima primer random menghasilkan pita DNA polimorfik dengan koefisien kesamaan 0.39 – 0.88 dan rata-rata koefisien kesamaan yaitu 0.63 (63%) atau  keragaman  genetik sampel sebesar 37%.  Dendrogram  membentuk delapan cluster yang berbeda sesuai koefisien kesamaan,terbentuknya clusters mengartikan bahwa ada keragaman genetik antar sampel DNA. Abstract

Penggunaan NAA (Napthalene Acetic Acid) dan Kinetin (6-furfurylaminopurine) pada Induksi Tunas Kubis Bunga Putih (Brassicaoleraceae L. var. Botrytis) secara in vitro. Penelitian inidilaksanakan di Laboratorium Bioteknologi Fakultas Pertanian Universitas SamRatulangi Manado, pada bulan Maret – Oktober 2018. Rancangan penelitianyaitu Percobaan Faktorial dalam Rancangan Acak Lengkap (RAL) yang terdiridari 16 kombinasi perlakuan dan diulang sebanyak 5 kali. Hasil analisis ragammemperlihatkan bahwa perlakuan kinetin 0 ppm menghasilkan tinggi tanamanterbesar, yaitu 5,31cm pada umur 4 minggu setelah kultur (MSK). Perlakuan NAA0 ppm menghasilkan jumlah daun terbanyak pada umur 2 MSK (2,2) danterbanyak pada umur 3 MSK (3,10). Perlakuan kinetin 0 ppm menghasilkanjumlah daun terbanyak pada umur 4 MSK (3,75). Perlakuan kinetin 3 ppmmenghasilkan jumlah tunas terbanyak pada umur 4 MSK (4,95). Jumlah tunasterbanyak pada perlakuan kombinasi NAA 0,3 ppm dan kinetin 2 ppm adalah 1,8pada umur 2 MSK.Kata Kunci : kubis, induksi tunas, NAA dan Kinetin, in vitroAbstractThe aim of this study was to determine the optimum concentration of NAAand kinetin for in vitro shoot induction of white cabbage (Brassica oleraceae L.var. Botrytis). This research was carried out in the Biotechnology Laboratory,Agricultural Faculty, Sam Ratulangi University, Manado in March - October 2018.Experimental design in this study was a Factorial Design in CompletelyRandomized Design that consisted of 16 treatment combinations with 5replication. The results of ANOVA showed that 0 ppm kinetin resulted in thelargest plant height, i.e 5.31cm at 4 weeks after culture. The highest leaf numberswere observed at 0 ppm NAA, i.e 2.2 at 2 weeks after culture and 3.10 at 3weeks after culture. The highest leaf numbers was also observed at 0 ppmkinetin, i.e 3.75 at 4 weeks after culture. The treatment of 3 ppm kinetin resultedin the highest shoot number (4.95) at 4 weeks after culture. The highest shootnumber at combination of 0.3 ppm NAA and 2 ppm kinetin was 1.8 at 2 4 weeksafter culture.Keywords: Cabbage, Shoot Induction, NAA and Kinetin, in vitro

Abstract

It has been done analysis for the total  phenolic, flavonoid  and tannin compounds at eceng gondok leafs (Eichornia crassipessolms). The research was done in several stages: sample preparation, maceration extraction, evaporation. The total phenolic content analysis by Folin Ciocalteu 50%, flavonoid by chloride aluminium 2% in ethanol, and tanin by vanillin 4% solution in methanol, continued by using ultra violet visible (UV-Vis) spectrophotometry methods. The results showed that the total phenolic content of eceng gondok with methanol 60% of 2.72mg galic acid/kg sample is the high total phenolic content and the lowest phenolic total  at aquades of 1.07mg galic acid/kg and the flavonoid with methanol 80% solution of 3.29mg kuersetin/kg is the high flavonoid content, and the lowest at the aquades of 1.65mg kuersetin/kg and the high condensed tannin content with methanol 80% of 0.7mg katekin /kg. The Fe2+ content photoreduction process, with methanol 60% solution showed the Fe2+  production increase of 17.67ppm, aquades of 2.94ppm. Extract that does not contain eritosin as sensitizer only produce Fe2+ of 5.6ppm and that is not illuminated only 0.06ppm   Keywords: phytochemical, agricultural waste, eceng gondok, biosentizer, ferric photoreduction mitratani
