Abstract

The availability and sustainability of the algae-based biodiesel industry depend on the adequate selection of microalgae strains. Based on this, it is important to explore local microalgae from Indonesia as a feedstock for biodiesel. This study aimed to isolate and screen freshwater microalgae with lipid-producing abilities. A total of three microalgal isolates have been successfully isolated from the Cengklik Reservoir (-7.506712, 110.723582). Based on the cell morphology, the three isolates were identified as Chlorella sp., Scenedesmus sp., and Oscillatoria sp. Nile red staining indicated that the three microalgal isolates contained neutral lipid droplets in their cells, as shown by the presence of a yellow or orange luminescence under a fluorescence microscope. The results of this study indicated that Scenedesmus sp. and Oscillatoria sp. had more potential as feedstocks for biofuels due to their higher lipid content and productivity. This research succeeded in obtaining microalgae isolates from Cengklik reservoir, Boyolali, Central Java which can be used as a source of lipids for biodiesel.

Abstract

Plankton and benthos are two communities of organisms that can be used as bioindicator of aquatic environment. The purpose of this research were to determine kinds, density and diversity of plankton and benthos in Pepe river Surakarta as bioindicator.Study site was devided into 3 station as sampling location. Data sampling of identifying the type, density and calculating the value of diversity index (ID) according to Shannon Wiener for-mula. Investigations carried out measurements of environmental parameters, namely tempera-ture, pH and Dissolved OxygenThe result showed that 1) identified 20 kinds of plankton in Pepe river Surakarta namely Spirogy- ra, Eustbidentat, Pleurosigma, Oscilatoria, Euglena, Aungilospora, Gonatozygon, Dendrospora, Amoeba, Blepharisma sp, Hapalosiphon, Skeletonema, Synura, Stentor, Worochinia, Leptomitus, Peridinium, Paramecium, Volvox, Rhizosolenia, and Lyngbia with the composition and density vary between each sampling site, 2) Identified 3 kinds of benthos in Pepe river Surakarta namely Melanoides sp, Lymnaea sp and Fimbria fimbriata with the composition and density vary between each sampling site and 3 ) the Shannon Wiener Diversity Index (ID) of plankton and benthos in Pepe river Surakarta ranged from 0.98 to 1.98, showing the condition of polluted waters from mild to severe, varied between sampling sites.

Abstract

Keju merupakan salah satu produk olahan susu dengan nilai gizi yang lengkap serta memiliki cita rasa yang khas, sehingga digemari oleh masyarakat. Pada proses pembuatannya, keju menghasilkan limbah cair (whey) saat protein keju diendapkan dan dipadatkan.  Whey yang dibuang begitu saja tanpa diolah akan menimbulkan dampak bagi lingkungan dan dapat menimbulkan protes masyarakat sekitar. Di sisi lain, limbah cair keju masih memiliki kandungan nutrisi yang cukup untuk menjadi bahan baku pembuatan produk fermentasi seperti minuman atau produk lainnya yang memiliki manfaat bagi kesehatan. Minuman sinbiotik berisi probiotik, bakteri yang baik, serta prebiotik, yang merupakan nutrisi selektif untuk bakteri baik.  Pada jangka panjang, pengolahan lebih lanjut limbah cair keju ini dapat memberikan tambahan penghasilan bagi pemilik usaha melalui diversifikasi produk, serta berkontribusi untuk memenuhi kebutuhan masyarakat terhadap bahan pangan fungsional, juga meningkatkan perekonomian di lingkungan usaha mitra. Tujuan kegiatan ini adalah memberikan solusi pengelolaan limbah produksi keju di PT. Mazaraat Lokanatura Indonesia menjadi produk berupa minuman sinbiotik dengan menerapkan hasil riset di Grup Riset Biomateri Mikroba.  Selain itu, kegiatan ini juga akan ditautkan dengan program Merdeka Belajar Kampus Merdeka (MBKM) di prodi Biologi FMIPA UNS.   Luaran kegiatan ini berupa ide pemanfaatan whey serta penerapan teknologi pembuatan produk dari whey yang bisa dipasarkan untuk  mengurangi dampak limbah cair, serta rancangan kegiatan MBKM berupa magang industri di institusi mitra yang dapat direkognisi sampai 21 SKS untuk mahasiswa. Dari kegiatan ini dihasilkan produk minuman sinbiotik menggunakan bakteri probiotik Lactobacillus acidophilus serta prebiotik biji nangka, dengan penambahan ekstrak kunyit dan ekstrak kulit naga sebagai pewarna dan antioksidan.  Produk sinbiotik ini juga dijadikan sebagai bahan pembuatan produk perawatan diri berupa sabun serta lulur.

Abstract

Food stocks, such as grains, fruits, and vegetables are vulnerable to spoilage by Aspergillus flavus, which typically controlled using chemical preservatives. Concerns about the health impact of these chemicals highlight the need for safer alternatives, such as biopreservatives. Lactic acid bacteria (LAB) are Generally Recognized as Safe (GRAS) microorganisms and have potential application as biopreservative agent, because LAB produced various metabolites with antifungal activities. LAB could be found in nutrient-rich environments, including rhizosphere soil and poultry house soil due to the nutrient residues for growth. The aims of this study were to isolate LAB from soils and to evaluate their antifungal activity with a focus on potential applications as biopreservatives. The LAB isolates from soil were characterized and screened for antifungal activity using the dual culture method. The LAB isolates inhibited A. flavus growth with the largest growth inhibition was showed by LAB isolate 1 that isolated from poultry house soil (17.65%). However, statistics analysis showed that fungal growth inhibition by LAB isolates were not significantly difference (p>0,05). Although the result was not statistically significant, all  LAB isolates significantly inhibited conidia development. This showed potential application of LAB isolates as biopreservative agent. Keywords: antifungal, Aspergillus flavus, biopreservative, food stock, lactic acid bacteria
