Abstract

Vaccination is one of the main prevention of the spread of Covid-19. Technological engineering based on virus attenuation has been applied in vaccine development. This study aims to obtain an epitope-based vaccine design with low risk of allergic reactions, safe, and inexpensive. The design of the vaccine was conducted by the collection of SARS-CoV-2 sequence data, phylogenetic analysis, prediction of protein antigenicity, and identification of CD 8+ T cell epitopes. Furthermore, epitope conservation and immunogenicity prediction, as well as molecular docking analysis were carried out to see the interaction between epitopes and alleles. The next step was B cell epitope prediction, population coverage prediction, construction and visualization of vaccine design, structural analysis and validation, interaction analysis between vaccines with TLR 3 and TLR 4, and evaluation of vaccine design immunogenicity. All stages were carried out using the appropriate webserver. The designed vaccine had an antigenicity of 0.5134, not toxic, and not allergenic. The physicochemical parameters met the requirements except for the molecular weight which was less than 40 KDa. The designed vaccine was predicted to have a population coverage of 95.14% for the Indonesian population. The results of the immunogenicity prediction of the vaccine design showed an increase of IgM and IgG until day 35.

Abstract

The ventilation system is a critical factor in underground mining operations, ensuring the availability of fresh air and the dilution of hazardous gases for workers. This study will be conducted at PT Bukit Asam Tbk’s Ombilin Mining Unit (UPO) and will focus on analyzing fan requirements to optimize the airflow system. The issues addressed in this study include the imbalance between the supplied air volume and the size of the working area, as well as the potential for methane (CH₄) and carbon monoxide (CO) accumulation. The research methodology involves direct field measurements of ventilation parameters- such as air velocity, airway cross-sectional area, humidity, and gas concentration levels. The collected data will be processed mathematically to calculate total fresh air requirements based on the number of workers, machinery, and occupational health standards. The study aims to provide technical recommendations regarding the appropriate specifications and placement of fans (both main and auxiliary) to create a safe and productive working environment in compliance with applicable mining safety regulations.

Abstract

The drainage system is a crucial aspect of underground mining operations at PT BA UPO to keep the work area dry and safe from the threat of high water discharge that could potentially hamper production activities. This research aims to conduct a technical study of the pumping system by calculating the total water discharge entering from groundwater and rain runoff, evaluating the actual pump capacity, and determining the optimal pump head requirements. Through field observation metphods and technical data analysis, the research results indicate that the current pumping system requires optimization in operational duration and settling pond management to improve efficiency. Based on the calculation of the total dynamic head, technical parameters were obtained that can serve as a reference for the company in adjusting pump performance to maximize the discharge of accumulated water out of the mine, ensuring that underground mining operations can be maintained more effectively. The expected optimization can reduce the risk of air pollution and improve the distribution system to support the safety and continuity of underground mining operations.

Abstract

Acid mine drainage (AMD) is one of the negative impacts of mining activities that can significantly reduce environmental quality due to its low pH and high concentration of dissolved heavy metals. Therefore, proper and effective treatment is required before the water is discharged into the surrounding environment. This study aims to analyze the management of acid mine drainage using an active treatment method with quicklime (CaO) and to determine the appropriate dosage required at KPL 18 Stockpile of RMKO (RMKO), Muara Enim Regency. The research methods include field observations and data collection consisting of primary data such as pH, water discharge, pond volume, and lime dosage, as well as secondary data including location maps and rainfall data. Data analysis was carried out through pH measurements at the inlet and outlet channels and supported by jar test experiments to evaluate the effectiveness of lime addition. The results indicate that the pH values at the inlet range from 4.41 to 4.47, while at the outlet they range from 7.81 to .94, which comply with environmental quality standards (pH 6–9). The treatment process involves water flow through channels, processing in settling ponds with the addition of quicklime , followed by discharge into receiving water bodies. The use of quicklime has proven effective in increasing pH and facilitating the precipitation of dissolved metals, indicating that this method is efficient and applicable for AMD management.
